Project description:T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematologic cancer derived from the malignant transformation of T-cell progenitors. Outcomes remain poor for T-ALL patients who have either primary resistance to standard-of-care chemotherapy or disease relapse. Notably, there are currently no targeted therapies available in T-ALL. This lack of next-generation therapies highlights the need for relevant preclinical disease modeling to identify and validate new targets and treatment approaches. Here, we adapted a spontaneously arising, genetically heterogeneous, thymic transplantation-based murine model of T-ALL, recapitulating key histopathological and genetic features of the human disease, to the preclinical testing of targeted and immune-directed therapies. Genetic engineering of the murine Notch1 locus aligned the spectrum of Notch1 mutations in the mouse model to that of human T-ALL and confirmed aberrant, recombination-activating gene (RAG)-mediated 5' Notch1 recombination events as the preferred pathway in murine T-ALL development. Testing of Notch1-targeting therapeutic antibodies demonstrated T-ALL sensitivity to different classes of Notch1 blockers based on Notch1 mutational status. In contrast, genetic ablation of Notch3 did not impact T-ALL development. The T-ALL model was further applied to the testing of immunotherapeutic agents in fully immunocompetent, syngeneic mice. In line with recent clinical experience in T-cell malignancies, programmed cell death 1 (PD-1) blockade alone lacked anti-tumor activity against murine T-ALL tumors. Overall, the unique features of the spontaneous T-ALL model coupled with genetic manipulations and the application to therapeutic testing in immunocompetent backgrounds will be of great utility for the preclinical evaluation of novel therapies against T-ALL.

Pre-clinical models for prediction of immunotherapy outcomes and immune evasion mechanisms in genetically heterogeneous multiple myeloma

Project description:Multiple myeloma (MM) is a neoplasia of bone marrow (BM) plasma cells that remains largely incurable with current treatment. Advancing therapeutic discoveries has been hampered by the lack of genetically heterogeneous models of MM. To circumvent this limitation, we engineered fifteen mouse models carrying combinations of eight MM genetic drivers, which fulfilled the pathogenesis of human disease including progression from premalignant states under immune surveillance. Integrative analyses of ⁓500 mice and ⁓1,000 patients revealed a MAPK-MYC genetic pathway that regulates time to progression and immune escape mechanisms across genetically heterogeneous tumors. During progression, MYC-dependent and independent remodeling of the BM microenvironment divided MM into immune categories with predominance of selected T-cell subpopulations, which dictated immunotherapy responses. Experimental targeting of the cytotoxic or immunosuppressive T-cell states observed in refractory MM patients enhanced immunotherapy effectiveness. Our resource enabled characterization of MM cell-intrinsic and immunological traits at unprecedented levels, which will accelerate the translation of personalized immunotherapy.

Characterization of a novel mouse model of undifferentiated pleomorphic sarcoma that is lymphocyte poor, macrophage rich, and resistant to immune checkpoint blockade

Project description:Sarcomas are rare, difficult to treat, mesenchymal lineage tumours that disproportionately affect children and young adults. Immunologically-based therapies have improved outcomes for numerous adult cancers, however, such approaches have proven ineffective for the majority of individuals with advanced sarcomas. Clinically relevant, immunologically-competent, and transplantable pre-clinical sarcoma models are needed in order to test and develop novel immunologically-based therapies for sarcoma. Herein we show that Cre- lox P-mediated simultaneous activation of Kras G12D , and deletion Trp53, in the hindlimb muscles of C57Bl/6 mice results in the development of highly penetrant, rapid onset undifferentiated pleomorphic sarcomas (UPS). The latter represents a model one of the most common human sarcoma subtypes both histologically and genetically. Furthermore, cell lines derived from the UPS tumors induced in C57Bl/6 mice can be transplanted into the hindlimbs or lungs of naïve, immune competent syngeneic mice. Immunological characterization of both the spontaneous and transplanted UPS tumours demonstrates an immunologically- ‘quiescent’ microenvironment, characterized by a paucity of lymphocytes and spontaneous adaptive immune pathways in addition to dense macrophage infiltrates. UPS cell line induced tumours demonstrate mildly increased lymphocytic infiltrates as compared with the spontaneous UPS tumours, furthermore, growth of transplanted UPS was unaltered in lymphocyte deficient hosts. Despite expression of PD-1 on the tumour infiltrating lymphocytes, UPS tumours were resistant to immunological checkpoint blockade. This syngeneic transplantable UPS model that recapitulates many of the immunological features of human UPS will prove invaluable for the preclinical development and evaluation of novel immunotherapeutic approaches for targeting sarcomas.

Cancer immunotherapy using checkpoint blockade.

Project description:The release of negative regulators of immune activation (immune checkpoints) that limit antitumor responses has resulted in unprecedented rates of long-lasting tumor responses in patients with a variety of cancers. This can be achieved by antibodies blocking the cytotoxic T lymphocyte-associated protein 4 (CTLA-4) or the programmed cell death 1 (PD-1) pathway, either alone or in combination. The main premise for inducing an immune response is the preexistence of antitumor T cells that were limited by specific immune checkpoints. Most patients who have tumor responses maintain long-lasting disease control, yet one-third of patients relapse. Mechanisms of acquired resistance are currently poorly understood, but evidence points to alterations that converge on the antigen presentation and interferon-? signaling pathways. New-generation combinatorial therapies may overcome resistance mechanisms to immune checkpoint therapy.
