Project description:This experiment was designed to monitor the gene expression changes of young bees raised with and without queen mandibular pheromone (QMP). This experiment was a timecourse, comparing QMP- to QMP+ bees over 4 days of exposure. Bees that had eclosed over 16 hours were collected from a brood frame and placed in cages, 35 bees/cage. 8 hours later, queen mandibular pheromone (QMP, 0.1 queen equivalents) treatment was initiated, and fresh QMP was placed in the cage every day. One hour after QMP treatment had been refreshed, QMP- and QMP+ cages were collected into liquid nitrogen. 8 cages of each were collected on each day of the timecourse (1 day of treatment = Day 1, as well as Day 2, Day 3, and Day4).

Project description:Responses to social cues, such as pheromones, can be modified by genotype, physiology, or environmental context. Honey bee queens produce a pheromone (queen mandibular pheromone; QMP) which regulates many aspects of worker bee behavior and physiology. Forager honey bees are less responsive to QMP than young nurse bees engaged in brood care, suggesting that physiological changes associated with behavioral maturation may modulate response to this pheromone. Since cGMP is a major regulator of behavioral maturation in honey bee workers, we examined its role in modulating worker responses to QMP. Treatment with a cGMP analog, 8-Br-cGMP, resulted in significant reductions in both behavioral and physiological responses to QMP in young caged workers. Treatment significantly reduced attraction to QMP (the retinue response) and inhibited the QMP-mediated increase in vitellogenin levels in the fat bodies of worker bees. Genome-wide analysis of brain gene expression patterns demonstrated that cGMP has a larger effect on expression levels than QMP, and that QMP has specific effects in the presence of cGMP, suggesting that some responses to QMP may be dependent on an individual beesM-^R physiological state. Several functional gene categories were significantly differentially expressed, including genes involved in regulating GTPase activity, phototransduction, immunity, and carboxylic acid transmembrane transporter activity. Overall, our data suggest that cGMP-mediated processes play a large role in modulating responses to queen pheromone in honey bees, at the behavioral, physiological and molecular levels.

Project description:Female honeybees are specified as workers or queens based on diet during early development. Workers are essentially sterile with a reduced number of ovarioles and no spermatheca. In the presence of the queen (queen mandibular pheromone) and her brood, worker ovaries are kept in an inactive quiescent state. If the queen is removed, or lost, worker bees are able to sense this change in their environment and their ovaries undergo complete remodelling producing unfertilised haploid eggs that will produce male (drone bees). In this study we analyse gene expression in queen, worker, and laying worker ovaries using RNA-seq and explore differences in the chromatin landscape (focussing on H3K27me3).

Project description:The Eurasian spruce bark beetle Ips typographus is known for its devasting attack on the host tree Picea abies, a common conifer in Europe. The beetle uses various pheromone components (2-methyl-3-buten-2-ol and cis-verbenol) for mass aggregation to overcome the tree defence compounds such as terpenes. Though this aggregation pheromone biosynthesis and respective precursors via terpenes detoxification mechanism was investigated for a few decades, gene-level understanding behind these biosynthesis pathways are uncertain yet in I. typographus. Though, applying Juvenile hormone (JH III) on the beetles have induced specific pheromone biosynthesis in many bark beetle species, irrespective of their life stage, it is not uniform found in all Ips species. While investigating pheromone biosynthesis among various life stages of I. typographus, we have also reported recently about the JHIII induction of aggregation pheromone biosynthesis from the gut tissue of the beetle. Thus, in this study, we have applied the concept of JHIII induction on I. typographus and analyzed the respective pheromone and possible biosynthesis precursors from via pathway gene families from the gut tissue of the beetle. A comparative approach from transcriptome and proteome study has revealed the mevalonate pathway genes including isoprenyl-di-phosphate synthase (IPDS) gene (Ityp09271) was upregulated over 5-fold change after JHIII induction in I. typographus. The identified IPDS is suspected to directly involve in 2-methyl-3-buten-2-ol, a vital aggregation pheromone of I. typographus. Added to that, a hydrolase gene family was found upregulated over 2-fold change, specifically in the male gut tissue after JHIII treatment. Furthermore, another vital gene family, CytochromeP450 have shown the upregulated (transcript) in the male gut tissue after treatment. Especially Previously reported CyP450 candidates Ityp3140 and Ityp03153 for pheromone compounds cis/trans- verbenol and ipsdienol biosynthesis respectively. Along with CyP450 candidates, the hydrolase gene candidates could possibly involve in braking down the detox compounds such as diglycosylated terpenes and stored wax esters (verbenyl oleate) from the gut possibly provided from the of the beetle body as a reservoir. An added metabolomic analysis has confirmed these compounds abundance was in the gut tissue. Especially, the abundance of the related fatty acid ester (verbenyl oleate) has reduced half in male gut tissue after the treatment. Hence, we have shed light on three possible genes from different families for the respective pheromone and its precursors biosynthesis after JHIII application over I. typographus. This approach would lead us to elucidate the molecular basis of stored pheromone biosynthesis and the derived knowledge from this study would lead to eco-friendly pest management for this aggressive pest. Project description:In honey bees (Apis mellifera), the reproductive queen produces a pheromonal signal that regulates many aspects of worker behavior and physiology and is critical for maintaining colony organization. Queen mandibular pheromone (QMP) inhibits worker reproduction, attracts workers from a short distance (retinue response), inhibits the rearing of new queens, modulates age-related division of labor and globally alters brain gene expression in worker bees. Interestingly, substantial variation in worker retinue responses to QMP has been found between colonies, but the molecular and physiological bases for variation in individual responses to the queen have not been characterized. Here, we demonstrate that individual retinue response is negatively correlated with traits associated with reproductive potential. Workers with low response to QMP have more ovarioles and higher levels of vitellogenin transcripts than workers with a high response to QMP, suggesting that workers with greater reproductive potential may be attempting to escape queen control. Retinue response appears to be associated with a suite of behavioral and physiological traits that may be pleiotropically linked. However, while these phenotypes are all correlated at the organismal level, the underlying brain expression patterns and gene networks associated with each trait are independent, suggesting that these phenotypes are uncoupled at the molecular level in adult bees. These studies provide insights into the ultimate and proximate causes of natural variation in pheromone response in honey bees.
