Metabolite annotation of rat epididymis with High-resolution imaging mass spectrometry

ABSTRACT: Infertility or subfertility is a widespread problem with approximately 25% of couples seeking medical help. For about half the cases it lies in the male partner. Male infertility is caused by problems that affect sperm production, maturation and/or transport. In this study the role played by the epididymis in the post-testicular maturation of spermatozoa during its transit through this organ was investigated using FT-ICR imaging mass spectrometry of metabolites and the FDR-controlled metabolite annotation platform MetaSpace for high-resolution imaging mass spectrometry. This study together with MTBLS313 provides the accompanying data for FDR-controlled metabolite annotation for high-resolution imaging mass spectrometry.

MALDI-IMS combined with shotgun proteomics identify and localize new factors in male infertility

Project description:Spermatogenesis is a complex multi-step process involving intricate interactions between different cell types in the male testis. Disruption of these interactions results in infertility. Combination of shotgun tissue proteomics with MALDI imaging mass spectrometry is markedly potent in revealing topological maps of molecular processes within tissues. Here, we use a combinatorial approach on a characterized mouse model of hormone induced male infertility to uncover misregulated pathways. Comparative testicular proteome of wildtype and mice overexpressing human P450 aromatase (AROM+) with pathologically increased estrogen levels unravels gross dysregulation of spermatogenesis and emergence of pro-inflammatory pathways in AROM+ testis. In situ MS allowed us to localize misregulated proteins/peptides to defined regions within the testis. Results suggest that infertility is associated with substantial loss of proteomic heterogeneity, which define distinct stages of seminiferous tubuli in healthy animals. Importantly, considerable loss of mitochondrial factors, proteins associated with late stages of spermatogenesis and steroidogenic factors characterise AROM+ mice. Thus, the novel proteomic approach pinpoints in unprecedented ways the disruption of normal processes in testis and provides a signature for male infertility.

Multiple reproductive tract-specific whey acidic protein four-disulfide core genes demonstrate functional requirement for sperm differentiation and function in mice

Project description:Our research defines the pivotal roles of the WFDC locus genes in male fertility, particularly in the regulation of sperm development and function. By employing CRISPR/Cas9 mediated gene editing strategy to study this gene family, we provided robust genetic evidence confirming their interconnected functions, which are crucial for sperm differentiation and maturation. Disruptions within these genes can result in infertility, highlighting their regulatory significance. These findings substantiate the potential of targeting EPPIN and related genes for male contraceptive development. Moreover, our study suggests that mutations in the WFDC locus could be instrumental in diagnosing and treating idiopathic male infertility, thereby presenting new therapeutic prospects for reproductive health.

Disruption of TEX38 impairs sperm morphogenesis and diverse functions including the migration of sperm into the oviduct

Project description:In male reproduction, spermatogenesis and sperm maturation are critical for the production of normal sperm and offspring, yet the underlying molecular mechanisms remain largely elusive. Tex38 is a testis-enriched gene, and its deficiency results in oligoasthenoteratospermia (OAT) with aberrant epididymis, leading to male infertility in mice. Tex38 knockout (KO) sperm primarily exhibited neck bending deformities and functional abnormalities, including impaired fertilization. Proteomic analysis identified ADAM3 and its maturation-associated chaperones (CALR3, CLGN, and PDILT) as the most significantly altered proteins among the differentially expressed proteins (DEPs) in both sperm and epididymis of Tex38 knockout mice. GO analysis revealed that DEPs were primarily involved in sperm morphogenesis, motility, and fertilization. ARRDC5 was identified as a novel interacting protein of TEX38, and its deletion resulted in similar male infertility phenotypes as Tex38 deletion. Immunoprecipitation-mass spectrometry identified TEX38 and ARRDC5 interacts with CLGN and PDILT. The interactions among TEX38, ARRDC5, PDILT, and CLGN were found to affect ADAM3 maturation, resulting in the failure of both Tex38-/- and Arrdc5-/- sperm to migrate to the oviduct. Overall, these findings establish TEX38 as an essential regulator of mammalian ADAM3-related migration, sperm formation, energy metabolism (ATP generation) sperm-egg binding, and fertilization. TEX38 represents a potential target for diagnosis and treatment of male infertility and male contraception.

Identification of a damaging variant rs35033974 associated with idiopathic male infertility through degradation of TEX101 protein and its transient interactome

Project description:Human TEX101 is a testis-specific cell membrane protein expressed exclusively in male germ cells and a validated biomarker of male infertility. TEX101 was suggested to function as a cell surface chaperone of numerous proteins involved in sperm migration and sperm-egg interaction. However, the precise functional roles of human TEX101 in spermatogenesis and fertilization are unknown. Here, we show that a common homozygous variant rs35033974 of TEX101 (G99V) with ~1% frequency in the general population may be associated with idiopathic male infertility. Spermatozoa from patients with homozygous rs35033974 exhibited near-complete degradation of variant G99V TEX101 protein and concomitant degradation of its interactome, as revealed by proteomic measurements. Substantially reduced levels of numerous testis-specific membrane proteins involved in sperm migration and sperm-oocyte fusion (including LY6K, IZUMO3 and ADAM29) were confirmed in spermatozoa of men with homozygous G99V variant. These men were previously diagnosed with idiopathic male infertility or oligospermia and failed the treatment by intrauterine insemination. Collectively, these data may facilitate diagnostics of idiopathic male infertility, provides a rational for selection of male infertility treatments and validate TEX101 and its interactome as targets to develop non-hormonal male contraceptives.
