Smell of Infection: a novel, non-invasive method for detection of fish excretory- secretory proteins

Project description:Chemical signals are produced by aquatic organisms following predatory attacks or perturbations such as parasitic infection. Ectoparasites feeding on host fish are likely to cause release of similar alarm cues into the environment due to the stress, wounding and immune response stimulated upon infection. Alarm cues are often released in the form of proteins and peptides and provide important insights into bodily function and infection status. Here we outline a non-invasive method to identify potential chemical cues associated with infection in fish by extracting, purifying and characterizing proteins from water samples from cultured fish. Gel free proteomic methods were deemed the most suitable for protein detection in saline water samples. It was confirmed that proteins could be characterized from teleost water samples and that variation in protein profiles could be detected between infected and uninfected individuals and fish and parasite only water samples. Our novel assay provides a non-invasive method for assessing the health condition of both wild and farmed aquatic organisms. Similar to environmental DNA monitoring methods, these proteomic techniques could provide an important tool in applied biology and aquaculture biology

Project description:A custom multi-species microarray was used to study gene expression in wild hornyhead turbot (Pleuronichthys verticalis), collected from polluted and clean coastal waters in Southern California and in laboratory male zebrafish (Danio rerio) following exposure to estradiol and 4-nonylphenol. A multi-gene cross species microarray was fabricated as a diagnostic tool to screen the effects of environmental chemicals in fish, for which there is minimal genomic information. The microarray measurement of gene expression in zebrafish, which are phylogenetically distant from turbot, indicates that this multi-species microarray will be useful for measuring endocrine responses in Pleuronectiformes and other fish for which there is minimal genomic sequence information.

Project description:The potential of an hepatic transcriptome expression profile, evaluated in a sentinel feral fish, to serve as an environmental biomarker was examined. Gene expression profiles of individuals of Lithognathus mormyrus were examined using a cDNA microarray and were correlated to the set of environmental exposure conditions at their site and date of collection. The clusters of individual gene expression profiles were reasonably related to the fish samples and several sample-specific clusters of genes, designated gene signatures of the sample, were determined. The relationship between transcriptome expression and the fish samples indicated a potential for their utilization as an environmental biomarker. Ten fish were sampled at each of two sites (Dor, Israel and Haifa, Israel) each of two years (2007 and 2008) for a total of 40 fish. All fish were hybridized to a common reference RNA population using a dye-swap design. Hence, each fish was hybridized twice vs. the reference RNA. The reference RNA was an equalized pool of the RNAs from all 10 fish comprising sample D7.

A comparison of gene transcription profiles of domesticated and wild Atlantic salmon (Salmo salar L.) at early life stages, reared under controlled conditions

Project description:Atlantic salmon (Salmo salar) has been selectively bred in Europe since the 1970s and the process of domestication has led to both phenotypic and genotypic differences between wild and farmed fish. Despite strict regulations large numbers of fish escape annually from fish farms, a concern for both aquaculturalists and those managing wild fish stocks. A better understanding of the interactions between domesticated and wild salmon is essential to the continued sustainability of the aquaculture industry and to the maintenance of healthy wild stocks. One major concern is that of potential interbreeding of escapees with wild fish leading to potentially detrimental genetic changes in wild populations. Advances in high throughput technologies allow the role of genome-wide gene transcription to be studied in relation to both micro- and macro- evolutionary change. In this study, we have compared the transcriptomes of Norwegian wild and domesticated stocks at two life stages: yolk sac and first-feeding salmon fry and reared under identical conditions. These preliminary data improve knowledge of potential transcriptional difference between domesticated and wild salmon and will hopefully provide a better understanding of the fitness consequences of such interactions.

Epigenetic Inheritance of DNA Methylation Changes in Fish Living in Hydrogen Sulfide-rich Springs

Project description:Although hydrogen sulfide is toxic to most organisms, a fish, Poecilia mexicana, has adapted to survive in environments with high levels of hydrogen sulfide. The epigenetic changes in response to this environmental stress were examined by assessing DNA methylation alterations in the nucleated red blood cells (RBC) in the fish. In addition to collecting wild males and females from sulfidic and non-sulfidic environments, wild males and females in these environments were collected and moved to a non-sulfidic environment in the laboratory and propagated for two generations in a non-sulfidic environment. We compared epimutations between sexes and field and laboratory populations. The F0 generation sulfidic wild fish were compared to the non-sulfidic wild fish and found to have significant differential DNA methylation regions (DMRs) in the RBC DNA. The F2 generation laboratory fish were also compared between the sulfidic and non-sulfidic populations, and a significant number of DMRs were also identified. The DMRs have stable generational inheritance in the absence of the sulfidic environment. The DMRs in the F0 generation wild fish had an over 80% overlap with the F2 generation laboratory non-sulfidic environment propagated fish. This is one of the first examples of epigenetic generational stability after the removal of an environmental stressor. The DMR associated genes were found to be relevant to sulfur toxicity and metabolism processes.
