Project description:If relapses occur in only 20% of T-ALL children, 70% will have a dismal outcome, justifying the need for new therapeutic options. Transcriptomic plasticity is a property of leukemic cells to adapt their functions and resist to treatments. The project aims at analyzing, at a single cell resolution, the variations of the gene expression, between diagnostic and relapse. This will allow identification of relapse-associated genes as potential new targets for future innovative treatments.

Molecular and clinical analyses of PHF6 mutant myeloid neoplasia provide clues as to their pathogenesis and therapeutic targeting

Project description:PHF6 mutations (PHF6MT) are identified in various myeloid neoplasms (MN). However, little is known about the precise function and consequences of PHF6 in MN. Our comprehensive genomic analyses focused on three main findings. Firstly, we revealed a different pattern of genes correlating with PHF6MT in male and female cases. When analyzing male and female cases separately, in only male cases, RUNX1 and U2AF1 were co-mutated with PHF6. In contrast, female cases revealed co-occurrence of ASXL1 mutations and X-chromosome deletions with PHF6MT. Next, proteomics analysis revealed a direct interaction between PHF6 and the pioneer transcription factor RUNX1. Both proteins co-localize in active enhancer regions that define the context of lineage differentiation. Finally, we demonstrated a negative prognostic role of PHF6MT, especially in association with RUNX1. The negative effects on survival were additive as PHF6MT cases with RUNX1 mutations had worse outcomes when compared to cases carrying single mutation or wild-type.

Targeting Electron Transport Chain Confers Therapeutic Vulnerability in Castration-Resistant Prostate Cancer

Project description:Most prostate cancer will develop into castration-resistant prostate cancer, which is the most frequent cause of death for prostate cancer patients. Despite novel androgen receptor antagonists have significantly improved clinical outcomes for these patients, some patients still could not benefit from existing treatment regimens. By analyzing the single-cell RNA-sequencing data and bulk-sequencing data, we discovered that oxidative phosphorylation and electron transport chain (ETC) pathway were enhanced in the prostate cancer microenvironment following tumor progression. Meanwhile, high ETC was related to poor clinical outcomes in prostate cancer patients. Both in vitro and in vivo castration-resistant prostate cancer models demonstrated that ETC inhibitor marked suppressed tumor growth and the synergistic antitumor efficacy of the combination of ETC inhibitor and androgen receptor antagonist. Our study indicates that ETC activity is a metabolic vulnerability for advanced prostate cancer and can serve as a novel therapeutic target.

Broad-Spectrum Therapeutic Suppression of Metastatic Melanoma Through Nuclear Hormone Receptor Activation

Project description:Melanoma metastasis is a devastating outcome in need of novel preventive therapies. We provide pharmacologic, nolecuar, and genetic evidence establishing the liver-X nuclear hormone receptor (LXR) as a therapeutic target in melanoma. Molecular and genetic experiments revealed these effects to be mediated by LXRb, which elicits these outcomes through transcriptional induction of tumoral and systemic apolipoprotein-E (ApoE). LXRb agonism robustly suppressed tumor growth and metastasis across a wide spectrum of melanoma lines of diverse mutational subtypes established in xenograft, immunocompetent, and genetically-initiated model. We propose a path for the clinical testing of LXRb targeting-a therapeutic approach that uniquely acts by transcriptionally acivating a metastasis suppressor gene. In this experiment we analyzed the effect of GW3965 treatment on gene expression in the MeWo human melanoma cell line. The cells were treated either with DMSO or GW3965 at 1 micromolar for 48 hours, after which the RNA was extracted and gene expression was analyzed by transcriptomic profiling
