Project description:The aim of this experiment is to characterize the genome composition of the two hybrids IF6 and MR25. IF6 and MR25 are hybrids among S.cerevisiae and S. kudriavzevii. The hybridization of the genome extraction with the microarray was at 65C, under this condition only S. cerevisiae genes can hybridize. In this case we are only studying the genome composition of S. cerevisiae parental in the hybrids. The study of S. kudriavzevii parental genome was done by other methods.

Project description:BackgroundAroma is one of the most important attributes defining wine quality in which yeasts play a crucial role, synthesizing aromatic compounds or releasing odourless conjugates. A present-day trend in winemaking consists of lowering fermentation temperature to achieve higher aroma production and retention. S. cerevisiae × S. kudriavzevii hybrids seem to have inherited beneficial traits from their parental species, like fermenting efficiently at low temperature or producing higher amounts of certain aromatic compounds. In this study, allelic composition and gene expression of the genes related to aroma synthesis in two genetically and phenotypically different S. cerevisiae × S. kudriavzevii hybrids, Lalvin W27 and VIN7, were compared and related to aroma production in microvinifications at 12 and 28 °C. In addition, the contribution of the allele coming from each parental to the overall expression was explored by RT-PCR.ResultsThe results indicated large differences in allele composition, gene expression and the contribution of each parental to the overall expression at the fermentation temperatures tested. Results obtained by RT-PCR showed that in ARO1 and ATF2 genes the S. kudriavzevii allele was more expressed than that of S. cerevisiae particularly at 12 °C.ConclusionsThis study revealed high differences regarding allele composition and gene expression in two S. cerevisiae × S. kudriavzevii hybrids, which may have led to different aroma profiles in winemaking conditions. The contribution of the alleles coming from each parental to the overall expression has proved to differently influence aroma synthesis. Besides, the quantitative contribution to the overall gene expression of the alleles coming from one parental strain or the other was clearly determined by the fermentation temperature for some genes.

Project description:This research work investigates the hybridization of the genes of Saccharomyces bayanus var. uvarum CECT 12600 and S. kudriavzevii IFO1802 employing S. cerevisiae microarrays. Two strains,one S. kudriavzevii and one S. bayanus var. uvarum were employed for the study. The S. cerevisiae strain S288C was used as control for microarray hybridizations. All experiments were performed using duplicate arrays, and Cy5-dCTP and Cy3-dCTP dye-swap assays were performed to reduce dye-specific bias.

Comparison of natural yeast hybrids

Project description:Analysis of genomic variation in several Saccharomyces cerevisiae x S. kudriavzevii natural hybrids S. cerevisiae and S. kudriavzevii yeasts have been described among wine strains. These strains exhibit enological properties of interest for the wine industry. A preliminary characterization of these hybrids showed a trend to reduce the S. kudriavzevii fraction of the hybrid genome. As a first approach to understand the mechanisms involved in the genome changes occurred after hybridization that may account for differences in their enological properties, we characterized the genomic constitution of several wine S. cerevisiae x S. kudriavzevii strains by using a combined approach based on the RFLP analysis of gene regions, comparative genome hybridizations with S. cerevisiae DNA arrays, ploidy analysis and gene dose determination by quantitative real-time PCR. Genomic DNA hybridizations of w27, w46, SPG1691 and 441 hybrids were compared against the laboratory strain FY1679. Hybridizations were performed by triplicate in order to obtain the final CGH ratio.
