Prenatal Exposure to Metals Alters the Placental Inflammatory Proteome in a Sex-Dependent Manner in Extremely Low Gestational Age Newborns

Project description:Prenatal exposure to toxic metals is associated with altered placental function and adverse health outcomes. The underlying mechanisms linking in utero toxic metal exposures with later-in-life health remain unclear, though placental inflammation is posited as a potential driver. The aim of this study was to evaluate whether in utero metals presence is associated with sex-specific changes in placental protein expression. We hypothesized that sex-specific patterns of metal-associated placental protein expression would be observed, and metals presence would be positively associated with the altered expression of inflammation-associated pathways Using samples banked from the Extremely Low Gestational Age Newborn Study (ELGAN), umbilical cord tissue samples were analyzed via ICP-MS/MS for trace elements, and placental samples underwent a global untargeted proteomics analysis via LC-MS/MS. This work highlights the linkage between prenatal metals exposure and an altered placental proteome, revealing that metals in cord tissue were associated with largely distinct differences in placental protein expression, in a sexually-dimorphic manner.

Occurrence and risk assessment of trace metals and metalloids in sediments and benthic invertebrates from Dianshan Lake, China.

Project description:The present study measured concentrations of Cr, Ni, Cu, Zn, As, Cd, Sb, and Pb in surface sediments and two benthic invertebrate species (Anodonta woodiana and Bellamya aeruginosa) collected from Dianshan Lake, located in the Yangtze River Delta. The Dianshan Lake acts as one of the most important drinking water sources to Shanghai, the biggest city in China. Concentrations of trace metals and metalloids ranged from 0.04 mg/kg for Cd to 288.0 mg/kg for Zn. Substantial bioaccumulation in invertebrates was observed for Zn and Cu based on the biota-sediment accumulation factor (BSAF) measurements. The results revealed that concentrations of metals and metalloids in sediments from Dianshan Lake were at the lower end of the range of levels found in other regions of China. The assessment of three significantly inter-related evaluation indices, including the geo-accumulation Index (Igeo), potential ecological risk factor (Eri), and mean probable effect concentration quotients (Qm-PEC), suggested that sediment-associated trace elements exhibited no considerable ecological risks in the studied watershed. However, the target hazard quotient and hazard index analysis suggested that selected elements (particularly As) accumulation in edible tissues of benthic invertebrates could pose potential health risks to local populations, especially fishermen. Given that wild aquatic organisms (e.g., fish and bivalves) constitute the diet of local populations as popular food/protein choices, further investigations are needed to better elucidate human health risks from metal and metalloid exposure via edible freshwater organisms.

Transcriptomic responses of the endangered freshwater mussel Margaritifera margaritifera to trace metal contamination in the Dronne River, France

Project description:The freshwater pearl mussel Margaritifera margaritifera is one of the most threatened freshwater bivalves worldwide. In this study, we aimed (i) to study the processes by which water quality might affect freshwater mussels in situ and (ii) to provide insights into the ecotoxicological significance of water pollution to natural populations in order to provide necessary information to enhance conservation strategies. M. margaritifera specimens were sampled in two close sites located upstream or downstream from an illegal dumping site. The renal transcriptome of these animals was assembled and gene transcription determined by RNA-seq. Correlations between transcription levels of each single transcript and the bioaccumulation of 9 trace metals, age (estimated by sclerochronology) and condition index were determined in order to identify genes likely to respond to a specific factor. Amongst the studied metals, Cr, Zn, Cd and Ni were the main factors correlated with transcription levels, with effects on translation, apoptosis, immune response, response to stimulus and transport pathways. However, the main factor explaining changes in gene transcription appeared to be the age of individuals with a negative correlation with the transcription of retrotransposons-related genes. To investigate this effect further, mussels were classified into 3 age classes. In young, middle-aged and old animals, transcription levels were mainly explained by Cu, Zn and age, respectively. This suggests differences in the molecular responses of this species to metals during its lifetime that must be better assessed in future ecotoxicology studies.
