Project description:It is assumed that climate and land-use changes cause increasing stress to pollinators, which play pivotal roles in almost all terrestrial ecosystems, with consequences on population growth, diversity and ecosystem functions. While these responses to global change drivers are well located, the molecular pathways triggering the response are poorly understood. We analysed the transcriptomic response of Bombus lucorum workers in their systematic responses to temperature and livestock grazing, sampled along an elevational gradient from 650 – 1930 m.a.s.l., and from differently managed grassland sites (livestock grazing vs. unmanaged) in and around the National Park Berchtesgaden (German Alps).

Project description:The altitude gradient limits the growth and distribution of alpine plants.Alpine plants have developed strategies to survive the extremely cold conditions prevailing at high altitudes; however, the mechanism underlying the evolution of these strategies remains unknown. The alpine plant Potentilla saundersiana is widespread in the Northwestern Tibetan Plateau. In this study, we conducted a comparative proteomics analysis to investigate the dynamic patterns of protein expression of P. saundersiana located at five different altitudes. We detected and functionally characterized 118 differentially expressed proteins. Our study confirmed that increasing levels of antioxidant proteins, and their respective activities, and accumulation of primary metabolites, such as proline and sugar, confer tolerance to the alpine environment in P. saundersiana. Proteins species associated with the epigenetic regulation of DNA stability and post-translational protein degradation were also involved in this process. Furthermore, our results showed that P. saundersiana modulated the root architecture and leaf phenotype to enhance adaptation to alpine environmental stress through mechanisms that involved hormone synthesis and signal transduction, particularly the cross-talk between auxin and strictosidine. Based on these findings, we conclude that P. saundersiana uses multiple strategies to adapt to the high-altitude environment of the Northwestern Tibetan Plateau.

Project description:The elevational diversity pattern for microorganisms has received great attention recently but is still understudied, and phylogenetic relatedness is rarely studied for microbial elevational distributions. Using a bar-coded pyrosequencing technique, we examined the biodiversity patterns for soil bacterial communities of tundra ecosystem along 2000-2500 m elevations on Changbai Mountain in China. Bacterial taxonomic richness displayed a linear decreasing trend with increasing elevation. Phylogenetic diversity and mean nearest taxon distance (MNTD) exhibited a unimodal pattern with elevation. Bacterial communities were more phylogenetically clustered than expected by chance at all elevations based on the standardized effect size of MNTD metric. The bacterial communities differed dramatically among elevations, and the community composition was significantly correlated with soil total carbon (TC), total nitrogen, C:N ratio, and dissolved organic carbon. Multiple ordinary least squares regression analysis showed that the observed biodiversity patterns strongly correlated with soil TC and C:N ratio. Taken together, this is the first time that a significant bacterial diversity pattern has been observed across a small-scale elevational gradient. Our results indicated that soil carbon and nitrogen contents were the critical environmental factors affecting bacterial elevational distribution in Changbai Mountain tundra. This suggested that ecological niche-based environmental filtering processes related to soil carbon and nitrogen contents could play a dominant role in structuring bacterial communities along the elevational gradient.

Project description:Tibet is one of the most threatened regions by climate warming, thus understanding how its microbial communities function may be of high importance for predicting microbial responses to climate changes. Here, we report a study to profile soil microbial structural genes, which infers functional roles of microbial communities, along four sites/elevations of a Tibetan mountainous grassland, aiming to explore potential microbial responses to climate changes via a strategy of space-for-time substitution. Using a microarray-based metagenomics tool named GeoChip 4.0, we showed that microbial communities were distinct for most but not all of the sites. Substantial variations were apparent in stress, N and C cycling genes, but they were in line with the functional roles of these genes. Cold shock genes were more abundant at higher elevations. Also, gdh converting ammonium into urea was more abundant at higher elevations while ureC converting urea into ammonium was less abundant, which was consistent with soil ammonium contents. Significant correlations were observed between N-cycling genes (ureC, gdh and amoA) and nitrous oxide flux, suggesting that they contributed to community metabolism. Lastly, we found by CCA, Mantel tests and the similarity tests that soil pH, temperature, NH4+M-bM-^@M-^SN and vegetation diversity accounted for the majority (81.4%) of microbial community variations, suggesting that these four attributes were major factors affecting soil microbial communities. Based on these observations, we predict that climate changes in the Tibetan grasslands are very likely to change soil microbial community functional structure, with particular impacts on microbial N cycling genes and consequently microbe-mediated soil N dynamics. Project description:Most studies of biodiversity-elevational patterns do not take species abundance into consideration. Hill numbers are a unified family of indices that use species abundance and allow a complete characterization of species assemblages through diversity profiles. Studies on dung beetle responses to elevation were essentially based on species richness and produced inconsistent results because of the non-distinction between different habitats and the use of gradients dispersed over wide areas. We analyzed dung beetle diversity in a Mediterranean mountain (central Italy) for different habitats (woodlands vs. grasslands) and taxonomic groups (scarabaeids and aphodiids). Scarabaeids were the most abundant. Since scarabaeids are able to construct subterranean nests, this indicates that the warm and dry summer climatic conditions of high elevations favor species capable of protecting their larvae from desiccation. Dung beetles were more abundant and diversified in grasslands than in woodlands, which is consistent with their preference for open habitats. In the woodlands, diversity increased with increasing elevation because of increasing tree thinning, whereas, in the grasslands, diversity decreased with elevation because of increasingly harsher environmental conditions. These results indicate a trade-off in the beetle response to elevation between the positive effects of increasing the availability of more suitable habitats and the decrease of optimal environmental conditions.

Project description:The extreme environments of the Tibetan Plateau offer significant challenges to human survival, demanding novel adaptations. While the role of biological and agricultural adaptations in enabling early human colonization of the plateau has been widely discussed, the contribution of pastoralism is less well understood, especially the dairy pastoralism that has historically been central to Tibetan diets. Here, we analyze preserved proteins from the dental calculus of 40 ancient individuals to report the earliest direct evidence of dairy consumption on the Tibetan Plateau. Our palaeoproteomic results demonstrate that dairy pastoralism began on the higher plateau by approximately 3,500 years ago, more than 2,000 years earlier than the recording of dairying in historical sources. With less than 1% of the Tibetan Plateau dedicated to farmland, pastoralism and the milking of ruminants were essential for large-scale human expansion into agriculturally-marginal regions that make up the majority of the plateau. Dairy pastoralism allowed conversion of abundant grasslands into nutritional human food, which facilitating adaptation in the face of extreme climatic and altitudinal pressures, and maximizing the land area available for long-term human occupation of the “roof of the world”.

Project description:Highlighting patterns of distribution and assembly of plants involves the use of community phylogenetic analyses and complementary traditional taxonomic metrics. However, these patterns are often unknown or in dispute, particularly along elevational gradients, with studies finding different patterns based on elevation. We investigated how patterns of tree diversity and structure change along an elevation gradient using taxonomic and phylogenetic diversity metrics. We sampled 595 individuals (36 families; 53 genera; 88 species) across 15 plots along an elevational gradient (2440-3330 m) in Ecuador. Seventy species were sequenced for the rbcL and matK gene regions to generate a phylogeny. Species richness, Shannon-Weaver diversity, Simpson's Dominance, Simpson's Evenness, phylogenetic diversity (PD), mean pairwise distance (MPD), and mean nearest taxon distance (MNTD) were evaluated for each plot. Values were correlated with elevation and standardized effect sizes (SES) of MPD and MNTD were generated, including and excluding tree fern species, for comparisons across elevation. Taxonomic and phylogenetic metrics found that species diversity decreases with elevation. We also found that overall the community has a non-random phylogenetic structure, dependent on the presence of tree ferns, with stronger phylogenetic clustering at high elevations. Combined, this evidence supports the ideas that tree ferns have converged with angiosperms to occupy the same habitat and that an increased filtering of clades has led to more closely related angiosperm species at higher elevations.
