Project description:Telomere shortening is universally acquired with aging in humans, but the contribution of telomere shortening to immune aging is not fully understood. Here, we studied T cells derived from short telomere syndrome patients and compared their T cell profile and function to controls and elderly individuals who had normal age-adjusted telomere length. The short telomere syndrome patients were all under age 40 and carried mutant telomerase or telomere genes and had telomere lengths below the 1st age-adjusted percentile. The T cell profile of young short telomere syndrome patients resembled those that were five decades older including a depletion of naive T cell population and accumulation of terminally differentiated effector cells. To test whether short telomeres affect the quality of these cells, we performed a gene expression microarray. The data are summarized here. We found that although numerically expanded in both short telomere and elderly cases, the gene expression microarrays were distinct with short telomere cells predominately upregulating gene expression of DNA damage pathways and elderly T cells upregulating cell extrinsic apoptotic pathways. The goal of the experiment is to highlight important genes and pathways that may drive immune aging in short telomere syndromes and which may also contribute to normal immune aging.

Project description:Studies in rodents have shown obesity and aging impair tissue nicotinamide adenine dinucleotide (NAD+) biosynthesis, which contributes to metabolic dysfunction. The availability of nicotinamide mononucleotide (NMN) is an important rate-limiting factor in mammalian NAD+ biosynthesis. We conducted a 10-week, randomized, placebo-controlled, double-blind trial to evaluate the effect of NMN supplementation on metabolic function in 25 postmenopausal women with prediabetes who were overweight/obese. Insulin-stimulated glucose disposal, assessed by using the hyperinsulinemic-euglycemic-clamp procedure, increased by 25±7% (P<0.01) in the NMN group, which was accompanied by an increase in insulin-stimulated phosphorylation of muscle AKT (P<0.01), whereas neither outcome changed after placebo treatment. Body composition (fat mass, fat-free mass, intra-abdominal fat, intrahepatic triglyceride content) and muscle mitochondrial respiratory capacity did not change after treatment with placebo or NMN. These results demonstrate NMN improves muscle insulin sensitivity in women with prediabetes who are overweight/obese, independent of changes in body composition or mitochondrial function.

Project description:Telomere length heterogeneity in various cell types including stem cells and cancer cells has been recognized. Cell heterogeneity also is found in pluripotent stem cells such as embryonic stem cells (ESCs). The implication and mechanisms underlying the heterogeneity remain to be defined. We have optimized a robust method that can simultaneously measure telomere length coupled with RNA-sequencing analysis (scT&R-seq) in the same human ES cell. Using this method, we show that telomere length varies with pluripotency state. Long telomere hESCs highly express TERF1/TRF1 as well as ZFP42/REX1, PRDM14 and NANOG for naïve pluripotency, in contrast to short telomere hESCs. hESCs express high telomerase activity as expected, and ubiquitously express NOP10 and DKC1, stabilizing components of telomerase complexes, regardless of telomere lengths. Moreover, new candidate genes such as MELK, MSH6 and UBQLN1 cluster with long telomeres and pluripotency network. Notably, short telomere hESCs exhibit higher oxidative phosphorylation primed for lineage differentiation, whereas long telomere hESCs show elevated glycolysis, another key feature for naïve pluripotency. Our data further suggest that telomere length is implicated in metabolism activity and pluripotency state of hESCs. Single cell analysis of telomere and RNA-sequencing can be exploited to further understand the molecular mechanisms of telomere heterogeneity.

Project description:Advanced maternal age, defined as 35 years or older, is associated with a decline in both ovarian reserve and oocyte quality, which leads to the female infertility, pregnancy loss, fetal anomalies, stillbirth, and obstetric complications. At present, the effective approaches to counteract the maternal age-related decay of oocyte quality are still not fully determined. Here, we report that in vivo supplementation of nicotinamide mononucleotide (NMN) efficaciously ameliorates the quality of oocytes from naturally aged mice by recovering nicotinamide adenine dinucleotide (NAD + ) levels in oocytes. NMN supplementation increases the number of antral follicles, ovulated oocytes and matured oocytes from aged mice. Specifically, NMN supplementation maintains the normal spindle/chromosome structure and dynamics of cortical granule component ovastacin to ensure the meiotic competency and fertilization ability of aged oocytes. Moreover, single cell transcriptome analysis shows that the beneficial effect of NMN on the aged oocytes is mediated by the restoration of the mitochondrial function, thereby reducing the accumulated ROS to suppress the occurrence of apoptosis. To sum up, our data reveal that supplementation of NMN is a feasible approach to prevent oocyte quality from advanced maternal age-related deterioration, contributing to improve the reproductive outcome of aged women and the assisted reproductive technology.

Project description:We sorted H2O2–treated HCC Huh7-cells harboring telomeres in the top 20th percentile (long telomeres) and bottom 20th percentile of telomere length (short telomeres) according to fluorescence-activated cell sorting based on the fluorescence intensity associated with the telomere-binding protein, followed by whole-genome mRNA-expression microarray analysis. In detail, a stable Huh7 cell line expressing GFP-tagged telomere-binding protein adrenocortical dysplasia protein homolog (TPP1) was established using 1 mg/mL G418 as a selection antibiotic. The telomere lengths in living cells were measured by assessing the expression of TPP1 by fluorescence-activated cell sorting (FACS) under the assumption that the expression level of the telomere binding protein is proportional to telomere length. The cells were dissociated at 37°C in 0.2 % w/v trypsin-EDTA for 10 min. After gentle pipetting to induce dissociation, the cells were washed twice with cold PBS solution containing 0.6 % w/v ultra-pure bovine serum albumins (A0100-010) and filtered using a cell strainer (BD Bioscience). HCC cells displaying weak or strong GFP expression were isolated from the mock- or H2O2-treated cells using a FACS Aria II (BD Bioscience), respectively. A whole genome mRNA expression microarray (Macrogen, Seoul, Korea) was used to compare the gene expression patterns between HCC cells possessing short or long telomeres.

Project description:Progressive attrition of telomeres triggers DNA damage response (DDR) and limits the regenerative capacity of adult stem cells during mammalian aging. Intriguingly, the telomere integrity is not only determined by telomere length but also by epigenetic status of telomeric/sub-telomeric regions. However, the functional interplay between telomere shortening-induced DDR and epigenetic modification in aging is unknown. Here we show that deletion of Gadd45a improves the maintenance and function of intestinal stem cells (ISCs), and prolongs lifespan of late generation of telomerase deficient mice (G3Terc-/-). Mechanistically, Gadd45a facilitates the generation of a permissive chromatin status for DDR signaling transduction through the demethylating of CpG islands specifically at sub-telomeric regions of short telomeres. Deletion of Gadd45a restores the heterochromatin compaction in the sub-telomeric regions and thereby ameliorating the DDR initiation at short telomeres of G3Terc-/- ISCs. Treatment with a small molecule inhibitor of BER alleviates DDR and improves the maintenance and function of G3Terc-/- ISCs. Taken together, our study discloses a potential therapeutic approach to enhance stem cell function and prolong lifespan by targeting epigenetic modulating molecules.
