Proteomic analysis of human microglia cells (HMC3) stimulated with anti-inflammatory cytokines using serum-enriched culture conditions

ABSTRACT: The immune functions of microglia in the brain are compromised in the presence of tumors that have the ability to “hijack” their pro-inflammatory nature by sending out signals that promote cancer progression. To analyze the tumor-supportive microglia phenotype, human microglia cells (HMC3) were treated with anti-inflammatory cytokines in serum-enriched cell culture media, and analyzed by LC-MS/MS using an Orbitrap QE instrument.

Frontiers in immunology 20210909

Background
Microglia safeguard the CNS against injuries and pathogens, and in the presence of certain harmful stimuli are capable of inducing a disease-dependent inflammatory response. When exposed to anti-inflammatory cytokines, however, these cells possess the ability to switch from an inflammatory to an immunosuppressive phenotype. Cancer cells exploit this property to evade the immune system, and elicit an anti-inflammatory microenvironment that facilitates tumor attachment and growt ...[more]
