Project description:To gain further insight into the molecular architecture of cuticulosome containing hair cells, we performed a transcriptomics experiment comparing RNA expression profiles of hair cells with and without cuticulosomes. To do so we adapted existing methods that enable ultra low input RNA sequencing, applying it to avian hair cells. RNA from 30 hair cells per sample was extracted, cDNA prepared using a full length RNA Seq from single cells protocol (Picelli et al. 2014), before next generation sequencing on an Illumina HiSeqV4 system. We generated approximately 42 million reads per sample that were aligned with TopHat against the Columba livia genome and subjected to differential expression analysis with DESeq2 . We report that 387 genes were upregulated (more than 3 fold increase) in cuticulosome positive cells, with 18 reaching significance (P<0.05) following correction for multiple testing.

Project description:A comparative profile of miRNAs in livers during pigeon development was performed by using high-throughput sequencing. We identified known pigeon miRNAs, novel miRNAs, and miRNAs that are conserved in other birds and mammals.Our results expanded the repertoire of pigeon miRNAs and may be of help in better understanding the mechanism of squab’s rapid development from the perspective of liver development.
