Site-dependent lineage preference of adipose stem cells

Project description:Adult stem cells have unique properties in both proliferation and differentiation preference. However, the molecular mechanisms remain unclear. In this study, we hypothesized that the matrix microenvironment plays a critical role in the fate determination of local stem cells. Four rabbits were used to provide donor-matched adipose stem cells from either subcutaneous adipose tissue (ScAT) or infrapatellar fat pad (IPFP). Proliferation and multi-lineage differentiation were evaluated in adipose stem cells from donor-matched ScAT and IPFP. RNA sequencing (RNA-seq) and proteomics were conducted to uncover potential molecular mechanisms underlying the interaction between adipose stem cells and the local matrix microenvironment. We found that stem cells from ScAT exhibited significantly higher proliferation and adipogenic capacity compared to those from donor-matched IPFP while stem cells from IPFP displayed significantly higher chondrogenic potential compared to those from donor-matched ScAT. Our findings are strongly endorsed by supportive data from transcriptome and proteomics analyses, indicating that a unique extracellular matrix microenvironment is critical in the determination of local stem cell fate.

SARS-CoV-2 infects adipose tissue in a fat depot- and viral lineage-dependent manner

Project description:Visceral adiposity is a risk factor for severe COVID-19, and a link between adipose tissue infection and disease progression has been proposed. Here we demonstrate that SARS-CoV-2 infects human adipose tissue and undergoes productive infection in human primary adipose-derived stromal-vascular cells differentiated into adipocytes. However, the permissiveness to infection and the cellular response depends on the anatomical origin of the cells and the viral lineage. Cells of visceral adipose tissue origin express more ACE2 and are more permissive to SARS-CoV-2 infection than their subcutaneous counterpart. SARS-CoV-2 infection leads to inhibition of lipolysis in cells of subcutaneous origin, while in visceral fat cells, it results in higher expression of pro-inflammatory cytokines. Viral load and cellular response are attenuated when visceral adipose tissue cells are infected with the SARS-CoV-2 gamma variant. A similar degree of cell death occurs 4-days after SARS-CoV-2 infection, regardless of the cell origin or viral lineage. Hence, SARS-CoV-2 infects human adipose tissue cells, replicating and altering cell function and viability in a fat depot- and viral lineage-dependent fashion.

Individual variation in preference behavior in sailfin fish refines the neurotranscriptomic pathway for mate preference

Project description:Social interactions can drive distinct gene expression profiles which may vary by social context. Here we use female sailfin molly fish (Poecilia latipinna) to identify genomic profiles associated with preference behavior in distinct social contexts: male-interactions (mate choice) versus female-interactions (shoaling partner preference). We measured behavior of 15 females interacting in a non-contact environment with either two males or two females for 30 minutes followed by whole brain transcriptomic profiling by RNA sequencing. We profiled females that exhibited high levels of social affiliation and great variation in preference behavior to identify an order of magnitude more differentially expressed genes associated with behavioral variation than by differences in social context. Using linear modeling (limma), we took advantage of the individual variation in preference behavior to identify unique gene sets that exhibited distinct correlational patterns of expression with preference behavior in each social context. By combining limma and weighted gene co-expression network analyses (WGCNA) approaches we identify a refined set of 401 genes robustly associated with mate preference that is independent of shoaling partner preference or general social affiliation. While our refined gene set confirmed neural plasticity pathways involved in moderating female preference behavior, we also identified a significant proportion of discovered that our preference-associated genes were enriched for ‘immune system’ gene ontology categories. We hypothesize that the association between mate preference and transcriptomic immune function is driven by the less well-known role of these genes in neural plasticity which is likely involved in higher-order learning and processing during mate choice decisions.
