Project description:Small proteins are ubiquitous in cells and are conserved in many species. With advances in genome sequencing technology and computational biology, more and more evidence indicate that small proteins perform important functions and participate in stress responses in both prokaryotes and eukaryotes. However, the mechanism of small proteins regulate stress response is unclearly. In this study we found an unknown function small protein Cmi7p rapidly reacted to acetic acid stress and increased its expression. Further study found that Cmi7p affected cell cycle process and increased the efficiency of cell division under stress.

LB-100 (PP2A Inhibitor) and Azenosertib (WEE1 Inhibitor) in Metastatic Colorectal Cancer Patients

Project description:This Phase Ib trial studies the side effects and best dose of LB-100 and azenosertib for the treatment of patients with metastatic colorectal cancer. Azenosertib blocks a protein that is involved in the repair of damaged DNA, this protein is called WEE1. Inhibiting WEE1 drives cancer cells into a state of cell division without repair of the damaged DNA, resulting in cell death. LB-100 has been shown to make anticancer drugs work better at killing cancer. LB-100 blocks a protein called PP2A. Blocking this protein increases the stress signals for the tumor cells that express PP2A. Research has shown that azenosertib and LB-100 may enhance each others effect when treating metastatic colorectal cancer.

Sfp1 regulates yeast cell growth and division through multiple promoter binding modes

Project description:Understanding how transcriptional programs help to coordinate cell growth and division is an important unresolved problem. Here we report that the nutrient- and stress-regulated transcription factor Sfp1 is rate-limiting for expression of a large suite of genes involved in yeast cell growth, including ribosomal protein, ribosome biogenesis, and snoRNA genes. Remarkably, the spectrum of Sfp1 transcription effects is concordant with a combination of chromatin immunoprecipitation and chromatin endogenous cleavage binding analyses, which together provide evidence for two distinct modes of Sfp1 promoter binding, one requiring a co-factor and the other a specific DNA-recognition motif. In addition to growth-related genes, Sfp1 binds to and regulates the promoters of key G1/S regulon genes and that of MRS6, whose product regulates Sfp1 nuclear localization. Our findings suggest that Sfp1 acts as a master regulator of cell growth and cell size by coordinating the expression of genes implicated in mass accumulation and cell division.

Project description:HU protein regulates expression of genes involved in cell-division, iron acquisition, and transcriptional regulation in Porphyromonas gingivalis Analysis involved RNA extracted from the parent strain W83 and the PG0121 (HU protein) mutant during exponential growth in rich medium. Each of the four experiments used a different preparation of RNA. Comparative expression profiles were determined by hybridizing the microarray slide with a mixture containing equal amount of cDNA generated from each strain. Dye swap experiments were performed.
