Project description:The cultivated almond exhibits self-incompatibility of the gametophytic type regulated by the S-locus, and expressed in pistil (S-RNase) and in pollen (SFB protein). The aim of this study is to clarify the transcription pattern of these 2 S-genes and to identify additional components of the gametophytic self-incompatibility system in almond. With this aim, A2-198 (self compatible) and ITAP-1 (self incompatible) almond selections were used: RNA-seq of pistils of these two accessions both un-pollinated and pollinated with A2-198 pollen were carried out.

Characterization of protein in the pistil after the compatible and incompatible pollination using iTRAQ quantitative proteomics

Project description:Analyze the different proteins in the pistil after the compatible and incompatible pollination of peony by iTRAQ. The number of identified proteins was 2900, 383 proteins of which were up-regulated expression, and 302 proteins were down-regulated. Those proteins mainly attended the metabolic pathways, biosynthesis of secondary metabolites, ribosome, protein processing in endoplasmic reticulum, spliceosome, RNA transport, glycolysis/gluconeogenesis etc., showed that the proteins and energy metabolism were active. Among them, the RNA degradation, calcium signaling pathway, MAPK signaling pathway, and phosphatidylinositol signaling system were involved in the cross-incompatibility between herbaceous peony and tree peony. Those showed that the energy synthesis of the growing pollen tubes and the polaized growth of pollen tubes were inhibited. Among Enolase, DnaK(HSP70), CALM were down regulating expression, and ANT was up regulating expression.

Involvement of different plant viruses in the activation of RNA silencing-related genes and the defensive response against Plum pox virus of ‘GF305’ peach (Prunus persica) grafted with ‘Garrigues’ almond (P. dulcis)

Project description:Plum pox virus (PPV) causes the serious sharka disease in Prunus trees. Peach [P. persica (L.) Batsch] trees are severely affected by PPV and no definitive source of genetic resistance has been identified at this moment. Previous results showed, however, that PPV-resistant ‘Garrigues’ almond [P. dulcis (Mill.) D.A. Webb] was able to transfer its resistance to ‘GF305’ peach through grafting, preventing these trees from PPV infection and reducing symptomatology and viral load in PPV-infected plants. A recent study tried to identify genes responsible for this effect by studying mRNA expression through RNAseq data in peach and almond plants, before and after grafting, and before and after PPV infection. In this work, we used the same peach and almond samples, but focused the high-throughput analyses on small RNAs (sRNAs) expression. We studied massive sequencing data and found an interesting pattern of sRNAs overexpression linked to antiviral defense genes that suggested activation of these genes followed by downregulation to basal levels. We also discovered that ‘Garrigues’ almond plants were infected by different plant viruses that were transferred to peach plants. The large amounts of viral sRNAs found in grafted peaches indicated a strong RNA silencing antiviral response and led us to postulate that these plant viruses could be collaborating by cross-protection in the observed ‘Garrigues’ effect.

Prunus dulcis response to novel defense elicitor peptides and control of Xylella fastidiosa infections

Project description:Xylella fastidiosa is a plant pathogenic bacterium that has been introduced in the European Union (EU), threatening the agricultural economy of relevant Mediterranean crops such as almond (Prunus dulcis). Plant defense elicitor peptides would be promising to manage diseases such as almond leaf scorch but their effect on the host has not been fully studied. In this work, the response of almond plants to the defense elicitor peptide flg22-NH2 was studied in-depth using RNA-seq, confirming the activation of the salicylic acid and abscisic acid pathways. Marker genes related to the response triggered by flg22-NH2 were used to study the effect of the application strategy of the peptide on almond plants and to depict its time course. The application of flg22-NH2 by endotherapy triggered the highest number of upregulated genes, especially at 6 hours after the treatment. A library of peptides that include BP100-flg15, HpaG23, FV7, RIJK2, PIP-1, Pep13, BP16-Pep13, flg15-BP100 and BP16 triggered a stronger defense response in almond plants than flg22-NH2. The best candidate, FV7, when applied by endotherapy on almond plants inoculated with X. fastidiosa, significantly reduced levels of the pathogen and decreased disease symptoms. Therefore, these novel plant defense elicitors are suitable candidates to manage diseases caused by X. fastidiosa, in particular almond leaf scorch.

Comparative Metabolomic Profiling of Compatible and Incompatible Interactions Between Potato and Phytophthora infestans

Project description:Late blight is one of the main biological stresses limiting the potato yield; however, the biochemical mechanisms underlying the infection process of Phytophthora infestans remain unrevealed. In this study, the late blight-resistant potato cultivar Ziyun No.1 (R) and the susceptible cultivar Favorita (S) were inoculated with P. infestans. Untargeted metabolomics was used to study the changes of metabolites in the compatible and incompatible interactions of the two cultivars and the pathogen at 0, 48 and 96 h postinoculation (hpi). A total of 819 metabolites were identified, and the metabolic differences mainly emerged after 48 hpi. There were 198 and 115 differentially expressed metabolites (DEMs) in the compatible and incompatible interactions. These included 147 and 100 upregulated metabolites during the compatible and incompatible interactions, respectively. Among them, 73 metabolites were identified as the P. infestans-responsive DEMs. Furthermore, the comparisons between the two cultivars identified 57 resistance-related metabolites. Resistant potato cultivar had higher levels of salicylic acid and several upstream phenylpropanoid biosynthesis metabolites, triterpenoids and hydroxycinnamic acids and their derivatives, such as sakuranetin, ferulic acid, ganoderic acid Mi, lucidenic acid D2 and caffeoylmalic acid. These metabolites play crucial roles in cell wall thickening and have antibacterial and antifungal activities. This study reports the time-course metabolomic responses of potatoes to P. infestans. The findings reveal the responses involved in the compatible and incompatible interactions of potatoes and P. infestans.

The first transcriptome analysis of sugar beet in compatible and incompatible interactions with Heterodera schachtii

Project description:Sugar beet (Beta vulgaris subsp. vulgaris) is an economically important crop and provides nearly one third of the global sugar production annually. The beet cyst nematode (BCN), Heterodera schachtii, causes major yield losses in sugar beet worldwide. The most effective and economic approach to control this nematode is growing tolerant or resistant cultivars. To identify candidate genes involved in susceptibility and resistance, the transcriptome of sugar beet and BCN in compatible and incompatible interactions at two time points, was studied using mRNA-seq. In total, 16 cDNA libraries were constructed and 442 691 707raw reads were obtained. In the compatible interaction, many alterations in phytohormone-related genes were detected. The effect of exogenous application of methyl jasmonate and ethephon was therefore investigated and the results revealed significant reduction of J2s infection and female development rates in treated susceptible plants. Our results revealed candidate genes putatively involved in the Hs1pro1-induced resistance, such as genes related to phenylpropanoid pathway, putative R genes and genes encoding F-box proteins, zinc finger and NAC transcription factors, ABC transporters, BURP and CYSTM proteins. Also, the transcriptome of BCN in the infected root samples was analyzed and several nematode effector genes were found. Our study is the first investigation of the transcriptome profile in the compatible and incompatible interactions between sugar beet and BCN.

Gene expression analysis in stigma during compatible and in-compatible pollination reactions in Lolium perenne

Project description:The expression analysis had two goals: (1) look at relative transcription within mature pollen grains (2) compare expression in the stigma during pollination with either compatible or in-compatible pollen. Two pairwise comparisons, (i) unpollinated stigma vs. stigma pollinated with compatible pollen, and (ii) unpollinated stigma vs stigma pollinated with incompatible pollen. The genotype where stigma samples were harvested from is F1-30, and this is also the pollen source during an incompatible pollination reaction. The compatible pollen source is the variety Foxtrot (heterogeneous populations).
