Project description:Strobilurins are an important class of agrochemical fungicides used throughout the world on a wide variety of crops to protect against fungal pathogens. In addition to their protective role against pathogens, they are reported to also positively influence plant physiology. In this study, we analyzed the effect of Stroby WG, a commercially available fungicide consisting of 50% (w/w) kresoxim-methyl (KM) as active strobilurin compound, on Arabidopsis plant growth. Treatment of seeds or seedlings with Stroby resulted in larger leaves due to an increase in cell number. Transcriptome analysis of Stroby-treated rosettes demonstrated an increased expression of genes involved in redox homeostasis, iron metabolism and sugar transport. Stroby treatment strongly induced the expression of the subgroup Ib basic helix-loop-helix (bHLH) transcription factors, which have a role in iron homeostasis under iron-limiting conditions.

Project description:Peptidases are known to play key roles in multiple biological processes in all living organisms. In a model plant Arabidopsis, the vast majority of many putative aminopeptidases remain uncharacterized. We therefore aim to explore physiological function of uncharacterized aminopeptidase in higher plants using Arabidopsis as a model to study.We performed functional and expression analyses of the Arabidopsis LAP2 through cDNA cloning, isolation of T-DNA insertional mutants, characterization of the enzymatic activity, characterization of gene expression, and transcriptomic and metabolomic analyses of the mutants. We found that LAP2, one of the 28 aminopeptidases in Arabidopsis, regulates plant growth, leaf longevity and stress response by controlling intracellular protein turnover. Loss-of-function alleles of LAP2 reduced in vegetative growth, accelerated leaf senescence and rendered plants more sensitive to various stresses. LAP2 is highly expressed in the quiescent center cells in the root meristem, cotyledons and leaf veins. Integration of global gene expression and metabolite analyses suggest that LAP2 controlled intracellular protein turnover. The mutant maintained free leucine by up-regulating key genes for leucine biosynthesis, however, this influenced the flux of glutamate strikingly. As a result, gamma-aminobutyric acid, a metabolite which is derived from glutamate, was diminished in the mutant. Decrements in these nitrogen-rich compounds are associated with morphological alterations and stress sensitivity of the mutant.Our results provide molecular and biochemical evidence that LAP2 is indeed an enzymatically active aminopeptidase. LAP2 plays key roles in senescence, stress response and protein turnover. Regulated proteolysis is an important mechanism in all stages of the plant life cycle. The present study would contribute to further understanding of the aminopeptidases which have several implications in higher plants.

Project description:The intricate interactions between plants and microorganisms have garnered substantial scientific interest. While previous studies have highlighted the potential influence of various fungal volatile compounds(VCs) on plant growth and development, the precise mechanisms underlying this modulation still need to be discovered. In this study, we discovered that fungal volatile organic compounds from the soil-borne fungus Tolypocladium inflatum GT22 enhance the growth of Arabidopsis. Remarkably, following the priming of Arabidopsis with GT22 VC, it displayed an enhanced immune response, thereby mitigating the detrimental effects caused by both pathogenic infections and copper stress. Transcriptomic analyses of Arabidopsis seedlings treated with GT22 VCs revealed the differential expression of 90, 83, and 137 genes after 3, 24, and 48 hours of volatile exposure, respectively. These responsive genes are involved in growth, hormone regulation, defense mechanisms, and signaling pathways. Notably, the induction of genes related to innate immunity, hypoxia, salicylic acid (SA) biosynthesis and camalexin biosynthesis by GT22 VCs were reported. Among the VCs emitted by GT22, limonene is particularly noteworthy. Arabidopsis seedlings exposed to limonene exhibited not only growth promotion effects but also alleviation of copper stress, indicating that limonene may play a role in the interaction between GT22 and plants. Overall, the findings of this study provide evidence supporting that fungal VCs can promote plant growth and enhance both biotic and abiotic tolerance. Furthermore, our results suggest that seedlings exposed to T. inflatum GT22 VCs holds promising potential for harnessing beneficial effects to improve crop productivity.

Project description:The developmental functions of SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) 10 have been extensively studied in Arabidopsis, but SPL10's role in regulating leaf curvature has not been well characterized. In this study, we observed an upward-curling leaf phenotype upon overexpression of a miR156/157-resistant version of SPL10 (rSPL10). DNA affinity purification sequencing (DAP-seq), RNA-seq, and transient transactivation assays revealed that SPL10 binds to the promoter of REVOLUTA (REV). Consistent with binding assays, overexpression of a miR165/166-resistant version of REV (rREV) in WT plants caused an upward-curling leaf phenotype. Conversely, overexpression of rSPL10 in a rev-6 mutant couldn't rescue the downward-curling leaf phenotype of rev-6. Notably, SPL10 physically interacts with REV, attenuating the upward-curling leaf phenotype of rREV mutants, suggesting a role as a molecular rheostat to prevent overamplification of REV transcripts. Furthermore, overexpression of the BrpREV-1 gene caused rosette leaves to change from flat to upward curving and accelerated heading in Chinese cabbage. Taken together, our findings identify the role of the SPL10-REV module in regulating leaf curvature in Arabidopsis and the crucial functions of BrpREV-1 in the heading of Chinese cabbage.

Project description:Transcriptome changes during leaf growth and development were profiled. We extracted RNA from Arabidopsis leaf number six from plants growing in stable and controlled experimental conditions at four different growth stages both at the end-of-day (EOD) and end-of-night (EON). The experiment was performed in long-day conditions with optimal soil water content. Four growth stages were selected for analysis: stage 1, with maximum relative area and thickness expansion rates coinciding with leaf emergence; stage 2, maximum area and thickness absolute expansion rates; stage 3, decreasing leaf area and thickness expansion rates, and stage 4, end of leaf area and thickness expansions.
