Project description:Harmful algal blooms (HABs) arise from the rapid growth of algae in both marine and freshwater ecosystems due to the continuous global temperature rise and anthropogenic eutrophication. Humans are at a great risk of exposure to toxins released from HABs through drinking water, food, and recreational activities, making HAB toxins contaminants of emerging concern (CECs). The impact of HAB toxins on women’s reproductive health, however, remains poorly understood. Here, we investigated the effects of microcystin-LR (MC-LR), the most common HAB toxin, on the ovary, the female gonad, and associated ovarian functions. The results of a chronic daily oral mouse exposure model and an acute exposure using the mouse superovulation model revealed that MC-LR accumulated in the ovary and environmentally-relevant exposure to MC-LR perturbed follicle-stimulating hormone (FSH)-dependent follicle maturation to disrupt ovulation and luteinization. Using an ex vivo mouse follicle maturation and ovulation assay and in vitro culture of human primary granulosa cells, mechanistic studies such as the single-follicle RNA sequencing analysis and others elucidated that MC-LR inhibited protein phosphatase 1 (PP1) and interfered with PP1-mediated PI3K/AKT/FOXO1 signaling in granulosa cells, which suppressed follicle maturation, ovulation, luteinization, and progesterone secretion. Together, our study demonstrates that environmentally-relevant exposure to MC-LR acts a PP1 inhibitor to interfere with the PI3K/AKT/FOXO1 signaling in granulosa cells, which disrupts follicle maturation and results in adverse female reproductive outcomes. As a newly identified ovarian endocrine disrupting chemical (EDC), exposure to MC-LR poses a serious threat to women’s reproductive health and fertility.

Project description:Due to the occurrence of more frequent and widespread toxic cyanobacteria events, the ability to predict freshwater cyanobacteria harmful algal blooms (cyanoHAB) is of critical importance for the management of drinking and recreational waters. Lake system specific geographic variation of cyanoHABs has been reported, but regional and state level variation is infrequently examined. A spatio-temporal modeling approach can be applied, via the computationally efficient Integrated Nested Laplace Approximation (INLA), to high-risk cyanoHAB exceedance rates to explore spatio-temporal variations across statewide geographic scales. We explore the potential for using satellite-derived data and environmental determinants to develop a short-term forecasting tool for cyanobacteria presence at varying space-time domains for the state of Florida. Weekly cyanobacteria abundance data were obtained using Sentinel-3 Ocean Land Color Imagery (OLCI), for a period of May 2016-June 2019. Time and space varying covariates include surface water temperature, ambient temperature, precipitation, and lake geomorphology. The hierarchical Bayesian spatio-temporal modeling approach in R-INLA represents a potential forecasting tool useful for water managers and associated public health applications for predicting near future high-risk cyanoHAB occurrence given the spatio-temporal characteristics of these events in the recent past. This method is robust to missing data and unbalanced sampling between waterbodies, both common issues in water quality datasets.

Project description:In January 2003, the US Environmental Protection Agency sponsored a "roundtable discussion" to develop a consensus on the relationship between eutrophication and harmful algal blooms (HABs), specifically targeting those relationships for which management actions may be appropriate. Academic, federal, and state agency representatives were in attendance. The following seven statements were unanimously adopted by attendees based on review and analysis of current as well as pertinent previous data: 1) Degraded water quality from increased nutrient pollution promotes the development and persistence of many HABs and is one of the reasons for their expansion in the U.S. and the world; 2) The composition - not just the total quantity - of the nutrient pool impacts HABs; 3) High biomass blooms must have exogenous nutrients to be sustained; 4) Both chronic and episodic nutrient delivery promote HAB development; 5) Recently developed tools and techniques are already improving the detection of some HABs, and emerging technologies are rapidly advancing toward operational status for the prediction of HABs and their toxins; 6) Experimental studies are critical to further the understanding of the role of nutrients in HAB expression, and will strengthen prediction and mitigation of HABs; and 7) Management of nutrient inputs to the watershed can lead to significant reduction in HABs. Supporting evidence and pertinent examples for each consensus statement is provided herein.

Project description:<p>Harmful algal blooms (HABs) of the toxic haptophyte <em>Prymnesium parvum</em> are a recurrent problem in many inland and estuarine waters around the world. Strains of <em>P. parvum</em> vary in the toxins they produce and in other physiological traits associated with HABs, but the genetic basis for this variation is unknown. To investigate genome diversity in this morphospecies, we generated genome assemblies for 15 phylogenetically and geographically diverse strains of <em>P. parvum</em> including Hi-C guided, near-chromosome level assemblies for 2 strains. Comparative analysis revealed considerable DNA content variation between strains, ranging from 115 to 845 Mbp. Strains included haploids, diploids and polyploids, but not all differences in DNA content were due to variation in genome copy number. Haploid genome size between strains of different chemotypes differed by as much as 243 Mbp. Syntenic and phylogenetic analyses indicate that UTEX 2797, a common laboratory strain from Texas, is a hybrid that retains 2 phylogenetically distinct haplotypes. Investigation of gene families variably present across strains identified several functional categories associated with metabolic and genome size variation in <em>P. parvum</em> including genes for the biosynthesis of toxic metabolites and proliferation of transposable elements. Together, our results indicate that <em>P. parvum</em> is comprised of multiple cryptic species. These genomes provide a robust phylogenetic and genomic framework for investigations into the eco-physiological consequences of the intra- and inter-specific genetic variation present in <em>P. parvum</em> and demonstrate the need for similar resources for other HAB-forming morphospecies.</p>

Project description:Marine phytoplankton comprise the foundation of oceanic food webs and generate most of the Earth's oxygen. Of the many phytoplankton species in the ocean, a few dozen produce potent toxins, and at high concentrations can form what are called Harmful Algal Blooms (HABs) or "red tides" that can discolor marine waters. Managers and scientists have been monitoring coastal waters and shellfish resources for HABs and their toxins to ensure seafood safety and understand why blooms occur. This educational activity focuses on a prominent HAB species that causes paralytic shellfish poisoning (PSP). Students will learn about the importance of HABs and PSP, as well as how scientists collect and use data to understand and predict blooms. Students will plot data on HAB species collected by scientists over multiple years of sampling. Students will also plot results over time and across regions, report on observed patterns, and complete grade-appropriate calculations. Lastly, group discussion will focus on determining whether geographic patterns exist that might influence where shellfish beds are closed. This activity is timely given the widespread wildlife mortalities and beach closures due to Florida red tide, as well as recent dog deaths attributed to exposure to freshwater cyanobacteria (blue-green algae) blooms.

Project description:Harmful Algal Blooms (HABs) are of global concern, as their presence is often associated with socio-economic and environmental issues including impacts on public health, aquaculture and fisheries. Therefore, monitoring the occurrence and succession of HABs is fundamental for managing coastal regions around the world. Yet, due to the lack of adequate in situ measurements, the detection of HABs in coastal marine ecosystems remains challenging. Sensors on-board satellite platforms have sampled the Earth synoptically for decades, offering an alternative, cost-effective approach to routinely detect and monitor phytoplankton. The Red Sea, a large marine ecosystem characterised by extensive coral reefs, high levels of biodiversity and endemism, and a growing aquaculture industry, is one such region where knowledge of HABs is limited. Here, using high-resolution satellite remote sensing observations (1km, MODIS-Aqua) and a second-order derivative approach, in conjunction with available in situ datasets, we investigate for the first time the capability of a remote sensing model to detect and monitor HABs in the Red Sea. The model is able to successfully detect and generate maps of HABs associated with different phytoplankton functional types, matching concurrent in situ data remarkably well. We also acknowledge the limitations of using a remote-sensing based approach and show that regardless of a HAB's spatial coverage, the model is only capable of detecting the presence of a HAB when the Chl-a concentrations exceed a minimum value of ~ 1 mg m-3. Despite the difficulties in detecting HABs at lower concentrations, and identifying species toxicity levels (only possible through in situ measurements), the proposed method has the potential to map the reported spatial distribution of several HAB species over the last two decades. Such information is essential for the regional economy (i.e., aquaculture, fisheries & tourism), and will support the management and sustainability of the Red Sea's coastal economic zone.

Project description:The relationship between algae and their zooplanktonic predators typically involves consumption of nutrients by algae, grazing of the algae by zooplankton which in turn enhances predator biomass, controls algal growth and regenerates nutrients. Eutrophication raises nutrient levels, but does not simply increase normal predator-prey activity; rather, harmful algal bloom (HAB) events develop often with serious ecological and aesthetic implications. Generally, HAB species are outwardly poor competitors for nutrients, while their development of grazing deterrents during nutrient stress ostensibly occurs too late, after the nutrients have largely been consumed already by fast-growing non-HAB species. A new mechanism is presented to explain HAB dynamics under these circumstances. Using a multi-nutrient predator-prey model, it is demonstrated that these blooms can develop through the self-propagating failure of normal predator-prey activity, resulting in the transfer of nutrients into HAB growth at the expense of competing algal species. Rate limitation of this transfer provides a continual level of nutrient stress that results in HAB species exhibiting grazing deterrents protecting them from top-down control. This process is self-stabilizing as long as nutrient demand exceeds supply, maintaining the unpalatable status of HABs; such events are most likely under eutrophic conditions with skewed nutrient ratios.

Project description:Harmful algal blooms are caused by specific members of microbial communities. Understanding the dynamics of these events requires comparing the strategies developed by the problematic species to cope with environmental fluctuations to the ones developed by the other members of the community. During three consecutive years, the meta-transcriptome of micro-eukaryote communities was sequenced during blooms of the toxic dinoflagellate Alexandrium minutum. The dataset was analyzed to investigate species specific gene expression dynamics. Major shifts in gene expression were explained by the succession of different species within the community. Although expression patterns were strongly correlated with fluctuation of the abiotic environment, and more specifically with nutrient concentration, transcripts specifically involved in nutrient uptake and metabolism did not display extensive changes in gene expression. Compared to the other members of the community, A. minutum displayed a very specific expression pattern, with lower expression of photosynthesis transcripts and central metabolism genes (TCA cycle, glucose metabolism, glycolysis…) and contrasting expression pattern of ion transporters across environmental conditions. These results suggest the importance of mixotrophy, cell motility and cell-to-cell interactions during A. minutum blooms.
