HeLa Pro-IL-1a TurboID LC-MS/MS


ABSTRACT: Pro-IL-1a is a pro-inflammatory cytokine that is localised to the nucleus in humans due to the presence of a nuclear localisation sequence in its pro-domain. However, some mammals have a predicted loss of fucntion to their nuclear localistion sequence. It is suggested that pro-IL-1a serves as a dual action protein, and that in addition to its role as a cytokine, it has an additional nuclear role. Therefore, we used biotin proximity labelling to explore the protein interactome of pro-IL-1a with the aim to investogate possible additional roles of this protein. We have suggested in our study that interaction with histone acetyl trasnferases may suggest a possible nuclear function of pro-IL-1a.

Interleukin-1α is a suggested dual-function cytokine that diverged from interleukin-1β in mammals potentially by acquiring additional biological roles that relate to highly conserved regions in the pro-domain of interleukin-1α, including a nuclear localisation sequence and histone acetyltransferase-binding domains.
