Project description:The aim of the study was to determine the influence of tissue degeneration due to prolonged storage of tissues on RNA integrity and the miRNA expression profile using microarrays. There is some dispute as to which RNA integrity level is still acceptable for miRNA expression profiling by microarrays. Some authors claim that miRNAs are stable even in degraded total RNA, others say the opposite. Therefore, we wanted to clarify if miRNA are stable in degraded tissues and which miRNAs are the most susceptible to RNA degradation.

Project description:Microglia seed the embryonic neuro-epithelium, expand and actively sculpt neuronal circuits in the developing CNS, but eventually adopt relative quiescence and ramified morphology in the adult. Here we probed the impact of post-transcriptional control by microRNAs (miRNAs) on microglial performance during development and adulthood by generating mice lacking microglial dicer expression at these distinct stages. RNA seq of conditional dicer ablation in adult microglia revealed that miRNAs were required to limit microglial responses to challenge but had only minor changes on non-challenged microglia. Specifically, following peripheral endotoxin exposure of the animals (LPS), Dicer-deficient microglia either from total brain or hippocampus overexpressed pro-inflammatory cytokines and as a result compromised hippocampal neuronal functions. In contrast, prenatal ablation resulted in spontaneous microglia activation and revealed Dicer involvement in DNA repair and preservation of genome integrity. Overall, our study shows miRNA and Dicer regulation of inflammatory response either following challenge in the adult or spontaneously in the newborn microglia. Moreover, Dicer is required for preservation of microglia DNA integrity required for their proliferation, longevity and radioresistance.

Project description:The neurovascular unit (NVU) is a complex multicellular structure that helps maintain cerebral homeostasis and blood-brain barrier (BBB) integrity. While extensive evidence links NVU alterations to cerebrovascular diseases and neurodegeneration, the underlying molecular mechanisms remain unclear. Here, we use zebrafish embryos carrying a mutation in Scavenger Receptor B2, a highly conserved endolysosomal protein expressed predominantly in Radial Glia Cells (RGCs), to investigate the interplay among different NVU components. Through live imaging and genetic manipulations, we demonstrate that compromised acidification of the endolysosomal compartment in mutant RGCs leads to impaired Notch3 signaling, thereby inducing excessive neurogenesis and reduced glial differentiation. We further demonstrate that alterations to the neuron/glia balance result in impaired VEGF and Wnt signaling, leading to severe vascular defects, hemorrhages, and a leaky BBB. Altogether, our findings provide novel insights into NVU formation and function and offer new avenues for investigating diseases involving white matter defects and vascular abnormalities. Project description:B cells are crucial for adaptive immunity, orchestrating humoral responses by producing antibodies essential for pathogen clearance. Here, we show that Poly(rC) binding protein 1 (Pcbp1), a multifunctional RNA-binding protein, is a key regulator of antibody production in B cells. Pcbp1 deficiency in B cells resulted in significant reductions in IgM expression at steady state and compromised differentiation of germinal center B cells and production of high-affinity antibodies upon immunization. These effects result from compromised mitochondrial integrity in Pcbp1-deficient B cells, including compromised mitochondrial electron transport chain (ETC) complex I and increased mitochondrial ROS production. Blocking mitochondrial ROS production rescued IgM production and improved germinal center responses caused by Pcbp1 deficiency. Mechanistically, Pcbp1 promotes the expression of Fdxr, a critical component of ETC complex I, via binding to its 3’UTR. Overall, our findings reveal a novel role for Pcbp1 in regulating mitochondrial function, protein synthesis, and antibody responses in B cells, providing novel insight into post-transcriptional regulation and mitochondrial functions.
