The transcriptional landscape of Nicotiana benthamiana and Verticillium dahliae interaction

Project description:Verticillium dahliae is a soil-borne fungus with a broad host range, including the model plants Nicotiana benthamiana and Arabidopsis thaliana. The plant immunity can be activated by Vd-derived patterns while V. dahliae secreted effectors. We report here the RNAseq analyses of samples from N. benthamiana infected by V. dahliae strains 592 and 171 at different time points. The data showed dynamic expression patterns of genes not only from plant defense signaling but also plant developmental signaling.

Genome-wide profiling of miRNAs and other small non-coding RNAs in the Verticillium dahliae inoculated cotton roots

Project description:MicroRNAs (miRNAs) and small interfering RNAs (siRNAs) are short (19â25 nucleotides) non-coding RNA molecules that have large-scale regulatory effects on development and on stress responses in plants.The objective of this study is to investigate the transcriptional profile of miRNAs and other small non-coding RNAs in Verticilliumâinoculated cotton roots. Four small RNA libraries were constructed from mocked and infected roots of two cotton cultured species which are with different Verticillium tolerance (âHai-7124â, Gossypium barbadense L., a Verticillium-tolerant cultivar, and âYi-11â, Gossypium hirsutum L. a Verticillium-sensitive cultivar). The length distribution of obtained small RNA pools was significantly different among libraries. A total of 215 conserved miRNA families were identified in the two cotton species, of them 14 are novel. There were >65 families with different expression between two libraries. We also identified two ta-siRNAs and thousands of endogenous siRNA candidates, and hundred of them exhibited altered expression after inoculation of Verticillium. The profiling of these miRNAs and other small non-coding RNAs lay the foundation for further understanding of small RNAs function in the regulation of Verticillium defence responses in cotton roots.

Transcriptome sequencing of Verticillium dahliae and △VdBre1 mutant

Project description:The soil-borne ascomycete Verticillium dahliae which causes wilt disease in many important crops results a severe reduction in crop yield and quality. During infection, the V.dahliae will secrete a lot of secondary metabolites which act as toxic factors to promote the infection process. However, the mechanism underlying how V.dahliae secondary metabolites regulate cotton infection remains largely unexplored. BRE1 is a highly conserved ubiquitin ligase (E3) enzymes and regulate expression of specific genes related to the production of secondary metabolites. To get an insight into the genes regulated by VdBre1, the comparative transcriptomic analysis was conducted between VdBre1 deletion mutant and the wild type V592. The conidia of wilde type and mutant were collected and total RNA was extracted using fungal RNA kit, sequencing was performed on an BGISEQ-500 platform.
