Intact Proteoforms from 10,000 Single Brain Cells

ABSTRACT: We performed intact proteoform profiling of 11303 single cells from the rat hippocampus using single-cell Proteoform imaging Mass Spectrometry (scPiMS). scPiMS realizes the benefits of direct extraction of intact proteins and demonstrates unprecedented throughput for mass spectrometry-based single cell proteomics. Utilizing a new informatics workflow dedicated for scPiMS, neurons, astrocytes or microglia cell types are assigned to 93% of the single cell population using proteoform signatures.
