Project description:The presence of DNA methylation in beetles (Coleoptera) has only been investigated with bisulfite sequencing of Tribolium castaneum, which produced no evidence of DNA methylation. Here, we used whole genome bisulfite sequencing to assay if DNA methylation was present in another beetle, Nicrophorus vespilloides. We used T. castaneum as a negative control.

Project description:Bark beetles (Coleoptera: Scolytinae) are pests of many forests around the world. The mountain pine beetle (MPB), Dendroctonus ponderosae Hopkins, is a significant pest of western North American pine forests. The MPB is able to overcome the defences of pine trees through pheromone-assisted aggregation that results in a mass attack of host trees. These pheromones, both male and female produced, are believed to be biosynthesized in the midgut and/or fat body of these insects. We have used transcriptomics (RNA-seq) to identify transcripts differentially expressed between sexes and between tissues, with juvenile hormone III treatment, which is known to induce pheromone biosynthesis.
Project description:To allow estimation of the complexity of the antennal transcriptome between sexes as well as in comparison with a non-antennal tissue, microarrays were designed based on an assembly of new 454 sequence data from the antenna and publicly available data including 454 data from the larval midgut. The microarrays were hybridized with samples generated from the repsecitve tissues from 3 (antenna) and 5 (midgut) animals per sample, with four independent samples per sex (antenna) and tissue (midgut).

Female Tribolium castaneum beetles were either jabbed with sterile saline, heat killed Bt, or left naïve. 15 Offspring from the first and second broods were infected with Bt and 24 hours later pooled by treatment and flash frozen Beetles from saline and primed maternal backgrounds are compared to naïve infected Pooled whole larvae, custom array for T. castaneum, 1 replicate per treatment comparison. Same naïve sample used for both arrays
Project description:Polyp size and count determines the follow-up intervals after colonoscopy. However, relying on the endoscopist’s optical diagnosis for size estimation has shown considerable variability, leading to erroneous surveillance intervals and increased colorectal cancer risk. This study aims to assess the effectiveness of a new polyp size estimation software, called POSEIDON, which uses the tip of the auxiliary water-jet as reference and is implemented together with the EndoMind polyp detection system.
