Human liver cell line LO2 LC-MS/MS

ABSTRACT: A hyperlipidaemia model of L02 cells was established using free fatty acids (FFA, oleate and palmitate, 2:1; Sigma-Aldrich, USA) at a final concentration of 1 mM for 24 h. Cells were allowed to attach overnight prior to HTG-medicated serum for 24 h and then exposed to FFA during drug treatment. After treatment, LO2 cells were harvest and stored at -80 ℃, lactylation proteomics was performed by Jingjie PTM BioLabs (Hangzhou, China).

Project description:3×Flag-atrA strain was grown in YEME and 50 mL were collected after 24 h. Cells were resuspended in phosphate buffer (pH=7.4) (PBS) and sonicated. After centrifugation at 12000 rpm, the supernatant was transferred and incubated with FLAG affinity gel (Yeasen) at 4 °C. The FLAG affinity gel was washed three times with 1 mL binding buffer (PBS contains 10% glycerol). After centrifugation at 2000 rpm, 500 μL PBS was added, and the interaction proteins were eluted after denaturation. WT strain was used as control. Protein identification and analysis was provided by Jingjie PTM BioLab (Hangzhou, China). Proteins were expressed in BL21 with the plasmids pET-28a-3×Flag-atrA and pET28a-clpX, respectively. The purified proteins were mixed and incubated at 30 °C for 3 h. The reaction system consisted of 4 μg AtrA, 4 μg ClpX and 3 mM ATP. The elution method is the same as for the pull-down assay after FLAG affinity gel purification at 4 °C. Samples were analyzed by western blot using anti-His mouse monoclonal antibody. AtrA was expressed in BL21 with the plasmid pET-32a-atrA and incubated with cell lysate of Δprc at 30 °C for 1 h. The protein processing and pupylation analysis were provided by Jingjie PTM BioLab (Hangzhou, China).

Pre-existing cross-reactive antibody responses do not significantly impact inactivated COVID-19 vaccine-induced neutralization

Project description:Recent exposure to seasonal coronaviruses (sCoVs) may stimulate cross-reactive antibody responses against SARS-CoV-2. Previous studies have shown divergent results regarding protective or damaging immunity induced by prior exposure to sCoVs. It is still unknown whether pre-existing humoral immunity may play a role in the vaccine-induced neutralization and antibody responses. In this study, we collected 36 paired sera in healthy volunteers before and after immunization with inactivated SARS-CoV-2 vaccines, and analyzed the distribution and intensity of pre-existing antibody responses at the epitope level before vaccine immunization, as well as the relationship between pre-existing sCoVs immunity and vaccine-induced neutralization.

Skin transcriptome profiles associated with skin color in chickens

Project description:This work was to study the transcriptome profiles in the skin of chickens with black versus white skin using high-throughput RNA deep-sequencing technology, to investigate the different expression profiles of the genes involved in skin pigmentation, then look for the main differences between black and white skin colors in Lueyang chickens. 16-week-old white and black female Lueyang chickens (5 birds per color) were selected for the sample collection. A piece of skin (8 mm in diameter) from the left back was collected . Total RNA was extracted from the sample using Trizol reagent . Three RNA samples from either the black or white skin samples were pooled following mRNA isolation. The sequencing of the library was performed using an Illumina HiSeq 2000 (LianChuan Sciences, Hangzhou, China). According the result of sequencing, some colored gene expressions were validated using Real time quantitative polymerase chain reaction (qPCR).

Identification of novel KRT32-associated proteins in primary keratinocytes cells lysate by GST-KRT32 pulldown assays coupled with mass spectrometry.

Project description:GST-KRT32 and its interacting proteins in primary keratinocytes cells were enriched by pull-down using glutathione-Sepharose beads. Then, KRT32-interacting proteins were identified by mass spectrum. Proteins obtained in Pulldown were subjected to SDS-PAGE gel electrophoresis and silver staining. The SDS-PAGE gel was sent to the mass spectrometry platform of the Jingjie PTM Biolab for subsequent processing and mass spectrometry detection.

A secretome profile indicative of oleate-induced proliferation of HepG2 hepatocellular carcinoma cell line

Project description:Increased fatty acid (FA) is often observed in highly proliferative tumors, contributing to promoting proliferation of tumor cells. FA affects secreted factors from tumor cells, which can modulate tumor microenvironment in favor of tumor survival. However, the secreted factors affected by the increased FA have not been systematically explored. Here, we performed comprehensive secretome profiling of oleate-treated and untreated HepG2 cells. Comparison of the secretomes identified 349 differentially secreted proteins (DSPs; 145 up-regulated and 192 down-regulated) in oleate-treated samples, compared to untreated samples. Functional enrichment and network analyses of the DSPs revealed that the 145 up-regulated secreted proteins by oleate treatment were mainly associated with cell proliferation-related processes, such as lipid metabolism, inflammatory response, and ER stress. Based on the network models of the DSPs, we selected four up-regulated secreted proteins (MIF, THBS1, PDIA3, and APOA1) that can represent such processes related to cell proliferation. Thus, our results provided a secretome profile indicative of oleate-induced proliferation of HepG2 cells.
