Identification of AF9-associated proteins by IP and mass spectrometry


ABSTRACT: To assess whether these proteins directly interact with each other in cells, we performed large-scale co-immunoprecipitation (co-IP) experiments using nuclear extract of HEL cells and the AF9 antibody against the N-terminus of AF9, and analyzed immunoprecipitated proteins by mass spectrometry. We identified the common subunits of the COMPASS-like complexes.

Mixed lineage leukemia-fusion proteins (MLL-FPs) are believed to maintain gene activation and induce MLL through aberrantly stimulating transcriptional elongation, but the underlying mechanisms are incompletely understood.
