Project description:Phytophthora cinnamomi is one of the most invasive tree pathogens that devastates wild and cultivated forests. Due to its wide host range, knowledge of the infection process at the molecular level is lacking for most of its tree hosts. To expand the repertoire of studied Phytophthora-woody plant interactions and identify molecular mechanisms that can facilitate discovery of novel ways to control its spread and damaging effects, we focused on the interaction between P. cinnamomi and sweet chestnut (Castanea sativa), an economically important tree for the wood processing industry. By using a combination of proteomics, metabolomics, and targeted hormonal analysis, we mapped the effects of P. cinnamomi attack on stem tissues immediately bordering the infection site and away from it. P. cinnamomi led to a massive reprogramming of the chestnut proteome and accumulation of the stress-related hormones salicylic acid (SA) and jasmonic acid (JA) indicating that stem inoculation can be used as an easily accessible model system to identify novel molecular players in P. cinnamomi pathogenicity

Project description:Chinese Chestnut (Castanea mollissima BL.) has high nutritional and ecological value, so it is widely planted.However, the number of male flowers far exceeds the number of female flowers is an important factor limiting the yield of Chinese chestnut.A naturally occurring mutation in male catkin has been found on a Chinese chestnut tree in the mountains of Beijing, China.This mutant is shorter than the normal catkin.Studies have shown that this mutation can significantly reduce the number of male flowers, promote the differentiation of female flowers, and improve the yield of Chinese chestnut.In this research, we provided the RNA sequencing data of two groups of mutants and normal catkin.The comparative analyses of catkins transcriptomes presented here provides a valuable resource for discovery of genes and networks involved in the regulate the sex of the flower.

Project description:Gene flow between cultivated and wild gene pools is common in the contact zone between agricultural lands and natural habitats and can be used to study the development of adaptations and selection of novel varieties. This is likely the case in the northern Adriatic region, where centuries-old cultivated orchards of sweet chestnut (Castanea sativa Mill.) are planted within the natural distribution area of the species. Thus, we investigated the population structure of several orchards of sweet chestnuts. Furthermore, the genetic background of three toponymous clonal varieties was explored. Six genomic simple sequence repeat (gSSR) and nine EST-derived SSR (EST-SSR) loci were utilized in this research, and both grafted and non-grafted individuals were included in this study. Five closely related clones were identified, which represent a singular, polyclonal marron variety, found in all three cultivation areas. Furthermore, many hybrids, a result of breeding between cultivated and wild chestnuts, have been found. Analyzed semi-wild orchards defined by a diverse genetic structure, represent a hotspot for further selection and could result in creation of locally adapted, high-yielding varieties.

Project description:The main threats to chestnut in Europe are climate change and emerging pathogens. Although many works have separately addressed the impacts on chestnut of elevated temperatures and Phytophthora cinnamomi Rands (Pc) infection, none have studied their combined effect. The objectives of this work were to describe the physiology, secondary metabolism and survival of 6-month-old C. sativa seedlings after plants were exposed to ambient temperature, high ambient temperature and heat wave events, and subsequent infection by Pc. Ten days after the warming scenarios, the biochemistry of plant leaves and roots was quantified and the recovery effect assessed. Plant growth and root biomass under high ambient temperature were significantly higher than in plants under ambient temperature and heat wave event. Seven secondary metabolite compounds in leaves and three in roots were altered significantly with temperature. Phenolic compounds typically decreased in response to increased temperature, whereas ellagic acid in roots was significantly more abundant in plants exposed to ambient and high ambient temperature than in plants subjected to heat waves. At recovery, leaf procyanidin and catechin remained downregulated in plants exposed to high ambient temperature. Mortality by Pc was fastest and highest in plants exposed to ambient temperature and lowest in plants under high ambient temperature. Changes in the secondary metabolite profile of plants in response to Pc were dependent on the warming scenarios plants were exposed to, with five compounds in leaves and three in roots showing a significant 'warming scenario' × 'Pc' interaction. The group of trees that best survived Pc infection was characterised by increased quercetin 3-O-glucuronide, 3-feruloylquinic acid, gallic acid ethyl ester and ellagic acid. To the best of our knowledge, this is the first study addressing the combined effects of global warming and Pc infection in chestnut.

Project description:The phyllosphere supports a large and complex bacterial community that varies both across plant species and geographical locations. Phyllosphere bacteria can have important effects on plant health. The sweet chestnut (Castanea sativa Mill.) is an economically important tree species affected worldwide by the fungal pathogens Cryphonectria parasitica and Phytophthora cinnamomi. We examined the culturable phyllosphere bacterial community of the sweet chestnut at two nearby locations in Central Spain in order to know its geographical variability and to explore its potential as source of biological control agents against these two pathogenic fungi. The bacterial diversity at strain level was high but it varied significantly between locations; however, phylotype richness and diversity were more comparable. The isolates were affiliated with the phyla Actinobacteria, Firmicutes and Proteobacteria. Most of them were members of recognized bacterial species, with a notable proportion of representative of the genera Dietzia and Lonsdalea, but a small fraction of the strains revealed the existence of several potential novel species or even genera. Antagonism tests showed the occurrence in the chestnut phyllosphere of bacterial strains potentially useful as biological control agents against the two pathogenic fungi, some of which belong to species never before described as fungal antagonists. Chestnut phyllosphere, therefore, contains a great diversity of culturable bacteria and may represent an untapped source of potential biocontrol agents against the fungi causing blight and ink diseases of this tree species.

Project description:UnlabelledBACKGROUND1471-2229-9-51: American chestnut (Castanea dentata) was devastated by an exotic pathogen in the beginning of the twentieth century. This chestnut blight is caused by Cryphonectria parasitica, a fungus that infects stem tissues and kills the trees by girdling them. Because of the great economic and ecological value of this species, significant efforts have been made over the century to combat this disease, but it wasn't until recently that a focused genomics approach was initiated. Prior to the Genomic Tool Development for the Fagaceae project, genomic resources available in public databases for this species were limited to a few hundred ESTs. To identify genes involved in resistance to C. parasitica, we have sequenced the transcriptome from fungal infected and healthy stem tissues collected from blight-sensitive American chestnut and blight-resistant Chinese chestnut (Castanea mollissima) trees using ultra high throughput pyrosequencing.ResultsWe produced over a million 454 reads, totaling over 250 million bp, from which we generated 40,039 and 28,890 unigenes in total from C. mollissima and C. dentata respectively. The functions of the unigenes, from GO annotation, cover a diverse set of molecular functions and biological processes, among which we identified a large number of genes associated with resistance to stresses and response to biotic stimuli. In silico expression analyses showed that many of the stress response unigenes were expressed more in canker tissues versus healthy stem tissues in both American and Chinese chestnut. Comparative analysis also identified genes belonging to different pathways of plant defense against biotic stresses that are differentially expressed in either American or Chinese chestnut canker tissues.ConclusionOur study resulted in the identification of a large set of cDNA unigenes from American chestnut and Chinese chestnut. The ESTs and unigenes from this study constitute an important resource to the scientific community interested in the discovery of genes involved in various biological processes in Chestnut and other species. The identification of many defense-related genes differentially expressed in canker vs. healthy stem in chestnuts provides many new candidate genes for developing resistance to the chestnut blight and for studying pathways involved in responses of trees to necrotrophic pathogens. We also identified several candidate genes that may underline the difference in resistance to Cryphonectria parasitica between American chestnut and Chinese chestnut.
