Project description:Apis mellifera syriaca is the native honeybee subspecies of Jordan and much of the Middle East. It expresses behavioral adaptations to a regional climate with very high temperatures, nectar dearth in summer, attacks of the Oriental wasp Vespa orientalis and in most cases it is resistant to varroa mites. The Thorax control sample of A. m. syriaca in this experiment was originally collected and stored since 2001 from Wadi Ben Hammad a remote valley in the southern region of Jordan. Using morphometric and Mitochondrial DNA markers it was proved that bees from this area had show higher similarity than other samples collected from the Middle East as represented by reference samples collected in 1952 by Brother Adam. The samples L1-L5 are collected from the National Center for Agricultural Research and Extension breading apiary which was originally established for the conservation of Apis mellifera syriaca. Goal was to use the genetic information in the breeding for varroa resistant bees and to determine the successfulness of this conservation program. Project description:Bees make honey from the nectar that they collect from flowers. The characteristics of honey are closely associated to original botanical species. Compare with sugars in honey, proteins are minor components but usually used as an important honey quality evaluation parameters. Flower-origin proteins could be a good marker for the authentication. However, as a minute component in honey proteome, plant origin proteins are hard to be detected in honey by regular proteomic approaches, such as gel-based techniques. In this study, Eriobotrya japonica Lindl. (loquat) nectar and its derivative monofloral honey were systematically compared, especially regarding the proteomes and enzymatic activities. Using two-dimensional electrophoresis and mass spectrometry, only bee-originated proteins were detected in loquat honey which were major royal jelly proteins and two uncharacterized proteins. Xylosidase, thaumatin, and two kinds of chitinases were detected in loquat floral nectar by the gel-based proteomic approach. To our knowledge, it is the first study to analysis nectar-originated enzymes’ activity in honey and we proposed that the zymography of chitinase is a potential marker for honey botanical origin authentication.

Project description:The ability of honey bees to evaluate differences in food type and value is crucial for colony success, but these assessments are made by individuals who bring food to the hive, eating little, if any, of it themselves. We tested the hypothesis that responses to food type (pollen or nectar) and value involve different subsets of brain regions, and genes responsive to food. mRNA in situ hybridization of c‐jun revealed that brain regions responsive to differences in food type were mostly different from regions responsive to differences in food value, except those dorsal and lateral to the mushroom body calyces, which responded to all three. Transcriptomic profiles of the mushroom bodies generated by RNA sequencing gave the following results: (1) responses to differences in food type or value included a subset of molecular pathways involved in the response to food reward; (2) genes responsive to food reward, food type and food value were enriched for (the Gene Ontology categories) mitochondrial and endoplasmic reticulum activity; (3) genes responsive to only food and food type were enriched for regulation of transcription and translation; and (4) genes responsive to only food and food value were enriched for regulation of neuronal signaling. These results reveal how activities necessary for colony survival are channeled through the reward system of individual honey bees.

Project description:Many angiosperms can secret at least two types of sugar-rich liquids, floral nectar (FN) and extrafloral nectar (EFN), by which plants can make use of the animal partner’s mobility for pollen transportation and attract predatory animals for indirect defense. Both FN and EFN contain considerable amount of proteins which play important roles in nectar biosynthesis process and protection. Hemerocallis citrina (HC) can secrete both FN and EFN on flower during the same developmental stage. Our objective was to compare the HC FN and EFN proteome to understand the difference between their biosynthesis and ecological function. FN was collected from adult HC flowers and concentrated by ultrafiltering. EFN was collected from young HC flower buds and concentrated by ultrafiltering. Proteins were digested with trypsin then analyzed by LC-MS/MS. HSPs are the main protein identified in HC FN but their function in floral nectar is still largely unknown. PR proteins are the main protein identified in HC EFN with antimicrobial activity. Our data provide a good characterization of a monocot nectar proteome. These data, may be useful in understanding the generation process and ecological function of floral and extrafloral nectar.
