NIH 3T3 fibroblast cells with Ceftriaxone Treatment

ABSTRACT: NIH 3T3 cells were plated at 1.5million cells per 10cM dish into 12 10cm dishes. 24hrs post plating media was changed on all plates. At the time of media change, 4 dishes were treated with a final concentration of 50uM ceftriaxone (24hrs ceftriaxone n=4). 23 hrs post media change, 4 separate dishes were treated with 50uM ceftriaxone for 1hr (1hr ceftriaxone n=4). 4 dishes were untreated (0hr, basal, n=4). 24 hrs post media change, media was quickly removed from all 12 plates, each plate was rapidly washed with MilliQ H2O and rapidly frozen with liquid nitrogen poured directly into the dishes. All plates were transferred to dry ice and stored at -80 until shipped.

Effect of atrazine and sucrose on xenobiotic tolerance in Arabidopsis

Project description:rs05-05_atrazine - effect of atrazine and sucrose on arabidopsis transcriptome - Growth in the presence of sucrose was shown to confer to Arabidopsis thaliana seedlings, under conditions of in vitro culture, a very high level of tolerance to the herbicide atrazine and to other photosynthesis inhibitors (Sulmon et al., 2004). The CATMA investigation will be useful to reveal the gene networks implicated in the mechanisms of xenobiotics tolerance. - Seeds of Arabidopsis thaliana (ecotype Columbia) were surface-sterilized inbayrochlore/ethanol (1:1, v/v), rinsed in absolute ethanol and dried overnight. Germination and growth were carried out under axenic conditions in square Petri dishes. After seeds were sowed, Petri dishes were placed at 4degreeC for 48 h in order to break dormancy and homogenize germination, and then transferred to a control growth chamber at 22degreeC under a 16-h light period regime at 4500 lux for 4 weeks. Growth medium consisted of 0.8% (w/v) agar in 1x Murashige and Skoog (MS) basal salt mix (Sigma, St. Louis, MO, USA) adjusted to pH 5.7. After 4 weeks of cultivation on vertical plates plantlets were transferred on fresh medium complemented or not with atrazine 10 uM and with sucrose 80mM or mannitol 80 mM which were directly added during preparation of agar-MS media prior to sterilisation. Atrazine was sterilized by microfiltration through 0.2 um cellulose acetate filters (Polylabo, Strasbourg, France) and then axenically added to melted agar-MS medium prior to pouring into Petri dishes. TMT analysis of LARP6 knockdown impact on protrusion induced changes in the proteome of MDA-MB231 cells

Project description:To assess if LARP6 is required for upregulation of ribosomal proteins upon protrusion induction, MDA-MB231 breast cancer cells were treated with non-targeting (NT) or LARP6 siRNA for 48 hrs, before being seeded on top of collagen-I coated 3 micron transwell filters (corning). No media was added to the bottom chamber and cells were allowed to adhere to filter overnight. The next day, the media on top of cells was refreshed, and the filters were either left with no media in the bottom chamber (closed pores), or with media also added to the bottom chamber (open pores) in order to allow formation of protrusions. Cells were allowed to form protrusions for 12 hrs before being lysed and analysed by mass spectrometry.

Project description:Osteoblasts from 4 donors were seeded onto 20 constructs. Constructs were constructed as described: Calcium phosphate (brushite / β-TCP) posts were pinned into a Sylgard 184 silicone elastomer (base and curing agent, Dow Corning Corporation) base layer in 6-well dishes. Cells were seeded in fibrinogen onto a thrombin gel and subsequently cultured in 3ml of proliferation media (osteoblast media, 50 μg/ml ascorbate-2-phosphate, 40 μg/ml proline) per well until cells had fully contracted the gel between the support posts. Week 0 constructs were harvested and snap-frozen at this point with the remainder transferred to osteogenic media (proliferation media, 10 mM β-glycerophosphate, 10 nM dexamethasone) and harvested at 2, 4, 6 and 8 weeks.

Project description:cea12-02_chlorosap - comparison after 4 and 52h - To observe the effects of guanosine penta/tetraphosphate on gene expression - Plants of four different genotypes (43A10.6, 44B1.3, H1 P3.2 and Col-0) were grown for ten days on MS/2 plates with 1% sucrose under 80µE light in long-day conditions (16 hours of light, 8 hours of dark). Plants were treated at 10:30 in the morning (Time 0; 10:30 is 3 and a half hours after the onset of the light period) by flooding with 30uM dexamethasone (in aqueous solution with 0,3 % DMSO and 0,033 % Silwet L-77) for 5 minutes . Samples were taken at 4 hrs and 52 hrs after T0. Aerial parts of the seedlings were rapidly harvested and frozen in liquid nitrogen.
