Identification of novel ovarian predominant miRNAs in medaka

Project description:The involvement of miRNAs during vertebrate oogenesis is poorly documented. Based on the assumption that ovarian-specific or ovarian-predominant genes usually play important roles during oogenesis, we searched for ovarian-predominant miRNAs in the medaka (Oryzias latipes) ovary. 10 tissus were collected from adult medaka (intestine, ovary, testis, liver, heart, gills, kidney, brain, muscle and bone) and RNAs were hybridized on a designed agilent microarray displaying 3800 distinct miRNAs from different teleost and vertebrate species. We identified 66 miRNAs sequences predominantly expressed in the ovary that had never been previously described in medaka.

Whole Genome Bisulfite Sequencing (WGBS) or zebrafish, medaka, and zebrafish-medaka hybrid embryos

Project description:During development, the inherited DNA methylation patterns from the parental gametes needs to be remodeled into a state compatible with embryonic pluripotency. In Zebrafish, this remodeling is achieved by the maternal methylome becoming hypomethylated to match the paternal methylome. However, how this is achieved in medaka (another teleost fish) is currently not known. Moreover, how DNA methylation remodeling is impacted in hybrid organisms, and the effects this may have on their development, is also not known. Here we address these questions by generation whole genome bisulfite sequencing data for zebrafish, medaka and zebrafish medaka embryos.

Changes in ovarian gene expression profiles in maturing European eel (Anguilla anguilla)

Project description:The goal of this study was to gain a better understanding of the genetic background of gonadal maturation of the European eel and to use gene expression profiles to identify predictive markers for broodstock selection that can be measured in blood samples. To find leads for maturation markers we performed a pilot deep-sequencing transcriptome analysis of ovarian tissue derived from a yellow eel, a prepubertal silver eel and a post-spawning matured eel. Among the best leads were two key players in steroidogenesis, namely pP450c17 and liver receptor homolog-1.

Transcription profiling of major osmoregulatory tissues of Anguilla anguilla in response to the sea water environment

Project description:The aim of this study was to use a microarray approach to screen the major osmoregulatory tissues of the eel for changes in gene expression as fish acclimate to the SW environment. The other species (few clones) were used to validate the specificity of arrays hybridizations : Of the 6144 clones spotted, 96 contained control cDNAs comprising either known eel genes implicated in osmoregulation and previously studied in our laboratory or coding regions of genes sequenced from other species (ie. Aspergillus nidulans, Arabidopsis thaliana, Pleuronectes flesus, Carcharhinus leucas, Scyliorhinus canicula, Squalus acanthias and Salmo salar). The labelled cDNA probes derived from the eel RNAs hybridised only with the spotted cDNAs from the eel clones indicating the stringency and probe specificities of the hybridisation conditions were appropriate.

Transcriptome sequencing analysis of swamp eel

Project description:The swamp eel or rice field eel (Monopterus albus) taxonomically belongs to the family Synbranchidae of the order Synbranchiformes (Neoteleostei, Teleostei, Vertebrata). It is not only an economically important freshwater fish in aquacultural production, but also an increasingly known model species for biological studies. Understanding molecular mechanisms underlying sex change is a major area of interest. The swamp eel thus offers a powerful system for studying sexual development and adaptive evolution in vertebrates.The whole genome sequencing provides valuable resources for sex control in fish production, species protection through manipulating sex reversal genes, and potentially enabling effective population control and promoting reproduction health in human. High throughput sequencing was employed for three samples,three kind s of sex gonad from swamp eel, testis,ovotestis and ovary, no replicates.
