Project description:Soil qualities and rootstocks are among the main factors that have been acknowledged to influence grape development as well as fruit and wine composition. Despite the role of the soil and rootstock in establishing a successful vineyard in terms of grape quality, almost no molecular evidence linking soil and rootstock properties to the gene expression have been reported. The transcriptome variation in response to different soils and rootstocks was investigated through microarray technology. The cv. Pinot Noir was grown on different soils: sand, turf and vineyard soil. The plants were grafted on the contrasting 101-14 and 1103 Paulsen rootstocks. Project description:Genotype, climate, soil qualities, and rootstocks are among the main factors that influence grape development as well as fruit and wine composition. In this work, the rootstock/scion interactions were studied using transcriptomic and metabolic approaches on leaves of Gaglioppo variety, grafted onto thirteen different rootstocks growing in the same locations. The experiments were performed in a vineyard in the south of Italy, characterized by the Mediterranean climate with warm and dry summers. Whole leaf transcriptome of ‘Gaglioppo’ grafted on five selected rootstocks showed high variability in gene expression. In particular, significant modulation of transcripts linked to primary (e.g. carbohydrate and transport) and secondary metabolism (e.g. phenylpropanoids pathways and response to stress) was observed. Interestingly, genes involved in the priming defense responses (e.g stilbenes and defense genes) were strongly activated in some graft combinations. These results were further confirmed by the quantification of stilbene contents. According with these observations, the leaves of ‘Gaglioppo’ grafted on 1103 Paulsen showing the lowest levels of stilbene synthase transcripts and stilbene metabolites, reveal the greater sensitivity to downy mildew in in vitro assay. Project description:Basal leaf removal has been widely practiced to increase cluster sunlight exposure, control berry rot and eventually improve grape berry quality. Early leaf removal performed in cool region appeared to benefit the accumulation of norisoprenoids and monoterpenes. However, effects of this canopy management and leaf moving on volatile profiles and transcriptome of grape berry in warm region characterized with strong sunshine and arid climate were paid less attention. To cope with multiple possible effects caused by global warming, researches in warm region are urgently needed to provide a guide for adjusting the sunlight exposure treatment to adapt the climate change.In Manas county of Xinjiang province, sunlight exposure treatments performed in this study resulted in decreased β-carotene and lutein, which mainly responsible for the reduced norisoprenoids in ripening grapes. Substrate competition due to the up-regulation of VvTPS-a and VvNCED3 may contribute to the decreased concentration of monoterpenes in exposed berries. The notable increased C6 alcohols in the grape berries treated by leaf removal applied at veraison (LR-V), was mainly attribute to the enhanced substrate availability of linoleic acid and linolenic acid together with the higher expression of VvLOXO and VvADH1. Free C6 aldehyde was reduced by half leaf removal applied at veraison (HLR-V), leaf moving at veraison (LM-V) and leaf removal applied at berry pepper-corn size (LR-PS). Moreover, transcriptomic analysis indicated that both LR-V and LM-V treated grape berry implemented multiple stress-mitigation strategies to acclimate the improved sunlight exposure. Interestingly, photosynthesis-related genes in grape berry were primarily up-regulated by LR-V as a consequence of carbohydrate source removal rather than improved sunlight exposure. Weighted gene co-expression network analysis (WGCNA) suggested that the genes encoding malate synthase and 3-oxoacyl-(acyl carrier protein) reductase show a significant correlation with the accumulation of C6 alcohol.

Project description:Grapevine is a perennial crop often cultivated by grafting a scion cultivar on a suitable rootstock. Rootstocks influence scions, particularly with regard to water uptake and vigor. Therefore, one of the possibilities to adapt viticulture to the extended drought stress periods is to select rootstocks conferring increased tolerance to drought. However, the molecular mechanisms associated with the ability of rootstock/scion combination to influence grape berry metabolism under drought stress are still poorly understood. The transcriptomic changes induced by drought stress in grape berries (cv. Pinot noir) from vines grafted on either 110R (drought tolerant) or 125AA (drought sensitive) rootstock were compared. The experiments were conducted in the vineyard for two years and two grape berry developmental stages (50% and 100 % veraison. The genome-wide microarray approach showed that water stress strongly impacts gene expression in the berries, through ontology categories that cover cell wall metabolism, primary and secondary metabolism, signalling, stress, and hormones, and that some of these effects strongly depend on the rootstock genotype. Indeed, under drought stress, berries from vines grafted on 110R displayed a different transcriptional response compared to 125AA concerning genes related to jasmonate, phenylpropanoid metabolism and PR-proteins. The data also suggests a link between jasmonate and secondary metabolism in water-stressed berries. Overall, genes related to secondary metabolism and jasmonate are more induced and/or less repressed by drought stress in the berries grafted on the drought-sensitive rootstock 125AA. These rootstock-dependent gene expression changes are relevant for berry composition and sensory properties.
