Project description:Gan mice express Wnt1, Ptgs2, and Ptges, which develop inflammation-associated gastric tumors (Oshima et al, Gastroenterology 131: 1086, 2006). We examined the role of TNF-alpha in tumorigenesis by construction of TNF-/- Gan mice. We also examined genetic background difference in tumor phenotype by changing Gan mouse background from C57BL/6(B6) to BALB/c. Microarray analyses were performed to examine changes of expression profiles in tumors by TNF gene disruption or by changing genetic background. Total RNA was prepared from B6 Gan mice (n=3: Gan1-Gan3), B6-Gan TNF-/- mice (n=3: Gan(TNF KO)1-Gan(TNF KO)3), BALB-Gan mice (n=3: Gan(BALB/c)1-Gan(BALB/c)3), and wid-type normal glandular stomach (n=3: WT1-WT3). We used Affymetrix microarrays for hybridization, and examined expression profiles.

Gene expression data from colitis-associated colonic tumors of WT vs. MDR1A KO mice

Project description:Human Ulcerative colitis (UC) is characterized by chronic colonic inflammation and has been associated with an increased risk of colorectal carcinoma. Gene and protein expression profiles of ABCB1/MDR1 have been shown to be dysregulated in UC and sporadic colorectal cancer. We demonstrated that in a murine model of colitis-associated tumorigenesis, MDR1A KO mice showed reduced tumor load when compared to wildtype (WT) mice. The aim of this study was to identify gene alterations in colitis-associated tumors in the context of MDR1A deficiency. We used microarrays to assess gene expression profiles of colitis-associated colonic tumors from WT or MDR1A KO mice.

RNAseq of murine MB49 tumors - WT and gal-9-KO - grown on C57BL/6 syngenic mice through serial transplantations

Project description:A number of studies have reported evidence of positive or negative contributions of galectin-9 (gal-9) to human and experimental malignancies. Some clinical observations and in vitro experiments suggest that cell-associated gal-9 has anti-metastatic effects. On the other hand, extra-cellular gal-9 consistently enhances tumor immune escape. So far, all animal studies on this subject have been focused on gal-9 released by infiltrating cells, without paying attention to gal-9 released by malignant cells. To address this issue, we derived by gene editing, isogenic clones - either positive or negative for gal-9 - from the MB49 murine bladder carcinoma cell line. A progressive reduction of tumor growth was observed when gal-9-KO cells were subjected to serial transplantations into syngenic mice but not into nude mice thus accounting the tumor growth reduction in syngenic mice to a better immune response. Tumor fragments were collected from WT and KO tumors at different steps of the experiment : 2nd growth cycle (WT = 5 samples ; KO = 6 samples ) ; 3rd growth cycle (WT = 5 samples ; KO = 7 samples) ; 4th growth cycle (WT = 4 samples ; KO = 3 samples), in order to study the differences between WT and KO tumors through the serial transplantations, by RNAseq analysis.
