Project description:In this study we used a maize multiparental advanced generation intercross (MAGIC) population, originating from nine parental lines (A632, B73, B96, F7, H99, HP301, Mo17, W153R and CML91) followed by 6 generations of self-pollination. A subset of 94 lines was chosen randomly from the set of 529 lines that was genotyped and phenotyped in the field (Dell'Aqua et al (2015) Genome Biology, 16:167) and sampled for RNA seq of proliferative tissue of growing leaf.

Transcriptome analysis of 103 RILs of a B73 x H99 RIL population

Project description:The RIL population used in this study is derived from a cross between parental lines B73 and H99, followed by 12 generations of self-pollination. In total 142 RILs were generated and 223 markers were used for mapping these RILs (Marino et al. (2009) Mol Genet Genomics, 281:163-179). The transcriptome of a randomly chosen subset of 103 RILs was analyzed in this study using RNA-seq.

Project description:The aim of the study was to identify candidate genes responsible for drought tolerance trait between a pair of wheat varieties ( WL711 and C306) and correspondng progeny bulks (10 drought susceptible RILs and 10 drought tolerant RILs) by combining QTLs analysis with expression analysis. Microarray analysis of RNA extracted from the flag leaves showed large number of differentially expressed genes. The number of differentially expressed genes was reduced to 37 on the basis of their occurance in a major QTL region (responcible for drought tolerance) detected in RIL population derived from WL711 and C306.

Project description:Whole genome transcriptome profiling of bulked RILs with high and low grain number per panicle derived from 2 cultivars at panicle primordia stage The aim of this study was to identify candidate genes responsible for grain number per panicle by combining QTLs analysis with expression analysis. Microarray analysis of RNA extracted from the panicle primordia showed 20 differentially expressed genes, respectively. The differentially expressed genes were shorted to 4 on the basis of their occurance in the QTL region (responcible for grain number regulation) detected in RIL population derived from Pusa 1266 and Pusa Basmati 1.

Gene expression profiling in control, heat-shock, and recovery treatment in a RIL population of Caenorhabditis elegans L4 larvae

Project description:This experiment investigates the genetic architecture of gene expression (eQTL) in three different treatments in a N2xCB4856 RIL population of Caenorhabditis elegans. The goal is to identify genetic variation linked to differences in gene expression. We exposed 48 RILs per treatment to a control, heat-stress, and recovery treatment. More specifically these three conditions can be characterized as: (i) the control treatment was grown for 48 hours at 20C, (ii) the heat-stress treatment was grown for 46 hours at 20C followed by 2 hours at 35C, and (iii) the recovery treatment was grown for 46 hours at 20C, followed by 2 hours at 35C and thereafter 2 hours at 20C. Thereafter RNA was isolated, labelled and hybridized on microarray. The gene expression profiles were used for eQTL mapping.
