Metabolomics of Saliva Samples Obtained from Subjects with Diabetes

ABSTRACT: In this research, were are investigating the metabolic profile changes associated with well- and poorly-controlled type 1 and 2 diabetes and if there are distinct metabolite compounds that may be associated with glycemic control. The PI of the study collected whole unstimulated saliva samples were from subjects with well- and poorly-controlled type 1 and type 2 diabetes. Subjects were selected based on the level of A1C (<7= well-controlled and >7 = poorly controlled). Saliva samples were shipped to RTI RCMRC for a broad spectrum reverse phase metabolomics analysis.

Microarray analysis of surgical liver biopsies from 13 obese (nine with type 2 diabetes) and five control subjects

Project description:Hepatic lipid accumulation is an important complication of obesity linked to risk for type 2 diabetes. To identify novel transcriptional changes in human liver which could contribute to hepatic lipid accumulation and associated insulin resistance and type 2 diabetes (DM2), we evaluated gene expression and gene set enrichment in surgical liver biopsies from 13 obese (9 with DM2) and 5 control subjects, obtained in the fasting state at the time of elective abdominal surgery for obesity or cholecystectomy. RNA was isolated for cRNA preparation and hybridized to Affymetrix U133A microarrays. Human liver samples were obtained from 5 lean control subjects undergoing elective cholecystectomy and 13 obese subjects (with or without Type 2 diabetes) undergoing gastric bypass surgery. Subjects with diabetes were classified as either well-controlled or poorly-controlled.

Comparative salivary proteomics of people with different blood glucose levels

Project description:To evaluate the effect of glycemic control status in middle-aged and elderly individuals with type 2 diabetes mellitus (T2DM) on oral proteomic by liquid chromatography tandem mass spectrometry (LC-MS/MS). Twenty-four individuals who met the criteria were included and divided into a well-controlled group and a poorly-controlled group according to their glycated hemoglobin levels. The two groups were matched in age, gender and clinical parameters. After unstimulated whole saliva samples were collected, LC-MS/MS technique in data independent acquisition (DIA) mode was used to detect the proteins in samples. In addition, we compared the differentially expressed proteins between groups with different glycemic levels in saliva. Finally, we performed functional annotation and enrichment analysis of proteins.

Chlorella ingestion and suppression of resistin gene expression in borderline diabetics: a randomized, placebo-controlled study

Project description:Diabetes and Arteriosclerosis progression are frequently observed in borderline Type 2 diabetes cases. Onset of complications (arteriosclerosis and renal damage) due to Type 2 diabetes is well documented; it is extremely important to prevent or delay their progression. Type 2 diabetes onset and progression has been controlled through dietary habits and exercise, although these remain insufficient. Chlorella ingestion improves blood glucose and cholesterol concentrations in mice and humans, although no reports have evaluated Chlorella effects in borderline diabetics. Therefore, we conducted a randomized, placebo-controlled trial for borderline diabetics using laboratory results and comprehensive gene analysis as outcomes. Chlorella ingestion suppressed resistin gene expression, suggesting that Chlorella may be useful for preventing diabetes onset and ameliorating arteriosclerosis. Subjects were randomly divided into two groups: Chlorella group (n = 28) ingesting Chlorella powder (8.0 g/day) and placebo group (n = 29) ingesting lactose formulation (8.0 g/day) for 12 weeks. Blood and urine were collected every 4 weeks for laboratory tests. Gene expression analysis used RNA extracted from peripheral blood samples before and after 12 weeks of Chlorella or lactose ingestion.

Project description:Changes in gene expression in pancreatic Î² cells from type 2 diabetes should provide insights into their abnormal insulin secretion and turnover. Gene expression profile of Î² cells obtained from type 2 diabetic subjects and non-diabetic controls was evaluated by microarray analysis. Frozen sections were obtained from cadaver pancreases of 10 control and 10 type 2 diabetic human subjects. Î²-cell enriched samples were obtained by laser capture microdissection. RNA was extracted, amplified and subjected to microarray analysis.

Protein microarrays were used to analyze saliva and plasma from HIV infected and HIV uninfected subjects to identify new biomarkers for HIV disease progression and pathogenesis. A 507 protein microarray was employed to provide a broad view of cytokines and chemokines in saliva and plasma in acutely HIV infected subjects as compared to uninfected subjects. A custom array derived from the initial results was refined to highlight those molecules with significant change relative to control subjects indicating the potential for biological impact.
