Project description:Dilated cardiomyopathy (DCM) is the leading cause of heart failure and transplantation worldwide. We used iPSCs to model this disease and compared gene expression change before and after gene therapy of cardiomyocytes derived from DCM-specific iPSCs. We used microarrays to detail the global gene expression of patient specific iPSCs, iPSC-derived cardiomyocytes and its response to gene therapy. Skin fibroblasts and iPSCs derived from a family exhibiting familial dilated cardiomyopathy and H7 human ESCs were subjected to RNA extraction and hybridization on Affymetrix microarrays.Global gene expression pattern were compared and analyzed. Cardiomyocytes derived from iPSCs generated from this DCM family were treated with or without adenoriral Serca2a and subjected to RNA extraction and hybridization on Affymetrix microarrays. Global gene expression pattern were compared and analyzed.

Project description:The key aim of this experiment is to characterize the iPSC-Mφ population before and after pulmonary transplantation. For this purpose the following cell populations were compared: (i) transplanted iPSC-Mφ, (ii) Mφ obtained by in vitro differentiation of murine lineage-negative bone marrow cells (BM-Mφ), (iii) non-differentiated CD45.1 iPSC, (iv) murine alveolar Mφ (AMφ) isolated from the BALF of healthy control mice, and (v) iPSC-Mφ recovered from the transplanted animals two months after (PMT-Mφ).

Expression data from the dermis of human established scars before and after hair follicle transplantation

Project description:Human hair follicles undergo repetitive cycles of growth throughout their lifetime. During the hair follicle cycle, functional and structural changes occur within the surrounding skin environment. However, skin that experienced a deep injury lacks cycling hair follicles and turns into a mass of unremodelled scar tissue. We hypothesise that re-introducing cycling hair follicles into human scars will stimulate skin remodelling improving fibrotic tissue. To determine the transcriptional events underlying remodelling of scar dermis after hair follicle transplantation, we used Affymetrix microarrays to perform profiling of scar dermis before (0 mo), and at three time points after hair transplantation transplantation: 2, 4, and 6 months.
