Description and determinants of the faecal resistome and microbiome of farmers and slaughterhouse workers: a metagenome-wide cross-sectional study.

Project description:Study generating and describing the faecal metagenomes of 194 persons occupationally exposed to antimicrobial resistance in livestock (including 46 control subjects). Highlights: - DNA of faecal samples of 194 persons occupationally exposed to antimicrobial resistance (AMR) in livestock, i.e. persons living or working on pig and poultry farms and pig slaughterhouse workers and control subjects, was sequenced and metagenomically analysed. - The faecal resistomes and microbiomes of farmers and slaughterhouse workers were described and compared between occupationally exposed groups and controls. - We found an increased ARG carriage in persons working in the Dutch pork production chain as compared to poultry farmers and controls. - Significant differences were found in the resistome and bacteriome composition of pig and pork exposed workers compared to a control group, as well as within-population (farms, slaughterhouse) compositional differences. - On-farm working hours and working or living on a pig farm (versus poultry farm) are determinants for the human faecal resistome. - Direct or indirect contact with AMR in livestock may be a determinant for human ARG carriage.

Project description:The objective of this study was to identify key genes associated with porcine muscle growth and adipose metabolism which different expression in porcine backfat tissue between Fat Type Pig(Taihu pig) and Lean Type Pig (Landrace), among developmental phases(Month 1,2,3,4,5). The gene expression analyses will increase understanding of impact factors of pork quality by identifying key genes and pathways controlling backfat development. Relative real-time RT-PCR was used to confirm differential expression of 3 different expression genes(ME,SCD and UCP3) which normalized by 3 housekeep genes(ACTB, TBP and TOP2B). Project description:To understand the molecular basis of distinct pork quality in Chinese indigenous and Western breed, longissimus dorsi samples were collected from three adult Northeastern Indigenous and from three adult Large White. Total RNA was extracted and subjected to porcine Affymetrix Genechip. The study helps to elucidate the genetic mechnism of divergent pork quality and provide the theory basis for selection and genetic improvement of meat quality traits in porcine. Six longissimus dorsi samples were collected from three Northeastern Indigenous and from three Large White. Three Large White were control samples. Total RNA was extracted from each sample.Gene-expression profiling was performed for each RNA sample separately on the GeneChipÂ® Porcine Genome Array at CapitalBio Corporation (Beijing, China).

Pig peri-implantation embryo development transcriptional profiling

Project description:A novel porcine cDNA microarray, containing PCR products selected for embryonic expression, was used for transcriptional profiling during conceptus development. Total RNA from quadruplicate samples of small spherical (1-5mm), large spherical (6-11mm), tubular (13-45mm), and filamentous (>100mm) was extracted, amplified, and converted to 32P labeled target and hybridized to membranes. Porcine peri-implantation embryo development order is from small spherical to large spherical, then to Tubular, then to filamentous. All this morphological transmission is within two days (day 10 to day 12). In this study, we investigated the gene expression changes by a specifically selected porcine embryo gene array. A number of genes were found to be differentially expressed during those developmental stages and confirmed by quantitative PCR.
