miR204-containing exosome ameliorates GVHD-associated dry eye disease

Project description:The graft-versus-host disease (GVHD) associated dry eye disease usually leads to refractory pain and visual impairment with limited treatments currently. Here we found exosome derived from mesenchymal stromal cell (MSC-exo) administered as eye drops significantly alleviates GVHD-associated dry eye disease in human and mouse models. To find out the essential elements during exosome treatment, we performed miRNA sequencing of exosomes derived from MSCs and L929 cells, and identified miR-204 in MSC-exo benefited the recovery of dry eye, which targeted IL-6/IL-6R/Stat3 signaling. Blockade of miR-204 abolished the therapeutic effect of MSC-exo while miR-204 overexpression from L929-exo markedly attenuates dry eye. Thus MSC-exo eye drops are efficacious in treating GVHD-associated dry eye and highlight miR-204 as a potential therapeutic agent.

A gene expression study of the developing human eye

Project description:There is a vast range of tools with which to study the cells and tissues of the human body, however the scarcity of human embryonic material as a resource for direct study means that there is still limited data on the specific expression patterns which exist during human development. While embryonic studies are common place in other mammalian organisms and these have contributed greatly to our knowledge, characterising the events which occur during human development specifically is crucial in order to extrapolate the differences which exist between humans and other organisms and understand the human situation more completely. Establishing and maintaining collections of human developmental tissue for direct study requires significant time and resources, and is not a viable option for all. For this reason the creation of an atlas defining the key events which occur during human eye development would be of great value to the field. Through the Human Developmental Biology Resource we have collected human developmental eye tissue from post conception week 4 to 19 and performed gene expression studies using RNA sequencing. This study begins to reveal the spatiotemporal gene expression patterns which occur during normal human eye ontogenesis and represents a benchmark for comparison with development, disease and cellular differentiation studies.

Proteomic profiling of various rabbit eye tissues

Project description:Rabbits have been widely used for studying ocular physiology and pathology due to their relatively large eye size and similar structures with human eyes. Various rabbit ocular disease models, such as dry eye, age-related macular degeneration, and glaucoma, have been established. Despite the growing application of proteomics in vision research using rabbit ocular models, there is no spectral assay library for rabbit eye proteome publicly available. Here, we generated spectral assay libraries for rabbit eye compartments, including conjunctiva, cornea, iris, retina, sclera, vitreous humor, and tears using fractionated samples and ion mobility separation enabling deep proteome coverage. The rabbit eye spectral assay library includes 9,830 protein groups and 113,593 peptides. We present the data as a freely available community resource for proteomic studies in the vision field.
