Project description:Unanswered key questions in bark beetle-plant interactions concern host finding in species attacking angiosperms in tropical zones and whether management strategies based on chemical signaling used for their conifer-attacking temperate relatives may also be applied in the tropics. We hypothesized that there should be a common link in chemical signaling mediating host location by these Scolytids. Using laboratory behavioral assays and chemical analysis we demonstrate that the yellow-orange exocarp stage of coffee berries, which attracts the coffee berry borer, releases relatively high amounts of volatiles including conophthorin, chalcogran, frontalin and sulcatone that are typically associated with Scolytinae chemical ecology. The green stage of the berry produces a much less complex bouquet containing small amounts of conophthorin but no other compounds known as bark beetle semiochemicals. In behavioral assays, the coffee berry borer was attracted to the spiroacetals conophthorin and chalcogran, but avoided the monoterpenes verbenone and ?-pinene, demonstrating that, as in their conifer-attacking relatives in temperate zones, the use of host and non-host volatiles is also critical in host finding by tropical species. We speculate that microorganisms formed a common basis for the establishment of crucial chemical signals comprising inter- and intraspecific communication systems in both temperate- and tropical-occurring bark beetles attacking gymnosperms and angiosperms.

Using subtracted cDNA libraries and nylon filter arrays, we analyzed the expression profile of 1536 expressed sequence tags (ESTs) of coffee plants from an hybrid progeny (C. arabica x C. racemosa), containg resistant (R) and susceptible plants (S) to the infestation of coffee leaf miner. Leaf discs were collected from non-infested plants (R control - RC; S control - SC), infested plants after moth oviposition (R oviposition - Ro; S oviposition - So) and infested after larvar eclosion (R eclosion - Re; S eclosion - Se). Project description:The coffee berry borer is the most devastating insect pest of coffee throughout the world. The insect spends most of its life cycle inside the coffee berry, which makes it quite difficult to observe its behaviour. Micro-computed tomography (micro-CT) was used to observe all developmental stages of the coffee berry borer inside coffee berries (Coffea canephora). An interesting oviposition pattern involving a sequential placement of eggs starting in the periphery of the seed and moving inwards was observed. Micro-CT should be useful in elucidating unknown life history aspects of other seed-feeding bark beetles as well as of bark and ambrosia beetles in general.

Project description:Coffee, after petroleum, is the most valuable commodity globally in terms of total value (harvest to coffee cup). Here, our bioeconomic analysis considers the multitude of factors that influence coffee production. The system model used in the analysis incorporates realistic field models based on considerable new field data and models for coffee plant growth and development, the coffee/coffee berry borer (CBB) dynamics in response to coffee berry production and the role of the CBB parasitoids and their interactions in control of CBB. Cultural control of CBB by harvesting, cleanup of abscised fruits, and chemical sprays previously considered are reexamined here to include biopesticides for control of CBB such as entomopathogenic fungi (Beauveria bassiana, Metarhizium anisopliae) and entomopathogenic nematodes (Steinernema sp., Heterorhabditis). The bioeconomic analysis estimates the potential of each control tactic singly and in combination for control of CBB. The analysis explains why frequent intensive harvesting of coffee is by far the most effective and economically viable control practice for reducing CBB infestations in Colombia and Brazil.

Project description:The coffee industry loses millions of dollars annually worldwide due to the Coffee Berry Borer (CBB); these losses imply a decrease in quality and production. Traps are used to monitor their flight and for pest control. The main objective of this study was to determine the capture pattern and trap capture percentages of the CBB population over time using column traps (CTs) in two independent field experiments. CTs were composed of four traps installed at four different heights 0.5, 1.5, 2.5, and 3.5 m above ground. Our results demonstrated a significant difference in CBB capture by traps placed at different heights above the ground. The CT capture maintained a pattern throughout this study's lag: the lower the height, the greater the percentage of CBBs captured. The study was conducted in two independent experiments (A and B). In Experiment A and B, the traps placed at 0.5 m caught 67% and 85% of the CBBs captured, respectively. Furthermore, the trap set at 1.5 m above the ground in the multi-level CT showed a higher capture percentage than the single placed trap (ST, also at 1.5 m about ground). The pattern of the capture and proportion of the CBB in the CTs was maintained throughout the study despite the season, changes in temperature, and relative air humidity. We suggest that CTs could be explored as a useful tool for capturing the CBB, considering its monitoring and management.

Project description:The coffee berry borer, Hypothenemus hampei, is the most economically important insect pest of coffee worldwide. We present an analysis of the draft genome of the coffee berry borer, the third genome for a Coleopteran species. The genome size is ca. 163 Mb with 19,222 predicted protein-coding genes. Analysis was focused on genes involved in primary digestion as well as gene families involved in detoxification of plant defense molecules and insecticides, such as carboxylesterases, cytochrome P450, gluthathione S-transferases, ATP-binding cassette transporters, and a gene that confers resistance to the insecticide dieldrin. A broad range of enzymes capable of degrading complex polysaccharides were identified. We also evaluated the pathogen defense system and found homologs to antimicrobial genes reported in the Drosophila genome. Ten cases of horizontal gene transfer were identified with evidence for expression, integration into the H. hampei genome, and phylogenetic evidence that the sequences are more closely related to bacterial rather than eukaryotic genes. The draft genome analysis broadly expands our knowledge on the biology of a devastating tropical insect pest and suggests new pest management strategies.

Project description:Traditionally, the study of anatomy in insects has been based on dissection techniques. Micro-computed tomography (micro-CT) is an X-ray based technique that allows visualization of the internal anatomy of insects in situ and does not require dissections. We report on the use of micro-CT scans to study, in detail, the internal structures and organs of the coffee berry borer (Hypothenemus hampei), the most damaging insect pest of coffee worldwide. Detailed images and videos allowed us to make the first description of the aedeagus and the first report of differences between the sexes based on internal anatomy (flight musculature, midgut shape, hindgut convolutions, brain shape and size) and external morphology (lateral outline of the pronotum and number of abdominal tergites). This study is the first complete micro-CT reconstruction of the anatomy of an insect and is also the smallest insect to have been evaluated in this way. High quality rendered images, and additional supplementary videos and 3D models are suitable for use with mobile devices and are useful tools for future research and as teaching aids.
