Liver proteome of red deer (Cervus elaphus) infected by liver fluke Fascioloides magna

ABSTRACT: The aim of the current study was to analyse differences in liver proteomes between F. magna infected and control, apparently healthy, red deer using a label-based high- throughput quantitative proteomics approach.

Project description:The objective of this study was to evaluate the impact of dietary Spirulina and lysozyme supplementation over the muscle proteome of piglets during the post-weaning stage. Thirty piglets were randomly distributed among three diets: control (no microalga), SP (10% Spirulina) and SP+L (10% Spirulina + 0.01% lysozyme). They were fed ad libitum for 4 weeks, after which they were sacrificed and samples of the longissimus lumborum muscle were taken. The muscle proteome was analysed using a Tandem Mass Tag (TMT)-based quantitative approach.

Systems toxicology assessment of CHTP1.2 [Liver Data]

Project description:Modified risk tobacco products (MRTPs) have the potential to reduce smoking-related health risks. The Carbon Heated Tobacco Product 1.2 (CHTP1.2) is a potential MRTP that uses a pressed carbon heat source to generate an aerosol by heating tobacco. This study reports the results from the systems toxicology arm of a 90-day rat inhalation study (OECD test guideline 413) to assess the effects of CHTP1.2 aerosol compared with cigarette smoke (CS). Rats were exposed to filtered air (sham), to CHTP1.2 aerosol (at 15, 23 and 50 µg nicotine / L), or to the 3R4F reference cigarette smoke (at 23 µg nicotine / L).

Rifampicin-mouse liver proteomics

Project description:Tuberculosis is one of top causes of death among curable infectious diseases; it is an airborne infectious disease that kills 2 million people worldwide. Anti-tuberculosis drug-induced liver injury is the primary cause of drug-induced liver injury (DILI). Rifampicin is one of the most common anti-tuberculosis therapies and has well-known hepatotoxicity. To understand the mechanism of rifampicin-induced liver injury, we performed a global proteomic analysis of liver proteins by LC-MS/MS in a mouse model after the oral administration of 177 and 442.5 mg/kg rifampicin (LD10 and LD25) for 14 days. Based on the biochemical parameters in the plasma after rifampicin treatment, the hepatotoxic effect of rifampicin in the mouse liver was defined as a mixed liver injury. In the present study, we identified 1,101 proteins and quantified 1,038 proteins. A total of 29 and 40 proteins were up-regulated and 27 and 118 proteins were down-regulated in response to 177 and 442.5 mg/kg rifampicin, respectively.

"Effects of cigarette smoke, cessation and switching to a modified risk tobacco product on the liver in Apoe-/- mice -- a systems toxicology analysis"

Project description:"Liver represents one of the most important organs involved in the elimination of xenobiotic and potentially toxic substances. Cigarette smoke (CS) contains more than 7000 chemicals, among which compounds that exert biological effects and cause smoking-related diseases. Although CS is not directly hepatotoxic, a growing body of evidence suggests that it may exacerbate (pre-existing) chronic liver diseases. Here we integrated toxicological endpoints with molecular measurements and computational analysis approaches to investigate exposure effects on liver in an Apoe-/- mouse study. The mice were exposed to high concentrations of 3R4F reference CS (600 mg/m3 TPM, 29.9 mg/m3 nicotine), an aerosol from a candidate reduced risk product (RRP) the Tobacco Heating System (THS) 2.2 at matching nicotine concentration, or filtered air (Sham) for up to 8 months. THS2.2 was conceived using the heat-not-burn technology that heats tobacco avoiding pyrogenesis and pyrosynthesis. After 2 months of CS exposure, some groups were either switched to the RRP or underwent exposure cessation. While clear signs of hepatotoxic effects were absent for any exposure group, the integrative analysis of proteomics and transcriptomics data showed a CS-dependent impairment of specific biological networks, including lipid and xenobiotic metabolism, and iron homeostasis, which likely in turn mutually contribute to worsening of the oxidative stress. In contrast, most of these changes were absent in mice exposed to THS2.2, and in the cessation and switching groups. Our findings shed light on the complex biological response of the liver to CS exposure and support the benefits of switching to the tested heat-not-burn product, THS2.2."

HNF4A is required to specify glucocorticoid action in the liver - ATACseq

Project description:The glucocorticoid receptor (GR) is a nuclear hormone receptor critical to the regulation of energy metabolism and the inflammatory response. The actions of GR are highly dependent on cell type and environmental context. Here, we demonstrate the necessity for liver lineage-determining factor hepatocyte nuclear factor 4A (HNF4A) in defining liver-specificity of GR action. In normal mouse liver, the HNF4 motif lies adjacent to the glucocorticoid response element (GRE) at GR binding sites found within regions of open chromatin. In the absence of HNF4A, the liver GR cistrome is remodelled, with both loss and gain of GR recruitment evident. Loss of chromatin accessibility at HNF4A-marked sites leads to loss of GR binding at weak GRE motifs. GR binding is gained at sites characterised by strong GRE motifs, which typically show GR recruitment in non-liver tissues. The functional importance of these HNF4A-regulated GR sites is further demonstrated by evidence of an altered transcriptional response to glucocorticoid treatment in the Hnf4a-null liver.

Project description:The microsomes are re-formed vesicle-like artifacts, not ordinarily present in living cells, from pieces of the endoplasmic reticulum (ER), plasma membrane, mitochondria, and Golgi apparatus of broken eukaryotic cells. Typically, microsomes are separated by differential centrifuged from homogenized of cell or tissue. Based on enrichment of ER fractions, prepared microsomes mainly show a large of metabolic activity following high amount of metabolic enzyme, as cytochrome P450 (CYP). CYPs are important components of xenobiotic-metabolizing monooxygenase accounting for an approximate 75% of biotransformation of clinically relevant drugs. Hepatic CYP are ER-anchored hemoproteins involved in the metabolism of numerous endo- and xenobiotics including drugs, steroids, and carcinogens. The CYPs catalytic activity is subject to regulation via phosphorylation. In several previous studies, 8 CYP phosphorylation sites had been discovered in human. Protein kinase A (PKA) and protein kinase C (PKC) were identified as a major catalyst for the phosphorylation of CYPs in 2003. The phosphorylation of CYPs process to their ubiquitination undergoing ubiquitin-dependent 26S proteosomal degradation.

Project description:Cigarette smoke (CS) causes adverse health effects that may occur shortly after smoking initiation and lead to the development of respiratory disease (chronic obstructive pulmonary disease), cardiovascular disease, and cancer. To reduce the risk of smokers developing smoking-related diseases, Philip Morris International is developing modified risk tobacco products (MRTP). Within a systems toxicology study, we integrated multi-omics measurements with classical endpoints to assess the effects in female ApoE-/- mice of six months of exposure to CS (at 29.9 µg nicotine/L) or to aerosols from one potential and one candidate MRTP (CHTP 1.2 and THS 2.2, respectively) at matched nicotine concentrations. The impact of cessation or switching to CHTP 1.2 after three months of CS exposure was also evaluated. This data set contains the results from the analysis of proteome changes in the liver using the iTRAQ® quantification approach.
