Project description:Small aquatic organisms harbour deep phylogeographic patterns and highly structured populations even at local scales. These patterns indicate restricted gene flow, despite these organisms' high dispersal abilities, and have been explained by a combination of (1) strong founder effects due to rapidly growing populations and very large population sizes, and (2) the development of diapausing egg banks and local adaptation, resulting in low effective gene flow, what is known as the Monopolization hypothesis. In this study, we build up on our understanding of the mitochondrial phylogeography of the halophilic rotifer Brachionus plicatilis in the Iberian Peninsula by both increasing the number of sampled ponds in areas where secondary contact is likely and doubling sample sizes. We analyzed partial mitochondrial sequences of 252 individuals. We found two deep mitochondrial DNA lineages differing in both their genetic diversity and the complexity of their phylogeographic structure. Our analyses suggest that several events of secondary contact between clades occurred after their expansion from glacial refugia. We found a pattern of isolation-by-distance, which we interpret as being the result of historical colonization events. We propose the existence of at least one glacial refugium in the SE of the Iberian Peninsula. Our findings challenge predictions of the Monopolization hypothesis, since coexistence (i.e., secondary contact) of divergent lineages in some ponds in the Iberian Peninsula is common. Our results indicate that phylogeographic structures in small organisms can be very complex and that gene flow between diverse lineages after population establishment can indeed occur.

Project description:The effects of climate change on organisms are now being extensively studied in many different taxa. However, the variation in body size, usually shrinkage in response to increasing temperature, has received little attention regarding to reptiles. During past periods of global warming, many organisms shrank in size, and current evidence and experiments manipulating temperature have shown a biomass decrease in some organisms with increasing temperatures. Here we test whether the body size of the Montpellier snake Malpolon monspessulanus from the southeastern Iberian Peninsula is changing and correlated with the increasing temperature in this region during a 39-year period (1976-2014). We measured the snout-vent length (SVL) of vouchers in scientific collections to check for trends in adult body size at the population level in relation with temperature, while controlling for the age of the individuals (estimated by skeletochronology, n?=141). Given the great ontogenetic variation in body size of the study species, we categorized age in 3 classes: "young adults" (under 5 years old), "intermediate adults" (from 5 to 7 years old), and "old adults" (from 8 to 14 years old). By means of linear mixed models, we found a negative relationship between SVL of "old adults" and average annual temperature in the region during the lifetime of each individual. Our results indicate that largest and oldest individuals of the Montpellier Snake, that is, males because of strong sexual size dimorphism in this species, disappeared from the study population, and suggest that it occurred in response to rising environmental temperature.

Project description:The hypothalamus is implicated in controlling feeding and adiposity, besides many other physiological functions, and thus can be of great importance in explaining productive differences between lean and fatty pig breeds. The present study aimed to evaluate the hypothalamic transcriptome of pure Iberian (IBxIB) and Large White x Iberian crossbreds (IBxLW) at 60 days-old, produced in a single maternal environment. Results showed the implication of gender and genotype in the hypothalamic transcriptome, with 51 differentially expressed genes (DEGs) between genotypes and 10 DEGs between genders. Fourteen genotype by sex interactions were found, due to a higher genotype effect on transcriptome found in males. In fact, just 31 DEGs were identified when using only females but 158 using only males. A higher expression of genes related to mitochondrial activity in IBxIB male animals (ND3, ND4, ND5, UQCRC2 and ATP6) was found, which was related to a higher oxidative phosphorylation and greater reactive oxygen species and nitric oxide production. IBxLW male animals showed higher expression of SIRT3 regulator, also related to mitochondrial function. When females were analysed, no such differences were found, since only some differences in genes related to the tricitric acid cycle were observed. Thus, the results indicate a significant effect and interaction of the breed and the sex on the hypothalamic transcriptome at this early age.
