Effects of methionine supplementation on seminal plasma miRNAs and proteome in sheep

ABSTRACT: These studies provide information demonstrating that SP components can be altered by paternal diet, however, knowledge is greatly lacking in livestock species regarding the effects of specific dietary changes that may impact production and reproduction traits. To our knowledge, this is the first study to evaluate the effects of dietary methionine on SP composition, specifically miRNAs and proteins.

Effect of maternal methionine supplementation on the transcriptome of bovine preimplantation embryos

Project description:Maternal nutrition exclusively during the periconceptional period can induce remarkable effects on both oocyte maturation and early embryo development, which in turn can have lifelong consequences. The objective of this study was to evaluate the effect of maternal methionine supplementation on the transcriptome of bovine preimplantation embryos. Holstein cows were randomly assigned to one of two treatments differing in level of dietary methionine (1.89 Met vs. 2.43 Met % of metabolizable protein) from calving until embryo flushing. High quality preimplantation embryos from individual cows were pooled and then analyzed by RNA sequencing. A total of eight Holstein dairy cows were used in this study. Preimplantation embryos recovered from each cow were pooled in order to generate two replicates per cow assayed. Each pool consisted of 2-4 expanded blastocysts with excellent quality. Overall, a total of 16 embryo pools underwent RNA extraction, amplification, and subsequent sequencing.

Project description:Maternal nutrition can induce modifications in the epigenome of the fetus, resulting in gene expression alterations and impacting future performance. Epigenetic changes are related to nutrition via the one carbon cycle, which involves aminoacid methionine. We hypothesized that an increased dietary methionine in beef cows during early gestation will impact the whole-genome DNA methylation and transcriptome of the offspring.

Dietary cholesterol supplementation to a plant-based diet for Atlantic salmon

Project description:The study evaluated effects of dietary cholesterol (1.5%) in Atlantic salmon fed a plant based diet for 77 days. Cholesterol supplementation did not affect growth or organ weights of Atlantic salmon, but promoted induction of cholesterol and plant sterol efflux in the intestine, whereas sterol uptake was suppressed. Microarray analyses in the liver indicated decreased cholesterol biosynthesis and enhanced conversion to bile acids. The marked effect of cholesterol on bile acid synthesis suggests that dietary cholesterol can be used to stimulate bile acid synthesis in fish. The study clearly demonstrated how Atlantic salmon adjusted metabolic functions in response to the dietary load of cholesterol, and has expanded our understanding of sterol metabolism and turnover that adds to the knowledge of these processes in fish. Atlantic salmon received feeds based on plant ingredients with (CH) and without (K) supplementation of cholesterol. Liver samples were collected after 77 days. Five individuals from each group were analyzed with microarrays, pooled liver sample of salmon fed with commerical fish meal based feed was used as a reference.

TMT-based proteomics of longissimus lumborum muscle in newly weaned piglets after dietary Spirulina inclusion and enzyme supplementation

Project description:The objective of this study was to evaluate the impact of dietary Spirulina and lysozyme supplementation over the muscle proteome of piglets during the post-weaning stage. Thirty piglets were randomly distributed among three diets: control (no microalga), SP (10% Spirulina) and SP+L (10% Spirulina + 0.01% lysozyme). They were fed ad libitum for 4 weeks, after which they were sacrificed and samples of the longissimus lumborum muscle were taken. The muscle proteome was analysed using a Tandem Mass Tag (TMT)-based quantitative approach.

Small RNAs and their targets are associated with the transgenerational effects of water-deficit stress in durum wheat

Project description:Water-deficit stress negatively affects wheat yield and quality. Abiotic stress on the parental plants during reproduction could have transgenerational effects on the progenies. Here we investigated the transgenerational influence of pre-anthesis water-deficit stress by detailed analysis of the yield components, grain quality traits, and physiological traits in durum wheat. Next-generation sequencing analysis profiled the small RNA-omics, mRNA transcriptomics, and mRNA degradomics in the progenies. Parental water-deficit stress had positive impacts on the progenies in certain traits like harvest index and protein content in given genotype. Small RNA-seq identified 1739 conserved and 774 novel microRNAs (miRNAs). Transcriptome-seq characterised the expression of 66,559 genes while degradome-seq profiled the miRNA-guided mRNA cleavage dynamics. Differentially expressed miRNAs and genes were identified, with significant regulatory patterns subject to trans- and inter- generational stress. Integrated analysis based on the three omics revealed the significant biological interactions between stress-responsive miRNA and targets, with possible contributions towards transgenerational stress tolerance via pathways such as hormone signalling and nutrient metabolism. Our study provides the first confirmation of the transgenerational effects of water-deficit stress in durum wheat. New insights gained on the molecular level indicate that key miRNA-mRNA modules are potential candidates in transgenerational stress improvement.
