Project description:Fusarium graminearum (teleomorph Gibberella zeae) is a prominent pathogen that infects major cereal crops, such as wheat, barley, and maize. Conidiogenesis had been intensively studied in Aspergillus nidulans and regulatory pathway genes have been known to regulate conidiogenesis in stage specific manner. We reported the functional analyses of flbD, abaA, and wetA orthologs in F. graminearum. To understand genome-wide transcriptional profiling of conidiation, we employed RNA-seq of the wild-type Fusarium graminearum Z-3639 and each gene deletion mutants with three time courses (0 h, 6 h and 12 h after induction of conidiogenesis). Project description:Bacterial pathogen Burkholderia glumae and fungal pathogen Fusarium graminearum cause similar disease symptoms and often co-isolated from rice heads, inferring interactions between the two pathogens. F. graminearum is resistant to the bacterial toxin toxoflavin, a strong anti-microbial activity, produced by B. glumae. We isolated toxoflavin-sensitive mutants from transcription factor deletion mutant library of F. graminearum. To understand genome-wide transcriptional profiling, we performed RNA-seq analyses of F. graminearum wild-type strain GZ03639 and toxoflavin-sensitive mutant strains (∆GzZC190, ∆GzC2H008, ∆GzbZIP005) under toxoflavin condition.

Project description:We report a complete transcriptomic study of Fusarium graminearum in response to glucose, cellulose, xylan and cell wall fragments with a whole genome microarray from febit. Fusarium graminearum was cultured at 25 °C on minimal M3 medium with glucose, birch wood xylan, carboxy methyl cellulose or hop cell wall as sole carbon source at a concentration of 10 g/L. Microarray experiments were achieved with a Geniom device (febit biomed, Germany). Each of the four tested condition was tested with two biological and two technical replicates.

Genome-wide expression profiling reveals transcriptional reprogramming in Fusarium graminearum by FgV1-DK21 virus infection

Project description:Using a 3′-tiling microarray covering the whole F. graminearum genes, we carried out genome-wide expression analyses of F. graminearum infected with FgV-DK21 (Mycovirus) at two different time stages (36 h and 120 h). Our study is the first report which elucidated the global alternation of the prominent cellular pathways in fungal host by FgV1-DK21 virus infection. A total of 12 chips were used for microarray. At 36 h and 120 h, total RNAs were extracted from virus free Fusarium and virus infected Fusarium with three biological replicates.

Project description:Using a 3'-tiling microarray covering the whole F. graminearum genes, we carried out genome-wide expression analyses of both a wild type and a FgVelB deletion strainn of F. graminearum at a sexual stage (3 days after perithecial induction on carrot agar) Our study is the first report elucidating regulatory pathways controlled by FgVelB in Fusarium graminearum. A total of 4 chips were used for microarray. Total RNAs were extracted from perithecia and/or ground mycelia formed on carrot agar cultures of a self-fertile wild-type and a self-sterile FgVelB-deleted strain of F. graminearum with two biological replicates.
