Project description:Roots of Arabidopsis thaliana do not engage in symbiotic association with mycorrhizal fungi but host taxonomically diverse fungal communities that influence health and disease states. We sequenced the genomes of 41 isolates representative of the A. thaliana root mycobiota for comparative analysis with 79 other plant-associated fungi. We report that root mycobiota members evolved from ancestors having diverse lifestyles and retained diverse repertoires of plant cell wall-degrading enzymes (PCWDEs) and effector-like small secreted proteins. We identified a set of 84 gene families predicting best endophytism, including families encoding PCWDEs acting on xylan (GH10) and cellulose (AA9). These genes also belong to a core transcriptional response induced by phylogenetically-distant mycobiota members in A. thaliana roots. Recolonization experiments with individual fungi indicated that strains with detrimental effects in mono-association with the host not only colonize roots more aggressively than those with beneficial activities but also dominate in natural root samples. We identified and validated the pectin degrading enzyme family PL1_7 as a key component linking aggressiveness of endophytic colonization to plant health.

Project description:Transcriptional profiles were carried out for four transcription factors and two receptor-like kinases mutants, corresponding to genes that are co-expressed in silico with the CESA7 gene in Arabidopsis. These mutants show enhanced disease susceptibility to the necrotrophic fungal pathogen Plecthosphaerella cucmerina. Several gene families of small secreted peptides, some of them with known hormone or signalling implications, were commonly up-regulated in all of the 6 genotypes, unveiling a functional implication of these secreted peptides in the defence pathway against fungi. Transcriptomics of 6 mutant plants, including four transcription factors and two receptor-like kinases, were compared to their background Col-0 accession. For this porpoise, plants were grown as described and leaves from three plants of each genotype were mixed and extracted as one single replicate. Three replicates from three different sets of plants were analysed for each genotype.

Project description:Arbuscular mycorrhizas (AM) are the most common symbiotic associations between plant’s root compartment and fungi. They can provide both nutritional benefit (mostly inorganic phosphate) leading to improved growth and non-nutritional benefits including defense responses to environmental cues throughout the host plant, which in return delivers carbohydrates to the symbiont. However how transcriptional changes occurring in AM leaves differ from those induced by phosphate fertilization is poorly understood. We investigated systemic changes in the leaves of mycorrhized Medicago truncatula compared to non-mycorrhized control plants, and in phosphate fertilized plants compared to control plants.

Project description:Most land plants form beneficial associations with arbuscular mycorrhizal (AM) fungi which improves mineral nutrition, mainly phosphorus and nitrogen in the host plant in exchange for photosynthetically fixed carbon. Most of our knowledge on the AM symbiosis derives from dicotyledonous species. We show that inoculation with the AM fungus Funneliformis mossease stimulates growth and increases Pi content in leaves of the rice cultivar Loto (O. sativa ssp japonica). Although rice is a host for AM fungi, the molecular mechanism underlying the AM symbiosis, in particular the systemic transcriptional responses of shoots to AM inoculation, remain largely elusive. Transcript profiling of the shoots indicated the systemic induction of genes involved in the biosynthesis of phospholipids (phosphoinositides, inositol polyphosphates) and down-regulation of non-phosphorus lipids (galactolipids, sulfolipids) in leaves of mycorrhizal rice. Regulation of phospholipid biosynthesis genes appears to be coordinated with a reduced expression of genes involved in jasmonic acid and ethylene biosynthesis and signaling. Genes involved in phosphate starvation responses and remobilization of Pi were also found to be down-regulated in leaves of mycorrhizal rice. These results demonstrated that the AM symbiosis is accompanied by complex alterations in gene expression in shoots which are potentially important to maintain a stable symbiotic relationship in rice plants.

Project description:Arbuscular mycorrhiza (AM) interactions between plants and Glomeromycota fungi primarily support phosphate aquisition of most terrestrial plant species. To unravel cell-type specific gene expression during early stages of Medicago truncatula root colonization by AM fungi, we used genome-wide transcriptome profiling based on laser-microdissected cells. We used Medicago GeneChips to detail the cell-type specific programme of gene expression in early stages of colonization by arbuscular mycorrhizal fungi and identified genes differentially expressed during these stages. Medicago truncatula Gaertn M-bM-^@M-^XJemalongM-bM-^@M-^Y genotype A17 plantlets were grown in the climate chamber. Plants grown for the collection appressorial root areas (APP) and the corresponding non-appressorial root areas (NAP) were mycorrhized after 3 weeks and roots were harvested at 5-6 dpi.

Project description:Endophytic fungi are fungi that live inside the roots of plants. They can promote plant growth through a variety of direct and indirect mechanisms. Direct mechanisms include the production of phytohormones, such as auxin and gibberellins, which can stimulate plant growth. Endophytic fungi can also fix nitrogen, solubilize phosphate, and produce siderophores, which are compounds that chelate iron and make it available to plants. In addition, some endophytic fungi produce antimicrobial metabolites that can protect plants from pests and pathogens. Indirect mechanisms include the induction of systemic resistance, which is a plant's ability to defend itself against pests and pathogens. Endophytic fungi can also help plants to tolerate abiotic stresses, such as drought, salinity, and heavy metals. In this study, we used a proteomic approach to identify the proteins that are expressed in rice plants after they are treated with endophytic fungi. We found that the treatment with endophytic fungi resulted in the expression of a number of proteins involved in plant growth, stress response, and defense. These results suggest that endophytic fungi can promote plant growth and improve plant resilience to stress.

Illinois protein lines (IPL) comprise lines known as Illinois high protein (IHP), Illinois low protein (ILP), Illinois reverse-low protein (IRLP) and Illinois reverse-high protein (IRHP) lines. These lines represent over a century old experiment containing lines that represent various levels of nitrogen metabolism potential as a function of their grain protein concentration. Project description:The study investigated the impact of environment on the composition of the gut microbiota and mucosal immune development and function at gut surfaces in early and adult life. Piglets of similar genotype were reared in indoor and outdoor environments and in an experimental isolator facility. Mucosa-adherent microbial diversity in the pig ileum was characterized by sequence analysis of 16S rRNA gene libraries. Host-specific gene responses in gut ileal tissues to differences in microbial composition were investigated using Affymetrix microarray technology and Real-time PCR. Experiment Overall Design: Animals were reared on the sow at an outdoor or indoor facility. Additional piglets from the indoor facility were transferred to individual isolator units at 24 hours of age, and given a daily dose of antibiotic cocktail for the duration of the study. Piglets were weaned at day 28. From day 29 onwards, piglets were fed creep feed ad libitum. Ileal tissue samples were excised from N=6 piglets per group at day 5, 28 and 56.
