Vehicle versus FASN-inhibited human metastatic breast cancer global whole cell lysate

ABSTRACT: Metastatic breast cancer cells (MCF10CA1a) were treated with vehicle (DMSO) or 20 uM of Fatty Acid Synthase (FASN) inhibitor TVB-3166 for three days. Untargeted global proteomics (Tryp/Lys-C; Lumos) was performed on the whole cell lysates.

Inflammatory Signaling is Regulated by Fucosylation in Metastatic Breast Cancer Cells

Project description:N-linked glycosylation results in a large branched tree-like sugar structure composed of several types of individual carbohydrates being attached to a protein. Fucosylation is one specific type of glycosylation that is defined by the addition of α-L-fucose to a carrier protein. Although fucosylation has been relatively well defined as a biomarker for progression in some human cancers, for example, pancreatic and hepatocellular carcinoma, its role in breast cancer is much less well defined. A growing body of evidence indicates that levels of fucosylation correlate with breast cancer progression and contributes to metastatic disease. However, very little is known about the signaling and functional outcomes that are driven by fucosylation. We performed tandem- mass-tag (TMT) proteomics on 4T1 metastatic mammary tumor cells that were treated with a fucosylation inhibitor, 2-fluorofucose (2FF) or vehicle (DMSO) control. This analysis identified >400 proteins that were significantly increased or decreased in 2FF treated samples compared to the DMSO treated samples. We found that two proteins, Tollip and Bcl-10, were downregulated in 4T1 cells in response to 2FF treatment. Functionally, Tollip and Bcl10 signaling induce pro-inflammatory responses through activation of NF-κB, a transcription factor that is pro-tumorigenic and a prime target in human cancer. Our results show that treatment of 4T1 cells with 2FF leads to a reduction in NF-κB expression and activity through increased IκBα. This observation is consistent with a reduction in inflammatory response due to the loss of Tollip and Bcl-10 expression. Collectively, our results indicate that fucosylation is a key mediator of inflammatory signaling in metastatic breast cancer cells.

Whole-genome expression profiling of metastatic versus non-metastatic mammary tumors in MMTV-Wnt1 transgenic mice

Project description:To identify genes that mediate lung metastasis in breast cancer, we compared expression profiles of metastatic versus non-metastatic mammary tumors in MMTV-Wnt1 transgenic mice. A subset of biologically relevant genes with statistically significant changes was selected for validation. These genes include Alox15, Ptn, Ror2, Sox9, Jag2 and Runx2. These genes encode proteins that play important roles in the immune and inflammatory responses as well as osteogenesis and skeletal morphogenesis.

Gene expression profiling of metastatic lung cancer cell lines

Project description:During malignant disease progression, the extracellular matrix (ECM) of epithelial tumors accumulates inter-molecular cross-links between collagen strands; these cross-links enhance ECM stiffness and trigger tumor cell invasion and dissemination, but the mechanisms that regulate intra-tumoral collagen maturation have not been fully defined. Using a new mouse model of metastatic lung adenocarcinoma driven by mutant K-ras expression and Cdkn1a inactivation, we showed that tumor cell invasion and metastasis are driven by high expression of lysyl hydroxylase 2 (LH2), an enzyme that hydroxylates telomeric lysine (Lys) residues on collagen. We interbred KrasLA1 mice, which develop non-metastatic lung adenocarcinomas at 8-10 months of age because of the expression of a somatically activated K-rasG12D allele, with Cdkn1a-/- mice, which develop sarcomas and B cell lymphomas because of depletion of p21. Cohorts were generated that had neither, either, or both mutant alleles. KrasLA1 mice that were p21-deficient (KC mice) had a shorter mean time to death than did p21-replete ones. We established cell lines from the tumors, and compared cell lines from highly metastatic tumors with those from poorly metastatic tumors.
