Project description:Global challenges with anthelmintic failure and resistance development lends impetus to the development of new nematocides (anthelmintics) with novel mechanism(s) of action. The free-living nematode Caenorhabditis elegans is as an important model organism used for drug discovery and a powerful tool for anthelmintic screening, evaluation and target deconvolution. Previously, we conducted a whole-organism phenotypic screen of the ‘Pandemic Response Box’ (from Medicines for Malaria Venture, MMV) and identified a hit compound, called ABX464, with activity against C. elegans. Here, we explored this nematocidal pharmacophore on C. elegans, and then tested a series of 46 analogues for human hepatoma (HepG2) toxicity, revealing five compounds whose potency was similar or greater than that of ABX464. Subsequently, we employed thermal proteome profiling (TPP), protein structure prediction and an in silico docking algorithm to identify prime ABX464-target candidates. Taken together, the findings from this study contribute significantly to the early-stage drug discovery of a new nematocide based on ABX464. Future work is aimed at validating the ABX464-protein interactions identified here, and at assessing of ABX464 and associated analogues against a panel of parasitic nematodes, towards the development of a new anthelmintic with a novel mechanism of action.

Project description:Quantitative high throughput screening (qHTS) pharmacologically evaluates libraries of drugs and investigational agents for potential therapeutic uses, toxicological risk assessment, and increasingly for academic chemical tool discovery. Phenotypic HTS assays aim to interrogate molecular pathways and networks, often relying on cell culture systems, historically with less emphasis on multicellular organisms. C. elegans has served as a powerful eukaryotic model organism for human biology and disease by virtue of genetic conservation and experimental tractability, as well as a surrogate for infectious parasitic nematodes. Here we describe a paradigm to enable C. elegans in qHTS using 384-well microtiter plate laser scanning cytometry for rapid signal acquisition with concurrent quantification of the fluorescent protein-encoded phenotype. E. coli ghost capsules are used as a non-replicating nutrient source to allow compound titration exposures over the full 7-day life cycle to mitigate complications from bacterial overgrowth. We demonstrate the method using 643 anti-infective biased agents tested in 7-pt titration to assess feasibility of nematode-based in vivo qHTS. A pharmacological profile from the primary screen confirmed the efficacy of known anti-parasitic molecules, such as ivermectin and levamisole as well as illuminating anthelmintic properties of general chemical classes, including -secretase, bromodomain and proteasome inhibitors. We anticipate a broader application of laser scanning cytometry-based qHTS will enable the analysis of C. elegans orthologous transgenic phenotypes of human pathologies to facilitate drug discovery for a range of therapeutic indications.

Project description:Parasitic nematodes of humans, animals and plants cause significant economic losses worldwide. Control of these pathogens is currently challenging due to the widespread problem of nematode anthelmintic resistance. Unfortunately, most efforts to develop anti-nematode vaccines for use in animals and humans have not succeeded. However, one effective (dead) anti-nematode vaccine (Barbervax) has been developed to protect livestock animals against the socioeconomically important parasitic nematode Haemonchus contortus (barber’s pole worm). This vaccine contains a major native antigen, termed H11. In its native form, H11 alone consistently induces high immunoprotection (75-95%) in animals, but recombinant forms thereof do not. Here, to test the hypothesis that post-translation modification of H11 is responsible for achieving the immunoprotection. We explored the N-glycoproteome and N-glycome of H11 using high-resolution mass spectrometry and assessed the roles of N-glycosylation in protective immunity against H. contortus. We demonstrated that N-glycans are a predominant protective component that induces protection and an associated IgG antibody response in immunised animals. We also showed that anti-H11 IgG antibodies confer specific, passive immunity in naïve animals. This study is the first detailed investigation of the relevance of protein glycosylation in protective immunity against a parasitic nematode, and should have important implications for developing vaccines against metazoan parasites.

Project description:Haemonchus contortus is a highly pathogenic parasitic nematode of that can infect a large number of wild and domesticated ruminant species and is the most economically important parasite of sheep and goats worldwide. Although originally a tropical parasite, it has been disseminated around the world by livestock movement and can now be found as far north as the arctic circle. Adult worms are blood feeders that reside in the abomasum (stomach) and are approximately 2cm in length when mature. They are dioecious with single females typically producing several thousand eggs per day which pass out of the host in faeces and develop to infective larvae on the pasture. H. contortus is a member of the superfamily trichostrongyloidea (Strongylida) which contains most of the economically important parasitic nematodes of grazing livestock. These parasites cost the global livestock industry billions of dollars per annum in lost production and drug costs. Resistance to all the major anthelmintic classes is now common worldwide often leading to failure of treatment and control. H. contortus is a close relative of the human hookworm species and belongs to the nearest phylogenetic group of parasites to the free-living model nematode Caenorhabditis elegans . This makes it an important model of parasitic nematode biology that is commonly used for experimental studies. Project description:Within the context of our anthelmintic discovery program, we recently identified and evaluated a quinoline derivative, called ABX464 or obefazimod, as a nematocidal candidate; synthesised a series of analogues which were assessed for activity against the free-living model nematode Caenorhabditis elegans; and predicted compound-target relationships by thermal proteome profiling (TPP) and in silico docking. Here, we logically extend(ed) this work and critically evaluate(d) the anthelmintic activity of ABX464 analogues on Haemonchus contortus (barber’s pole worm) – a highly pathogenic nematode of ruminant livestock. First, we tested a series of 44 analogues on H. contortus (larvae and adults) to investigate ABX464’s nematocidal pharmacophore, and identified one compound with greater potency than the parent compound, and also showed moderate activity against a select number of other parasitic nematodes (including Ancylostoma, Heligmosomoides and Strongyloides species). Using TPP and in silico modelling studies, we predicted protein HCON_00074590 (a predicted aldo-keto reductase) as a target candidate for ABX464 in H. contortus. Future work aims to optimise this compound as a nematocidal candidate and investigate its pharmacokinetic properties. Overall, this study presents the first steps towards the development of a new anthelmintic with a novel, distinct mechanism of action.

Project description:Infections and diseases caused by parasitic nematodes have a major adverse impact on the health and productivity of animals and humans worldwide. The control of these parasites often relies heavily on the treatment with commercially available chemical compounds (anthelmintics). However, the excessive or uncontrolled use of these compounds in livestock animals has led to major challenges linked to drug resistance in nematodes. Therefore, there is a need to develop new anthelmintics with novel mechanism(s) of action. Recently, we identified a small molecule, KM08948, with nematocidal activity against the free-living model organism Caenorhabditis elegans. Here, we evaluated KM08948’s potential as an anthelmintic in a structure-activity relationship (SAR) study in C. elegans and in the highly pathogenic, blood-feeding Haemonchus contortus (barber’s pole worm), and explored the compound-target relationship using thermal proteome profiling (TPP). First, we synthesised and tested a series of 25 KM08948 analogues for nematocidal activity in both H. contortus (larvae and adults) and C. elegans (young adults), establishing a preliminary nematocidal pharmacophore for both species. We identified several compounds with marked activity against either H. contortus or C. elegans which had greater efficacy than KM08948, and found a significant divergence in compound bioactivity between these two nematode species. We also identified a KM08948 analogue, 25, that moderately inhibited the motility of adult female H. contortus in vitro. Subsequently, we inferred three H. contortus proteins (HCON_00134350, HCON_00021470 and HCON_00099760) and five C. elegans proteins (F30A10.9, F15B9.8, B0361.6, DNC-4 and UNC-11) that interacted directly with KM08948; however, no conserved protein target was shared between the two nematode species. Future work aims to extend the SAR investigation in these and other parasitic nematode species, and validate individual proteins identified here as targets of KM08948. Overall, the present study describes an evaluation of this anthelmintic candidate and highlights some challenges associated with early anthelmintic discovery.

Subsequent life-stage and proteomics analysis helped elucidate the physiologic consequence of specific compounds on C. elegans viability. For example, bortezomib induced a parallel upregulation of core proteasome subunits and the transcription factor skn-1-activating DDI aspartyl protease, while the kinase inhibitor trametinib upregulated the C. elegans MEK-1 ortholog, Mnk-1, downstream aminoacyl tRNA synthetases and ribosomal survival pathway proteins. We anticipate a broader application of this qHTS-coupled proteomics approach will enable the analysis of C. elegans orthologous transgenic phenotypes of human pathologies to facilitate drug and probe profiling from high-impact chemical libraries for a range of therapeutic indications.

Project description:Expulsion of parasitic gastrointestinal nematodes requires diverse effector mechanisms coordinated by a Th2-type response. The evolutionarily conserved JmjC protein ribosomal oxygenase 2 (Riox2, formerly Mina) has been shown to repress IL4, a key Th2 cytokine, suggesting Riox2 may negatively regulate parasite expulsion. Here we report that expulsion of parasitic nematodes was indeed accelerated in Riox2 deficient mice. Unexpectedly, this was associated not with an elevated Th2 but rather an impaired Th1-type response. Further, in vitro T cell differentiation, reciprocal bone marrow chimera and conditional KO experiments demonstrated that retarded parasite expulsion and a normal Th1-type response both required Riox2 in intestinal epithelial cells (IECs). Gene profiling experiments in IECs revealed anti-microbial α-defensin peptides to be the major target of Riox2-dependent repression. In vitro exposure to recombinant α-defensin peptides caused cytotoxic damage to whipworms. These results identify a latent IEC-intrinsic anthelmintic pathway actively constrained by Riox2 and point to α-defensins as important effectors that together with Riox2 would be attractive therapeutic targets for the control of nematode infection.

Project description:Angiostrongylus costaricensis is a relatively uncharacterized nematode that causes abdominal angiostrongyliasis in Latin America, a human parasitic disease. Currently, no effective pharmacological treatment for angiostrongyliasis exists. Peptidases are known to be druggable targets for a variety of diseases and are essential for several biological processes in parasites. Therefore, this study aimed to systematically characterize the peptidase activity of A. costaricensis in different developmental stages of this parasitic nematode.
