Androgen signaling in cell fate and cell-cell communication during early prostate development at single-cell resolution

Project description:Androgens/androgen receptor (AR) mediated signaling pathways are essential for prostate development, morphogenesis, and regeneration. Here, using newly generated ArIRES-Cre mice with single-cell RNA sequencing (scRNAseq) and other experimental approaches, we profiled the transcriptomes of more than 9000 cells isolated from murine E17.5 UGS tissues and identified a variety of urogenital mesenchyme (UGM) and epithelia (UGE) cell populations. The expression of AR and other regulators were further assessed in different cell types at single cell resolution to determine their roles and interactions during early prostate development. Psuedotime analyses further explored the differentiation trajectory of UGM and UGE cell populations, and identified a Zeb1-expressing progenitor population. Using mouse genetic tools, the decisive role of mesenchymal AR action in prostate initiation was further demonstrated. These data provide fresh, high resolution, comprehensive insight into AR signaling and its roles in initiating dynamic interactions of with distinct signaling pathways between prostatic mesenchymal-epithelial cells.

Comparative analysis of cell-cell communication at single-cell resolution

Project description:Inference of cell-cell communication from single-cell RNA-sequencing data is a powerful technique to uncover intercellular communication pathways, yet existing methods perform this analysis at the level of the cell type or cluster, discarding single-cell level information. Here we present Scriabin, a flexible and scalable framework for comparative analysis of cell-cell communication at single-cell resolution that is performed without cell aggregation or downsampling. We use multiple published atlas-scale datasets, genetic perturbation screens, and direct experimental validation to show that Scriabin accurately recovers expected cell-cell communication edges and identifies communication networks that can be obscured by agglomerative methods. Additionally, we use spatial transcriptomic data to show that Scriabin can uncover spatial features of interaction from dissociated data alone. Finally, we demonstrate applications to longitudinal datasets to follow communication pathways operating between timepoints. Our approach represents a broadly applicable strategy to reveal the full structure of niche-phenotype relationships in health and disease.

Androgen action in cell fate and communication during prostate development at single-cell resolution.

Project description:Androgens/androgen receptor (AR)-mediated signaling pathways are essential for prostate development, morphogenesis and regeneration. Specifically, stromal AR signaling has been shown to be essential for prostatic initiation. However, the molecular mechanisms underlying AR-initiated mesenchymal-epithelial interactions in prostate development remain unclear. Here, using a newly generated mouse model, we have directly addressed the fate and role of genetically marked AR-expressing cells during embryonic prostate development. Androgen signaling-initiated signaling pathways were identified in mesenchymal niche populations at single-cell transcriptomic resolution. The dynamic cell-signaling networks regulated by stromal AR were additionally characterized in relation to prostatic epithelial bud formation. Pseudotime analyses further revealed the differentiation trajectory and fate of AR-expressing cells in both prostatic mesenchymal and epithelial cell populations. Specifically, the cellular properties of Zeb1-expressing progenitors were assessed. Selective deletion of AR signaling in a subpopulation of mesenchymal rather than epithelial cells dysregulated the expression of the master regulators and significantly impaired prostatic bud formation. These data provide novel, high-resolution evidence demonstrating the important role of mesenchymal androgen signaling in the cellular niche controlling prostate early development by initiating dynamic mesenchyme-epithelia cell interactions.
