Genotyping-by-sequencing-based identification of Arabidopsis pattern recognition receptor RLP32 recognizing proteobacterial translation initiation factor IF1

ABSTRACT: Pattern-triggered immunity (PTI) is a central component of plant immunity. Activation of PTI relies on the recognition of microbe-derived structures, termed patterns, through plant encoded surface-resident pattern recognition receptors (PRRs). We have identified proteobacterial translation initiation factor 1 (IF1) as an immunogenic pattern that triggers PTI in Arabidopsis thaliana and some related Brassicaceae species.

Project description:In plants, recognition of immunogenic molecular patterns, such as bacterial flagellin (flg22 epitope), leads to an enhanced state of immunity, designated pattern-triggered immunity (PTI). Following cognate ligand perception, pattern recognition receptors initiate sequential phosphorylation events to activate defense responses against invading pathogens. To gain further insight into PTI signaling, we conducted phosphoproteome analyses in Arabidopsis seedlings with immunogenic molecular patterns.

Function of Arabidopsis PUB2 and PUB4 in plant immunity

Project description:Plants recognize and respond to pathogens relying on pattern recognition receptors (PRRs) which usually are receptor like kinases (RLKs) or receptor like proteins (RLPs). Upon binding to conserved structures or molecules, the so-called pathogen-associated molecular patterns (PAMPs), in pathogens, PRRs form complex with coreceptors and activate pattern triggered immunity (PTI). Here, we show that two U-box type E3 ligases (PUBs), PUB2 and PUB4, play important roles in PTI responses and disease resistance in Arabidopsis.

Expression analysis of Arabidopsis transgenic plants carrying DEX-inducible MKK4DD in Col or sid2 background treated with DEX or control

Project description:Effector-Triggered Immunity (ETI) and Pattern-Triggered Immunity (PTI) are well-defined modes of plant immunity triggered by recognition of pathogen effector proteins and microbe-associated molecular patterns, respectively. While ETI and PTI network extensively share signaling components, the shared components are used in different ways, resulting in distinct network properties in the model plant Arabidopsis: immunity is highly robust against network perturbations in ETI but relatively sensitive in PTI. However, the molecular mechanism how the shared network leads to the different properties is not known. Here we show that sustained MAPK activation compensate salicylic acid (SA) signaling. Interactome of Arabidopsis BIR3 protein

Project description:Arabidopsis BAK1/SERK3, a co-receptor of leucine-rich repeat pattern recognition receptors (PRR), mediates pattern-triggered immunity (PTI). Genetic inactivation of BAK1 or BAK1-interacting receptors (BIR) causes cell death. We foundthat the TIR-NBS-LRR protein CONSTITUTIVE SHADE-AVOIDANCE 1 (CSA1) physically interacts with BIR3, but not with BAK1 and mediates ETI-type cell death in the absence of BAK1 and BIR3.

An immune-inducible trihelix transcription factor suppresses bacterial resistance in Arabidopsis thaliana

Project description:The plant innate immunity consists of the two interconnected mechanisms, pattern-triggered immunity (PTI) and effector-triggered immunity (ETI). Although much is known about how plants trigger immune responses upon pathogen recognition, the genetic program by which plants avoid an overshoot in pathogen-triggered immune responses remains largely unknown. Here, we discovered a trihelix transcription factor, GT-3a, as an immune-inducible negative regulator of bacterial resistance in Arabidopsis thaliana. Analysis of public RNA-seq data revealed that GT-3a is specifically induced by ETI activation not by PTI activation. Overexpression of GT-3a suppressed resistance against the bacterial pathogen Pseudomonas syringae pv. tomato DC3000 (Pto) and Pto-elicited expression of salicylic acid (SA)-responsive genes. Our results suggest that transcriptional induction of GT-3a circumvents the overshooting of SA-mediated defense responses during ETI.

Differential phosphoproteomics upon chitin- or blue-light treatment in Marchantia polymorpha

Project description:Pattern-recognition receptor (PRR)-triggered immunity (PTI) plays a pivotal role in plant immunity to ward off a wide range of pathogenic microbes. The model liverwort Marchantia polymorpha is gaining popularity in investigating the evolution of plant-microbe interactions. The M. polymorpha is capable of triggering defense-related gene expression by sensing components in bacterial and fungal extracts, suggesting existence of PTI in this plant model. However, the molecular components that would form PTI in M. polymorpha have not yet been described. We show that, in M. polymorpha, lysin motif (LysM) receptor-like kinase (LYK) MpLYK1 and LYK-related (LYR) MpLYR, among four LysM receptor homologs, are required for sensing chitin and peptidoglycan (PGN) fragments and thereby triggering a series of immune responses. Phosphoproteomic analysis of M. polymorpha in response to chitin treatment comprehensively identified regulatory proteins that would shape LysM-mediated PTI. The identified proteins covered homologs of well-described PTI components in angiosperms as well as proteins whose roles in PTI are not yet determined including the blue-light receptor phototropin MpPHOT. We revealed that MpPHOT is required for a negative feedback of defense-related gene expression during PTI. Taken together, this study provides the basic framework of LysM-mediated PTI in M. polymorpha and demonstrates the utility of M. polymorpha as a plant model for discovering novel or fundamental molecular mechanisms underlying PRR-triggered immune signaling in plants.
