Project description:The need to understand the biodegradation of oil and chemical dispersants in Arctic marine environments is increasing alongside growth in oil exploration and transport in the region. We chemically quantified biodegradation and abiotic losses of crude oil and Corexit 9500, when present separately, in incubations of Arctic seawater and identified microorganisms potentially involved in biodegradation of these substrates based on shifts in bacterial community structure (16S rRNA genes) and abundance of biodegradation genes (GeoChip 5.0 microarray). Incubations were performed over 28-day time courses using surface seawater collected from near-shore and offshore locations in the Chukchi Sea. Within 28 days, the indigenous microbial community biodegraded 36% (k = 0.010 day-1) and 41% (k = 0.014 day-1) of oil and biodegraded 77% and 33% (k = 0.015 day-1) of the Corexit 9500 component dioctyl sodium sulfosuccinate (DOSS) in respective near-shore and offshore incubations. Non-ionic surfactants (Span 80, Tween 80, and Tween 85) present in Corexit 9500 were non-detectable by 28 days due to a combination of abiotic losses and biodegradation. Microorganisms utilized oil and Corexit 9500 as growth substrates during the incubation, with the Corexit 9500 stimulating more extensive growth than oil within 28 days. Taxa known to include oil-degrading bacteria (e.g., Oleispira, Polaribacter, and Colwellia) and some oil biodegradation genes (e.g., alkB, nagG, and pchCF) increased in relative abundance in response to both oil and Corexit 9500. These results increase our understanding of oil and dispersant biodegradation in the Arctic and suggest that some bacteria may be capable of biodegrading both oil and Corexit 9500.

Project description:C5aR1, a receptor for the complement activation proinflammatory fragment, C5a, is primarily expressed on cells of the myeloid lineage, and to a lesser extent on endothelial cells and neurons in brain. Previous work demonstrated C5aR1 antagonist, PMX205, decreased amyloid pathology and suppressed cognitive deficits in Alzheimer Disease (AD) mouse models. In the Arctic AD mouse model, genetic deletion of C5aR1 prevented behavior deficits at 10 months. However, the molecular mechanisms of this protection has not been definitively demonstrated. To understand the role of microglial C5aR1 in the Arctic AD mouse model, we have taken advantage of the CX3CR1GFP and CCR2RFP reporter mice to distinguish microglia as GFP-positive and infiltrating monocytes as GFP and RFP positive, for subsequent transcriptome analysis on specifically sorted myeloid populations from wild type and AD mouse models. Immunohistochemical analysis of mice aged to 2, 5, 7 and 10 months showed no change in amyloid beta (Ab) deposition in the Arctic C5aR1 knockout (KO) mice relative to that seen in the Arctic mice. Of importance, no CCR2+ monocytes/macrophages were found near the plaques in the Arctic brain with or without C5aR1. RNA-seq analysis on microglia from these mice identified inflammation related genes as differentially expressed, with increased expression in the Arctic mice relative to wildtype and decreased expression in the Arctic/C5aR1KO relative to Arctic. In addition, phagosomal-lysosomal proteins and protein degradation pathways that were increased in the Arctic mice were further increased in the Arctic/C5aR1KO mice. These data are consistent with a microglial polarization state with restricted induction of inflammatory genes and enhancement of clearance pathways.

Project description:Deep-sea sedimentary hydrocarbonoclastic bacteria are still not widely used in the bioremediation field, especially for crude oil spill biodegradation. This study utilized a mixed culture of Raoultella sp., Enterobacter sp., and Pseudomonas sp. isolated from deep-sea sediment to determine the abilities of bacteria to degrade petroleum hydrocarbons while incorporating environmental variations in a microcosm study. The oil biodegradation extent was determined by measuring the remaining oil and grease in the sample vials. The highest percentage of biodegradation was 88.6%, with a constant degradation rate of 0.399 day-1. GC-MS analysis showed that the most degradable compound in the oil samples was paraffin. This study also observed that microbial degradation was optimized within three days of exposure and that degradation ability decreased at 35 °C. The salinity variation effects were insignificant. Based on all analyses, deep-sea sediment bacteria have great potential in oil spill biodegradation in a microcosm scale.
