The impact of acute Colony Stimulating Factor 1 treatment on serum and liver metabolites in fed and fasted mice – SERUM

ABSTRACT: The aim of the study was to investigate the impact of expanding tissue macrophage populations on systemic metabolism. Groups of male mice were treated with 4 x daily injections of Colony Stimulating Factor (1), followed by normal feeding or a 24 h fast from day 6 to day 7. Serum and liver were collected for GC-MS metabolomic analysis on a Shimadzu TQ8050.

Influence of hyperthyroid conditions on gene expression in rat tibialis anterior

Project description:Extraocular muscles (EOMs) are a highly specialized type of tissue with a wide range of unique properties, including characteristic innervation, development, and structural proteins. Even though EOMs are frequently and prominently involved in thyroid-associated diseases, little is known about the immediate effects of thyroid hormone on these muscles. In order to create a comprehensive profile of changes in gene expression levels in EOMs induced by thyroid hormone, hyperthyroid conditions were simulated by treating adult Sprague-Dawley rats with intraperitoneal injections of 25 ?g T3 per 100 g body weight over the course of six weeks; subsequently, microarray analysis was used to determine changes in mRNA levels in EOMs from T3-treated animals relative to untreated controls. RNA isolated from tibialis anterior muscles of the same animals was used as a reference fast-twitch muscle. Adult Sprague-Dawley rats (initial body weight 200 ± 40 g) were fed ad libitum with standard laboratory diet and tap water. Eight rats were randomly divided into a T3-treated and a control group, three males and one female each. To simulate a chronically hyperthyroid state, T3 treatment was administered by intraperitoneal injections of 25 µg T3 (Sigma-Aldrich, St. Louis, MO, U.S.A.) per 100 g body weight. Injections were administered every second day over a period of six weeks. 24 hours after the last injection, the animals were sacrificed by CO2 asphyxiation, muscles were dissected immediately after death. Each of the four biological replicates in each group was used for a single microarray each. RNA from EOMs was used. Tibialis anterior RNA from the same animals served as a fast-twitch reference muscle.

Single-nucleus RNA sequencing of the mouse hypothalamus in the fed and fasted state

Project description:To provide a summary of cells within the mouse hypothalamus in the fed and fasted state, we performed single nucleus RNA sequencing on 12 mice fed ad libitum, and 6 mice that were fasted overnight. Hypothalami were dissected and stored at -80ºC overnight. The next day single nucleus supensions were made, pooling together hypothalamic from the same feeding condition (2 x ad libitum prepared on separate days; 1 x overnight fast). Susepensions were FACS sorted for DRAQ5 positive events. Single nucleus RNA sequencing libraries were generated using the 10X Chromium single cell 3' reagents.

Elevated Serum Fibroblast Growth Factor 21 in Humans with Acute Pancreatitis.

Project description:BackgroundThe metabolic regulator Fibroblast Growth Factor 21 (FGF21) is highly expressed in the acinar pancreas, but its role in pancreatic function is obscure. It appears to play a protective role in acute experimental pancreatitis in mice. The aim of this study was to define an association between FGF21 and the course and resolution of acute pancreatitis in humans.Methods and principal findingsTwenty five subjects with acute pancreatitis admitted from May to September 2012 to the Beth Israel Deaconess Medical Center (BIDMC) were analyzed. Serial serum samples were collected throughout hospitalization and analyzed for FGF21 levels by ELISA. Twenty healthy subjects sampled three times over a four week period were used as controls. We found that, in patients with pancreatitis, serum FGF21 rises significantly and peaks four to six days after the maximum lipase level, before slowly declining. Maximum FGF21 levels were significantly greater than baseline levels for acute pancreatitis subjects (1733 vs. 638 pg/mL, P = 0.003). This maximum value was significantly greater than the highest value observed for our control subjects (1733 vs. 322 pg/mL, P = 0.0002). The ratio of active to total FGF21 did not change during the course of the disease (42.5% vs. 44.4%, P = 0.58). Fold increases in FGF21 were significantly greater in acute pancreatitis subjects than the fold difference seen in healthy subjects (4.7 vs. 2.0, P = 0.01). Higher fold changes were also seen in severe compared to mild pancreatitis (18.2 vs. 4.4, P = 0.01). The timing of maximum FGF21 levels correlated with day of successful return to oral intake (R2 = 0.21, P = 0.04).ConclusionsOur results demonstrate that serum FGF21 rises significantly in humans with acute pancreatitis. The pancreas may be contributing to increased FGF21 levels following injury and FGF21 may play a role in the recovery process.

Transcriptional profiling of human monocyte-derived macrophages

Project description:In this study we compared the response of human monocyte-derived macrophages differentiated with either granulocyte-macrophage colony-stimulating factor (GM-CSF) or macrophage colony-stimulating factor (M-CSF) to the most common activation stimuli: LPS plus interferon-γ to induce macrophage polarization towards the M1 type or IL-4 to induce macrophage polarization towards the M2a type. Additionally, IL-10 was used to drive M-CSF-primed macrophages into the M2c state. We used the the whole-human genome microarray to determine genes that were up- or downregulated by the activation stimuli in both macrophage lineages, with focus on genes implicated in immune response.
