Transcription profiling of sugarcane mosaic virus (SCMV) resistance in maize

Project description:The molecular mechanisms underlying the development and progression of sugarcane mosaic virus (SCMV) infection in maize are poorly understood. A transcript profiling study based on maize unigene-microarrays was conducted to identify genes associated with SCMV resistance in the near isogenic line (NIL) pair F7+ (SCMV resistant) and F7 (susceptible). Four comparisons were conducted, which is addressing constitutive genetic discrepancy (Non-infected F7 vs. Non-infected F7+), inducible genetic discrepancy (Infected F7 vs. Infected F7+), compatible reaction (Non-infected F7 vs. Infected F7), and incompatible reaction (Non-infected F7+ vs. Infected F7+).

Relationship between Sugarcane <i>eIF4E</i> Gene and Resistance against <i>Sugarcane Streak Mosaic Virus</i>.

Project description:Sugarcane mosaic disease, mainly caused by Sugarcane streak mosaic virus (SCSMV), has serious adverse effects on the yield and quality of sugarcane. Eukaryotic translation initiation factor 4E (eIF4E) is a natural resistance gene in plants. The eIF4E-mediated natural recessive resistance results from non-synonymous mutations of the eIF4E protein. In this study, two sugarcane varieties, CP94-1100 and ROC22, were selected for analysis of their differences in resistance to SCSMV. Four-base missense mutations in the ORF region of eIF4E resulted in different conserved domains. Therefore, the differences in resistance to SCSMV are due to the inherent differences in eIF4E of the sugarcane varieties. The coding regions of eIF4E included 28 SNP loci and no InDel loci, which were affected by negative selection and were relatively conserved. A total of 11 haploids encoded 11 protein sequences. Prediction of the protein spatial structure revealed three non-synonymous mutation sites for amino acids located in the cap pocket of eIF4E; one of these sites existed only in a resistant material (Yuetang 55), whereas the other site existed only in a susceptible material (ROC22), suggesting that these two sites might be related to the resistance to SCSMV. The results provide a strong basis for further analysis of the functional role of eIF4E in regulating mosaic resistance in sugarcane.

Genetic diversity and biological characterization of sugarcane streak mosaic virus isolates from Iran.

Project description:Sugarcane streak mosaic virus (SCSMV; genus Poacevirus, family Potyviridae) is a major causal agent of sugarcane mosaic disease in Asia. A survey of SCSMV was conducted in cultivated fields in Khuzestan province, southwestern Iran. Sixty-five sugarcane leaf samples showing mosaic symptoms were collected and investigated by RT-PCR. Almost one-fourth of the samples were found to be infected by SCSMV. To verify molecular variability, 12 SCSMV isolates were sequenced and analyzed by comparing partial NIb-CP gene sequences. The nucleotide identity among Iranian isolates was 83.1-99.8%, indicating high nucleotide variability, while amino acid identity was 95.2-100%, which suggesting selection for amino acid conservation. They shared nucleotide identities of 76.2-99.1% with those of other SCSMV isolates available in GenBank, the highest with isolates from Pakistan (PAK), India (IND671) and China (M117, KT257289). Further analysis was conducted based on complete CI coding region to gain more insight into the phylogenetic relationships of Iranian SCSMV compared to those from other Asian countries. Iranian isolates shared identities of 79.8-89.0% (nucleotide) and 94.8-98.6% (amino acid) with those from other geographical regions in the CI gene. The highest nucleotide identity of Iranian isolates was with isolates PAK (Pakistan), M121 (JQ975096, China) and IND671 (India), respectively. The phylogenetic trees (based on CI and NIb-CP) revealed the segregation of SCSMV isolates into two major divergent evolutionary lineages that reflect geographical origin of the isolates (with minor exception). Phylogenetic analyses grouped Iranian SCSMV isolates together with isolates from Pakistan, India and just one Chinese isolate in group II. Biological results showed that Iranian SCSMV isolates infect sugarcane, sorghum, maize and some wild grasses, causing mosaic symptoms on the leaves.

Identification of the RNA silencing suppressor activity of sugarcane streak mosaic virus P1 gene.

Project description:Sugarcane streak mosaic virus (SCSMV) belonging to Poacevirus, is a causative virus of mosaic disease in sugarcane in many Asian countries with substantial genomic variation. Although the virus infects the crop with Sugarcane mosaic virus (SCMV) a Potyvirus, it predominates over SCMV in spread as well as titre. We have taken up detailed studies to identify the functional activity of viral suppressors of SCSMV genome. Transient expression assay was performed with SCSMV-P1 and HC-Pro genes in the model plant Nicotiana tabacum to establish suppressor role of these genes. The plasmid constructs of both the genes were co-infiltrated with the reporter green fluorescent protein (GFP) and the suppressor activity was measured as enhancement in the GFP fluorescence. Further, the phenotypic expressions were validated by respective gene expression through semi quantitative and qRT-PCR. In the P1 co-infiltrated GFP leaves, suppression in the PTGS mechanism took place that allowed a long term expression of GFP. However, GFP co-infiltrated with HC-Pro did not sustain the GFP expression level for a prolonged period and the expression level was close to GFP control. The study concluded that unlike in other Potyviridae genera, P1 gene of SCSMV is playing the role of RNA silencing suppressor. This study helps in unveiling a new and promising way to understand the regulatory pathway in the host at the time of viral infection. Targeting the P1 gene of SCSMV through RNA silencing approach will be a viable strategy to develop mosaic resistant transgenic sugarcane varieties as they are directly involved in counter defence against the host.

Project description:Using nylon filter arrays, we analyzed the expression profile of 1536 expressed sequence tags (ESTs) of sugarcane (Saccharum sp.) exposed to the phytormone methyl jasmonate (MeJA) for 0-12 h. 26 ESTs were differentially expressed, including novel genes and also genes that had not previously been reported as being MeJA-inducible.
