Retrons are prokaryotic genetic retroelements encoding a reverse transcriptase that produces multi-copy single-stranded DNA<sup>1</sup> (msDNA). Despite decades of research on the biosynthesis of msDNA<sup>2</sup>, the function and physiological roles of retrons have remained unknown.
