Deciphering Black Extrinsic Tooth Stains composition in Children using metaproteomics

Project description:The present study focuses on the use of a metaproteomic approach to analyse Black Extrinsic Tooth Stains, a specific type of pigmented extrinsic substance, in a cohort of 96 Children. Metaproteomics is a powerful emerging technology that successfully enabled human protein and bacterial identification of this specific dental biofilm using mass spectrometry. 1600 bacterial proteins were identified in black stains (BS) samples and 2058 proteins in dental plaque (DP) samples whereas 607 and 582 human proteins identified in (BS and DP, respectively). 132 genera bacteria in black stains and dental plaque were identified using phylopeptidomic analysis, showing prevalence of Rothia, Kingella, Nesseria and Pseudopropionibatcterium in black stains samples. We additionally confirmed the metaproteomic approach by performing 16S rRNA. In this work, we showed an interesting diversity of the microbiota and proteome including significant difference between Black stain and dental plaque samples.

Remediation of adult black dental stains by phototherapy.

Project description:Introduction and aims:This study investigates the effects of the application of antimicrobial phototherapy on black-stained tooth surfaces. Key variables were area, color and time taken to reappearance of black stain following dental prophylaxis with and without antimicrobial phototherapy. Differences in bacterial composition of black stain (specifically Aggregatibacter actinomycetemcomitans, Tannerella forsythia, and Porphyromonas gingivalis) were analyzed. Material and methods:The phototherapy device used was GLO™ Science LLC, New York, America, emitting light at 475?nm and a power of 3?W. Light was applied in 2 cycles of 8?min on 31 volunteers. Microbial identification was carried out on DNA extracted from black plaque. This study is registered with ClinicalTrials.gov, number NCT03309748. Results:Following antimicrobial phototherapy, 64.5% of patients displayed reduced pigmentation area. Plaque color was lighter in 48.4% of subjects. Pigmentation area and depth of color returned to normal levels during the course of the study. Colonization by the three bacterial species decreased, although the changes were not statistically significant. We report a key novel finding showing elevated levels of colonization by Tannerella forsythia (83.9%) in adult black stain. Conclusions:Application of phototherapy results in a reduction in area, color and bacterial colonization of black plaque in adults. The changes were not found to be statistically significant, perhaps owing to the low illumination power of the home-whitening device. For the first time, we document the elevated presence of Tannerella forsythia in adult black stain. We also demonstrate the potential application of a commercially available home-whitening device for black plaque treatment.

Dark stains on rock surfaces in Driny Cave (Little Carpathian Mountains, Slovakia).

Project description:Mycobiota are important in underground ecology. In 2014, we discovered dark stains on clayey sediments on the walls of Driny Cave, Slovakia. Our description is based on the morphology of the fungus and the phylogenetic relationships of the internal transcribed spacer (ITS) region. In addition, data on its capacity for the production of extracellular enzymes, growth, and survival in vitro at different temperatures are reported. Our analyses revealed that this dark stains on the wall was produced by Penicillium glandicola. The fungus was able to synthesize amylases, proteases and cellulases, but not pectinases and keratinases. The vegetative structures of mycelium of this fungus are viable in vitro after storage at cool temperatures (from -72 to 5 °C), and show active growth at temperatures from 5 to 25 °C, but without spore germination, and without active growth at 30 and 37 °C. Penicillium glandicola is a psychrotolerant species and belong to var. glandicola.
