Proteomic Insights into Psychrophile Growth in Perchlorate-Amended Subzero Conditions: Implications for Martian Life Detection

ABSTRACT: Forty-five Colwellia psychrerythraea 34H (Cp34H) samples were individually prepared for mass spectrometry proteomics to identify proteomic effects of perchlorate oxy-ions and putative perchlorate-tolerant biosignatures. Cultures of Cp34H were grown in half-strength 2216 medium at -1°C to late log phase (to an optical density (OD) at 600 nm of 0.2-0.3, three proteomic samples are from this initial culture. 100 µl of this culture was used to inoculate 1 ml of 1) half-strength 2216 medium (control) and 2) half-strength 2216 medium amended with perchlorate ions (WCL medium), at a 30:1 ratio (2216:WCL) and incubated at -1℃ and -5℃ for 15 days. Twenty-one proteomic samples are control samples, grown in half strength 2216 medium, and the other twenty-one proteomic samples were grown in half strength 2216 medium with perchlorate ions (WCL) at a 30:1 ratio (2216:WCL). Biological triplicate samples from certain time points (7,9, and 11 at -1℃; 5,9,11,13 at -5℃) were selective for proteomic analysis.
